The Detection of CatSper1 and CatSper3 Expression in Men with Normozoospermia and Asthenoteratozoospermia and Its Association with Sperm Parameters, Fertilization Rate, Embryo Quality

CatSper affects sperm function and male fertilization capacity markers, including sperm motility and egg penetration. The study has aimed to evaluate the mRNA expression of CatSper1, and CatSper3 in the spermatozoa of men with normozoospermia and Asthenoteratozoospermia, and to assess the correlation between genes expression and sperm parameters, fertilization rate, and embryo quality in intracytoplasmic sperm injection (ICSI). Reverse transcription-polymerase chain reaction was utilized to evaluate the mRNA expression of CatSper1 and CatSper3 in sperm in two patient groups: Normozoospermia (NOR; n = 32), and Asthenoteratozoospermia (AT; n = 22). In all patients receiving intracytoplasmic sperm injection, the fertilization rate and embryo quality were evaluated. CatSper1, and CatSper3 mRNA expression in sperm was significantly lower in AT males than in NOR (P < 0.05). Levels of these genes demonstrated a significant positive correlation with sperm motility, mitochondrial membrane potential (MMP), capacitation, fertilization rate, cleavage rate, and embryo quality (P < 0.05) following ICSI. However, a negative correlation was found between mRNA expression of CatSper1, 3 and sperm DNA fragmentation (P < 0.05). Findings indicate low levels of CatSper1 and CatSper3 mRNA expression in men with Asthenoteratozoospermia, which resulted in poor sperm quality and impaired embryo development following ICSI therapy.

New gold(III) complexes TGS 121, 404, and 702 show anti-tumor activity in colitis-induced colorectal cancer: an in vitro and in vivo study

AbstractBackgroundChronic inflammation in the course of inflammatory bowel disease may result in colon cancer, or colitis-associated colorectal cancer (CACRC). It is well established that CACRC is associated with oxidative stress and secretion of multiple pro-inflammatory cytokines, e.g. tumor necrosis factor-α. Recently, we proved that the administration of gold(III) complexes resulted in the alleviation of acute colitis in mice. The aim of the current study was to assess the antitumor effect of a novel series of gold(III) complexes: TGS 121, 404, 512, 701, 702, and 703.MaterialsAnalyzed gold(III) complexes were screened in the in vitro studies using colorectal cancer and normal colon epithelium cell lines, SW480, HT-29, and CCD 841 CoN, and in vivo, in the CACRC mouse model.ResultsOf all tested complexes, TGS 121, 404, and 702 exhibited the strongest anti-tumor effect in in vitro viability assay of colon cancer cell lines and in in vivo CACRC model, in which these complexes decreased the total number of colonic tumors and macroscopic score. We also evidenced that the mechanism of action was linked to the enzymatic antioxidant system and inflammatory cytokines.ConclusionsTGS 121, 404, and 702 present anti-tumor potential and are an attractive therapeutic option for colorectal cancer.

Identification of Potential Biomarkers Associated with Spermatogenesis in Azoospermia

BACKGROUNDAzoospermia, characterized by the absence of spermatozoa in the ejaculate, affects approximately 1% of all men and 10 - 15% of infertile males, representing the most severe form of male infertility. It is classified into obstructive azoospermia (OA) and nonobstructive azoospermia (NOA), with the latter often resulting from unexplained failures in spermatogenesis. This study endeavored to clarify the molecular underpinnings of sper-matogenesis in NOA and to identify viable therapeutic targets.METHODSWe analyzed expression data from NOA and normal spermatogenesis samples obtained from the GEO database. Differential expression analysis was performed to identify differentially expressed genes (DEGs). We then intersected these DEGs with genes known to be related to spermatogenesis to pinpoint spermatogenesis-related DEGs specific to NOA. Subsequent analyses, including gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) pathway enrichments, aimed to elucidate potential signaling pathways involved. A protein-protein interaction (PPI) network was constructed to highlight hub genes, whose diagnostic potential was assessed by using ROC curve analysis. Additionally, miRNA and transcription factor (TF) regulatory network for hub genes were analyzed. The efficacy of identified hub genes as biomarkers was validated through RT-qPCR and Western blotting in a mouse model of NOA.RESULTSThis study identified 68 NOV-specific spermatogenesis-related genes. Enrichment analyses in GO and KEGG pathways highlighted their involvement in cellular processes related to reproduction in multicellular organism and endocrine and other factor-regulated calcium reabsorption. Seven hub genes were identified, with ROC curve analysis affirming their significant diagnostic value. Constructed networks revealed intricate interactions among miRNAs, hub genes, and TFs.CONCLUSIONSWe identified seven hub genes (CATSPER1, CATSPER3, CATSPER4, CATSPERG, OAZ3, ODF1, and SUN5) significantly associated with spermatogenesis in NOA, demonstrating their potential as biomarkers for diagnosing and monitoring the disease.
