Introduction

Essential peptidyl-tRNA hydrolase component of the mitochondrial large ribosomal subunit (PubMed:20186120, PubMed:33878294). Acts as a codon-independent translation release factor that has lost all stop codon specificity and directs the termination of translation in mitochondrion, possibly in case of abortive elongation (PubMed:33878294). Involved in the hydrolysis of peptidyl-tRNAs that have been prematurely terminated and thus in the recycling of stalled mitochondrial ribosomes (PubMed:20186120, PubMed:33878294).

Comparative proteomic analysis reveals the response mechanism of freshwater leech (Whitmania pigra) under heat-stress challenge

Temperature is an environmental parameter that remarkably affects the survival and organism health of poikilothermal animal-Whitmania pigra Whitman. Heat stress destroys the physiological homeostasis of intestine tissue. However, no studies on the intestinal mucosa response of leech exposure to heat stress have been reported so far. To identify the biomarker proteins involved in heat stress response, we performed a Tandem mass tag (TMT)-based comparative proteomic analysis on leech's intestine after exposing to 27 °C and 35 °C. The cumulative mortality in the 35 °C heat stress group increased on the third day after stress compared with the control group. Mean-while, intestinal malondialdehyde (MDA) content was significantly up-regulated whereas total antioxidant capacity (T-AOC) was significantly down-regulated. A total of 3935 proteins were identified through proteomic analysis. Heat stress resulted in 144 differential proteins; 75 were up-regulated, and 69 were down-regulated. The differential expression proteins in response to heat stress are mainly involved in calcium-binding proteins, molecular chaperones, cytoskeleton integrity, immune function and inflammation response, DNA damage and repair, ribosomal protein synthesis, stress hormones and neuro-transmitters, tumorigenesis, and apoptosis. Protein-protein network interaction analysis showed that HSP, CD8A, C3, CD63, CD81, MRPL58, MRPL15, HNRNPA1, AMBP, IGHEP1, SERPINC1, CYP3A4, ATP12A, RPS13, CaM, HSP70 binding protein, HSPA6, C4A, ANXA7, ARG1, ARG2, ATP1B, CALCA, HTR1B, and GNGT1 are the critical proteins of leeches resistance to heat stress. Our results systematically provide substantial responsive candidate proteins combating heat stress and enhance our understanding of the intrinsic response mechanisms of thermal sensitivities animal W. pigra facing heat exposure.

In Vivo Contribution of Cyp24a1 Promoter Vitamin D Response Elements

AbstractCYP24A1 is a multifunctional, P450 mitochondrial enzyme that catabolizes the vitamin D hormone (calcitriol, 1,25(OH)2D3), its precursor (calcifediol, 25(OH)D3), and numerous vitamin D metabolites. In the kidney, Cyp24a1 is induced by 1,25(OH)2D3 and fibroblast growth factor 23 (FGF23) and potently suppressed by PTH to control the circulating levels of 1,25(OH)2D3. Cyp24a1 is controlled by a pair of promoter proximal (PRO) vitamin D response elements (VDREs) that are aided by distal, downstream (DS) enhancers. The downstream 1 region of Cyp24a1 (DS1) enhancer is kidney-specific and responsible for PTH and FGF23 actions, and the downstream 2 region of Cyp24a1 enhancer responds to 1,25(OH)2D3 in all tissues. Despite this knowledge, in vivo contributions of the PRO VDREs to basal expression, FGF23 activation, and PTH suppression of Cyp24a1 remain unknown. In this study, we selectively mutated the PRO VDREs in the mouse to address these questions. We found mutation of the VDREs leads to a dramatic loss of VDR occupancy, a reduction of 1,25(OH)D3-induced kidney Cyp24a1 expression, and near elimination of intestinal Cyp24a1 induction. FGF23 induction of Cyp24a1 was reduced but not eliminated and still showed a synergistic increase with 1,25(OH)2D3. PTH suppression of Cyp24a1 was unchanged, despite minor reductions in total for phosphorylated cAMP-response element binding protein occupancy. Finally, VDR recruitment was dramatically reduced across the DS enhancers in the Cyp24a1 locus. Taken together, our data suggest a cooperative relationship between the DS and PRO enhancers in the regulation of Cyp24a1 by 1,25(OH)2D3 and FGF23 and points to the DS1 region as a crucial basal switch for Cyp24a1 activity that further defines the interconnected genomic control in vitamin D catabolism.

Variability in soybean yield responses to elevated atmospheric CO2: Insights from non-structural carbohydrate remobilisation during seed filling

The increasing atmospheric CO₂ concentration (e[CO₂]) has mixed effects on soybean most varieties' yield. This study elucidated the effect of e[CO₂] on soybean yield and the underlying mechanisms related to photosynthetic capacity, non-structural carbohydrate (NSC) accumulation, and remobilisation. Four soybean cultivars were cultivated in open-top chambers at two CO₂ levels. Photosynthesis rates were determined from R2 to R6. Plants were sampled at R5 and R8 to determine carbohydrate concentrations. There were significant variations in yield responses among the soybean cultivars under e[CO₂], from no change in DS1 to a 22% increase in SN14. DS1 and SN14 had the smallest and largest increase, respectively, in daily carbon assimilation capacity. Under e[CO₂], DS1, MF5, and XHJ had an increase in Ci, at which point the transition from Rubisco-limited to ribulose-1,5-bisphosphate regeneration-limited photosynthesis occurred, in contrast with SN14. Thus, the cultivars might have distinct mechanisms that enhance photosynthesis under e[CO₂] conditions. A positive correlation was between daily carbon assimilation response to e[CO₂] and soybean yield, emphasising the importance of enhanced photosynthate accumulation before the R5 stage in determining yield response to e[CO₂]. E[CO₂] significantly influenced NSC accumulation in vegetative organs at R5, with variation among cultivars. There was enhanced NSC remobilisation during seed filling, indicating cultivar-specific responses to the remobilisation of sucrose and soluble sugars, excluding sucrose and starch. A positive correlation was between leaf and stem NSC remobilisation and yield response to e[CO₂], emphasising the role of genetic differences in carbohydrate remobilisation mechanisms in determining soybean yield variation under elevated CO₂ levels.
