Introduction

Constitutes one of the E3 ubiquitin-protein ligases that mediate monoubiquitination of 'Lys-119' of histone H2A, thereby playing a central role in histone code and gene regulation. H2A 'Lys-119' ubiquitination gives a specific tag for epigenetic transcriptional repression and participates in X chromosome inactivation of female mammals. Essential component of a Polycomb group (PcG) multiprotein PRC1-like complex, a complex class required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones, rendering chromatin heritably changed in its expressibility. Compared to RNF2/RING2, it does not have the main E3 ubiquitin ligase activity on histone H2A, and it may rather act as a modulator of RNF2/RING2 activity.

A Cervus Elaphus and Eucommia Ulmoides Extract Blend Attenuates Muscle Atrophy by Regulating Protein Metabolism and Antioxidant Activity

Here, we investigated whether a mixture of Cervus elaphus and Eucommia ulmoides (1:3, KGC01CE) could suppress muscle atrophy in H₂O₂-induced C2C12 cells and dexamethasone-injected mice. Our results revealed that KGC01CE effectively safeguarded against H₂O₂-induced muscle atrophy in C2C12 cells compared with the same mixture at other ratios. We demonstrated that dexamethasone elicited oxidative stress in muscle tissue and decreased the grip strength and cross-sectional areas of muscle fibers; however, oral administration of KGC01CE (1:3) suppressed these dexamethasone-induced changes. Furthermore, oral KGC01CE (1:3) administration suppressed the expression of protein degradation-associated factors, myostatin, muscle RING finger 1, and atrogin-1 in muscle tissue from dexamethasone-injected mice. Oral KGC01CE administration stimulated protein synthesis by stimulating the PI3K/Akt/mTOR pathway in muscle tissue from dexamethasone-injected mice. These findings indicate that KGC01CE can suppress dexamethasone injection-induced muscle atrophy by inhibiting protein degradation and promoting muscle hypertrophy. Therefore, KGC01CE supplementation helps prevent muscle atrophy in mice and may be beneficial against various components of muscle atrophy.

Relationship between macular microvasculature and visual function in idiopathic macular epiretinal membrane by using OCT angiography and multifocal electroretinogram

PURPOSETo investigate the relationship between microvasculature and visual function in idiopathic epiretinal membrane (iERM).METHODSThis retrospective study included 30 iERM eyes. Macular microvasculature was evaluated by optical coherence tomography (OCT) and optical coherence tomography angiography (OCTA). Visual function was evaluated by best corrected visual acuity (BCVA) and multifocal electroretinogram (mfERG). All parameters were measured in the iERM patients at baseline, 1 month, and 3 months postoperatively. Associations between macular microvasculature and visual function were evaluated.RESULTSBCVA, central retinal thickness (CRT) and parafoveal vessel density (PFVD) of the deep capillary plexus (DCP) changed significantly 3 months after surgery. P1 amp density and amplitude of ring 1 decreased significantly 3 months postoperatively. P1 implicit time of ring 1 was shortened after surgery. Baseline PFVD of DCP was negatively correlated with the postoperative BCVA, and baseline BCVA was positively correlated with the postoperative BCVA. Baseline foveal avascular zone (FAZ) and P1 amplitude of ring 1 were positively correlated with the postoperative P1 amplitude of ring 1 (p < 0.05). Multiple linear regression analysis revealed that baseline PFVD of DCP was a predictor of the postoperative BCVA, and baseline FAZ was a predictor of the postoperative P1 amplitude of ring 1 (p < 0.05).CONCLUSIONAlthough visual acuity and CRT improved after iERM surgery, visual function estimated by mfERG had a limited recovery 3 months postoperatively. Baseline PFVD of the DCP and FAZ were predictors of visual function after the iERM surgery.

Fermented antler extract attenuates muscle atrophy by regulating the PI3K/Akt pathway and inflammatory response in immobilization-treated C57BL/6J mice

Muscle atrophy or muscle wasting, which is featured by reduced muscle function and mass, typically results from disuse, aging, and chronic diseases. The deer antler, which refers to the young and non-ossified antlers of various species of deer-related animals, is not fully calcified and comprises of densely growing hair. Here, we investigated whether Bacillus subtilis-fermented antler extract (FAE) inhibits immobilization-induced muscle atrophy in C57BL/6J mice. Oral administration of FAE increased grip strength, exercise performance, muscle mass, and volume in mice. FAE stimulated the phosphatidylinositol 3-kinase (PI3K)/Akt pathway, enhancing the mammalian target of rapamycin pathway for muscle synthesis. FAE phosphorylated Forkhead box O3 and downregulated muscle RING finger-1 and atrogin-1 for proteolysis. FAE inhibited the mRNA expression of tumor necrosis factor alpha and interleukin-6 through nuclear factor kappa B. Consequently, FAE attenuated muscle atrophy by regulating the PI3K/Akt pathway and inflammation.
