The zinc absorption of the novel peptide‐Zn complex in Caco‐2 cells: effects of soybean peptides charge and hydrophobicity

AbstractBACKGROUNDThe supplemental effect of zinc depends not only on adequate intake, but also on how efficiently it is absorbed in the small intestine. In the present study, weak hydrophobic peptides (WHP), strong hydrophobic peptides (SHP), positively charged peptides (PCP) and negatively charged peptides (NCP) were isolated from soybean peptides (SP). The peptide‐Zn complexes (PCP‐Zn, NCP‐Zn, WHP‐Zn, SHP‐Zn and SP‐Zn) were prepared to compare their promotion zinc absorption capacity in the Caco‐2 cells monolayers model.RESULTSWe found that the carboxyl, carbonyl and amino groups in peptide were the primary binding sites of Zn. Compared with zinc sulfate, the peptide‐Zn complexes with different charge and hydrophobic peptides could improve zinc solubility at different pH. NCP‐Zn had a lower Zn‐binding capacity but a higher zinc absorption capacity compared to that of PCP‐Zn in Caco‐2 cells. In addition, the capacity of PCP‐Zn to promote zinc absorption was lower than the control group (SP‐Zn). There were no significant differences in transport rates, retention rates and uptake rates of WHP‐Zn, SHP‐Zn and SP‐Zn. NCP‐Zn could improve the activity of Zn‐related enzymes, and the expression levels of PepT1 and ZnT1 were higher than other peptide‐Zn complexes.CONCLUSIONThe promotion zinc absorption capacity of peptide‐Zn complexes was not completely dependent on the Zn‐binding capacity, but also depended on the charge and hydrophobicity of peptides. © 2024 Society of Chemical Industry.

To identify novel regulators of endoplasmic reticulum (ER)-linked protein degradation and ER function, we determined the entire inventory of membrane-spanning RING finger E3 ubiquitin ligases localized to the ER. We identified 24 ER membrane-anchored ubiquitin ligases and found Nixin/ZNRF4 to be central for the regulation of calnexin turnover. Ectopic expression of wild type Nixin induced a dramatic down-regulation of the ER-localized chaperone calnexin that was prevented by inactivation of the Nixin RING domain. Importantly, Nixin physically interacts with calnexin in a glycosylation-independent manner, induces calnexin ubiquitination, and p97-dependent degradation, indicating an ER-associated degradation-like mechanism of calnexin turnover.

BACKGROUNDOne of the major obstacles in using artificial insemination to manage genetics of elephant population in captivity is the large variations in semen quality among ejaculates within the same and among individuals. The objectives of this study were to determine the influences of (1) age (2) seasonality (3) and circulating testosterone (SrTest), triiodothyronine (SrT3) and tetraiodothyronine (SrT4), as well as seminal (4) testosterone (SpTest), zinc (SpZn) and protein (SpTP) on semen quality in the Asian elephantMETHODSAnalyses, including motility, viability and morphology were performed in semen samples collected twice monthly from 13 elephant bulls (age range, 10-to 72-years) by manual stimulation between July 2004 and June 2005. Serum samples obtained monthly were assessed for SrTest, SrT3, SrT4, and seminal plasma samples were evaluated for, SpTest, SpZn and SpTP.RESULTSThe highest semen quality was observed at age 23 to 43 years. Percentages of progressive motility and viable sperm were lowest at age 51 to 70 years (P < 0.05); on the other hand, sperm concentration was lowest at age 10 to 19 years (P < 0.05). Percentage of sperm with normal morphology was highest at age 23 to 43 years. The levels of SrT3, SrTest, SpTest and SpZn were lowest at age 51 to 70 years, whereas SrT4 was lowest at age 23 to 43 years. Seasonality significantly affected semen characteristics in which percentage of viable sperm and cell concentration were highest during rainy season and lowest during summer months (P < 0.05). However, percentage of sperm with normal morphology was highest in summer and lowest in rainy season (P < 0.05). Seasonality significantly influenced SrTest with elevated concentrations observed in rainy season and winter (P < 0.05).CONCLUSIONThis study indicates that age and seasonality had influence on semen characteristics in the Asian elephant. The knowledge obtained in this study will improve our understanding of the reproductive biology of this species.
