Epigenetic regulation of physiological resilience to ammonia nitrogen stress in the Pacific whiteleg shrimp Penaeus vannamei: Evidence from genome-wide DNA methylation dynamics

Although DNA methylation has emerged as an essential epigenetic mechanism modulating organismal responses to abiotic stresses, its involvement in the physiological resilience of marine invertebrates like shrimp to ammonia nitrogen toxicity remains enigmatic. Here, we performed the first comprehensive dissection of genome-wide DNA methylation dynamics in the Pacific whiteleg shrimp Penaeus vannamei exposed to ammonia nitrogen, based on whole-genome bisulfite sequencing and transcriptome analyses. In the genome of P. vannamei, three DNA methyltransferases (DNMT1, DNMT2 and DNMT3a), one DNA demethylase (TET2) and four methyl-CpG binding proteins (MBD2, MBD4, Kaiso, and UHRF1) were present. About 1.68-1.87 % of cytosine nucleotides were methylated, and higher percentages of cytosines in the CpG context (5.23 %-6.34 %) was methylated compared with the CHG and CHH contexts. Methylated cytosines were mostly enriched in the coding DNA sequence, and methylation peaks occurred near the transcription end sites. Following ammonia exposure, 4203 differentially expressed genes (DEGs) and 1100 differentially methylated genes (DMGs) were identified. The DMGs accounted for 4.4 % of the total gene reservoir in P. vannamei genome, and 212 shared genes were found between the DEGs and DMGs. Genes exhibiting significant methylation and expression changes were enriched in various pathways including the FoxO signaling pathway, autophagy and endocytosis. Among them was a group of genes related to energy metabolism, antioxidation response and detoxification metabolism, highlighting involvement of DNA methylation in fine-tuning these crucial physiological processes. These findings provide new insights into the regulatory roles of DNA methylation in the physiological resilience of marine invertebrates to aquatic stressors.

Multigenerational immunotoxicity assessment: A three-generation study in Drosophila melanogaster upon developmental exposure to triclosan

Triclosan (TCS) is widely used as an antibacterial agent, nevertheless, its presence in different environmental matrices and its persistent environmental nature pose a significant threat to the organism, including humans. Numerous studies showed that TCS exposure could lead to multiple toxicities, including immune dysfunction. However, whether parental TCS exposure could impair the offspring's immune response remains limited. Maintaining the immune homeostasis is imperative to neutralize the pathogen and crucial for tissue repair and the organism's survival. Thus, this study aimed to assess the multigenerational immune response of TCS using Drosophila melanogaster. TCS was administered to organisms (1.0, 10, and 100.0 μg/mL) over three generations during their developing phases, and its effect on the immunological response of the unexposed progeny was evaluated. Total circulatory hemocyte (immune cells) count, crystal cell count, phagocytic activity, clotting time, gene expression related to immune response and epigenetics, ROS generation, and cell death were assessed in the offspring. A concentration-dependent decline in total hemocytes, crystal cells, phagocytic activity, and increased clotting time in the subsequent generations was observed. Furthermore, parental TCS exposure enhanced the ROS levels, induced cell death, and altered the expression of antimicrobial peptides drosomycin, diptericin, and inflammatory genes upd1, upd2, and upd3, in the offspring's hemocytes across successive generations. The upregulation of reaper hid, and grim suggests that TCS promotes apoptotic death in the offspring's hemocytes. Notably, the increased mRNA expression of epigenetic regulators dnmt2 and g9a in the hemocytes of the offspring indicates epigenetic modifications. Further, we also observed that the antioxidant N-acetylcysteine (NAC) supplementation to the parents alleviated TCS toxicity and improved immunological functions in the progeny, indicating the role of ROS in the TCS-induced multigenerational immune toxicity. This finding provides valuable insights into the potential immune risk of prenatal TCS exposure to their offspring in the higher organism.

Mitochondrial dysfunction and cell senescence are triggered by reactive oxygen species (ROS) in granulosa cells (GCs), leading to premature ovarian insufficiency (POI). Human umbilical cord mesenchymal stem cell-derived exosome (HuMSCs-Ex, H-Ex)-based treatments have been shown to alleviate ROS-induced POI, but knowledge about the underlying therapeutic mechanisms is limited. Here, we observed that the 5-methylcytosine (m5C) RNA methyltransferase tRNA aspartic acid methyltransferase 1 (TRDMT1) promoted the translation of COX subunit 5B (COX5B) in a manner dependent on its catalytic activity and downstream m5C reader Y-box binding protein 1 (YBX1), which was decreased in prematurely senescent GCs but abundant in H-Ex. Mechanistically, YBX1 released by H-Ex recognizes the TRDMT1-mediated m5C modification of COX5B and directly binds to COX5B^C-153 via LYS-92, thereby reducing ROS accumulation and improving mitochondrial function in GCs under oxidative stress, providing new insights into the theoretical basis for the great clinical potential of H-Ex in the treatment of POI.
