Introduction

Secretory protein that plays a role in various cellular processes (PubMed:20969804, PubMed:24706764, PubMed:31883645). Acts as a chemorepellent acting on gonadotropin-releasing hormone (GnRH) expressing neurons regulating their migration to the hypothalamus (PubMed:31883645). Also promotes neuron migration, growth and survival as well as neurite outgrowth and is involved in the development of the olfactory system (PubMed:20969804, PubMed:31883645). May also act through the regulation of growth factors activity and downstream signaling (PubMed:31883645). Also regulates extracellular matrix assembly and cell adhesiveness (By similarity). Promotes endothelial cell survival, vessel formation and plays an important role in the process of revascularization through NOS3-dependent mechanisms (PubMed:24706764).

The perception of the sensory world in mammals requires information flow from the thalamus to the cortex. Although the first-order sensory thalamus and its surrounding nuclei are considered the major hub for feedforward thalamocortical transmission, it remains unknown whether any other thalamic input could also contribute to this transmission. We found a thalamic region, the basal region of the ventromedial nucleus of the thalamus (bVM), that sends dense, tonotopically arranged projections to auditory cortex (AuC) fields. Silencing these AuC-projecting neurons severely impaired the mouse's ability to discriminate sound frequencies. These projections exhibited strong frequency-tuning preferences that matched the cortical tonotopic map. Moreover, bVM inputs were excitatory and primarily terminated on neuron-derived neurotrophic factor-positive interneurons in cortical layer 1. Silencing these inputs significantly reduced sound-evoked responses of AuC neurons. Our results reveal a non-canonical, tonotopically arranged thalamic input to cortical layer 1 that contributes to sound processing, in parallel to the classic auditory thalamocortical pathway.

Heterogeneity of Layer 1 Interneurons in the Mouse Medial Prefrontal Cortex

ABSTRACTCortical Layer 1 (L1) acts as a critical relay for processing long‐range inputs. GABAergic inhibitory interneurons (INs) in this layer (Layer 1 interneurons [L1INs]) function as inhibitory gates, regulating these inputs and modulating the activity of deeper cortical layers. However, their characteristics and circuits in the medial prefrontal cortex (mPFC) remain poorly understood. Using biocytin labeling, we identified three distinct morphological types of mPFC L1INs: neurogliaform cells (NGCs), elongated NGCs (eNGCs), and single‐bouquet cell‐like (SBC‐like) cells. Whole‐cell recordings revealed distinct firing patterns across these subtypes: NGCs and eNGCs predominantly exhibited late‐spiking (LS) patterns, and SBC‐like cells displayed a higher prevalence of non‐LS (NLS) patterns. We observed both electrical and chemical connections among mPFC L1INs. Optogenetic activation of NDNF+ L1INs demonstrated broad inhibitory effects on deeper layer neurons. The strength of inhibition on pyramidal neurons (PyNs) and INs displayed layer‐specific preference. These findings highlight the functional diversity of L1INs in modulating mPFC circuits and suggest their potential role in supporting higher order cognitive functions.
