Molecular correlates of glycine receptor activity in human β cells

OBJECTIVESGlycine acts in an autocrine positive feedback loop in human β cells through its ionotropic receptors (GlyRs). In type 2 diabetes (T2D), islet GlyR activity is impaired by unknown mechanisms. We sought to investigate if the GlyR dysfunction in T2D is replicated by hyperglycemia per se, and to further characterize its action in β cells and islets.METHODSGlyR-mediated currents were measured using whole-cell patch-clamp in human β cells from donors with or without T2D, or after high glucose (15 mM) culture. We also correlated glycine-induced current amplitude with transcript expression levels through patch-seq. The expression of the GlyR α1, α3, and β subunit mRNA splice variants was compared between islets from donors with and without T2D, and after high glucose culture. Insulin secretion from human islets was measured in the presence or absence of the GlyR antagonist strychnine.RESULTSAlthough gene expression of GlyRs was decreased in T2D islets, and β cell GlyR-mediated currents were smaller, we found no evidence for a shift in GlyR subunit splicing. Glycine-induced currents are also reduced after 48 h culture of islets from donors without diabetes in high glucose, where we also find the reduction of the α1 subunit expression, but an increase in the α3 subunit. We discovered that glycine-evoked currents are highly heterogeneous amongst β cells, inversely correlate with donor HbA1c, and are significantly correlated to the expression of 92 different transcripts and gene regulatory networks (GRNs) that include CREB3(+), RREB1(+) and ZNF697(+). Finally, glucose-stimulated insulin secretion is decreased in the presence of the GlyR antagonist strychnine.CONCLUSIONSWe demonstrate that glucose can modulate GlyR expression, and that the current decrease in T2D is likely due to the receptor gene expression downregulation, and not a change in transcript splicing. Moreover, we define a previously unknown set of genes and regulons that are correlated to GlyR-mediated currents and could be involved in GlyR downregulation in T2D. Among those we validate the negative correlation between EIF4EBP1 expression and GlyR activity.

UVR regulation of photoprotection in Phaeodactylum tricornutum (Bacillariophyceae): roles of light energy doses

The ability of diatoms to adapt to variable light conditions such as photosynthetically active radiation (PAR) and ultraviolet radiation (UVR) is crucial for their survival and ecological role. The study aimed to investigate how different doses of UVR regulate the photoprotection mechanism in Phaeodactylum tricornutum. Five treatments were established: Control (PAR only), PAR + UVR D_6h (17.5 W m^- 2, 6 h, 378 KJ m^- 2), PAR + UVR 2D_6h (35 W m^- 2, 6 h, 756 KJ m^- 2), PAR + UVR 2D_12h (17.5 W m^- 2, 12 h, 756 KJ m^- 2), and PAR + UVR 4D_12h (35 W m^- 2, 12 h, 1.512 KJ m^- 2). The growth of P. tricornutum was significantly affected by UVR doses, with a growth pattern of 42-55% below the control (PAR only). Increasing the UVR dose also had negative effects on photosynthesis parameters and the levels of chlorophyll-a and fucoxanthin. The de-epoxidation state showed a high rate in treatments subjected to higher UVR doses, promoting an attempt to activate cellular protection mechanisms. In the transcriptional genes related to the xanthophyll cycle, a reduction in transcript levels of ZEP1 and ZEP2 genes was observed in PAR + UVR treatments, including a reduction up to 75% at 72 h of exposure. Also, an increase in transcript levels of VDE and VDL1 genes was observed for treatments with the same UVR dose, reaching about 9-fold increase for the 2D_12h dose and 10-fold increase for the 2D_6h dose, both at 72 h, suggesting a modification in the cell's ability to respond to UVR light stress.

Clinical, Morphologic, and Genomic Findings in Spitz Tumors With RET Fusion: A Series of 31 Cases

RET-fused Spitz neoplasms are rare and poorly characterized. We describe clinical, histologic and molecular findings of 31 Spitz neoplasms with RET fusion diagnosed as 16 Spitz nevus (SN), 13 atypical Spitz tumors (AST) and 2 Spitz melanoma (SM). The lesions mainly occurred in children and young adults of both sexes with predilection for the extremities. Microscopically, there were mainly symmetrical compound melanocytic neoplasms with a dome-shaped/slightly raised silhouette predominantly composed of epithelioid, spindled and/or smaller nevoid melanocytes arranged in confluent nests. Dyscohesive melanocytes within the nests in the upper part of the lesions, prominent Kamino bodies, giant multinucleated melanocytes, variable pigmentation and increased vascularity with vascular ectasia were frequent features. RNA sequencing detected 9 different 5' (N-terminus) fusion partners, including KIF5B (n= 8), LMNA (n= 7), CCDC6 (n= 6), OPTN (n= 3), MYO5A (n= 2) and NCOA4, ERC1, MYH9, AGAP3 (n= 1). Of these, OPTN::RET and AGAP3::RET represent novel fusions, and 3 further 5' fusion partners, namely NCOA4, ERC1 and MYH9 have never been reported in Spitz tumors. Although as a whole group, the tumors showed a heterogenous histopathologic presentation, correlation of the morphological features and the 5' fusion partners demonstrated certain associations. Nevoid melanocytes were exclusively encountered in cases with KIF5B fusion partner. Neuroid-like appearances with intersecting fascicles of spindled cells typified both MYO5A-fused cases. Epithelioid melanocyte population dominated cases with LMNA and CCDC6 fusion partners. Transepidermal elimination/floating intraepidermal nests of pigmented both spindled and epithelioid melanocytes were observed in the OPTN subgroup. The remaining cases with less frequent 5' fusion partners manifested in general more atypical histopathologic features, including nuclear pleomorphism, high mitotic count, atypical mitoses and sheet-like growth pattern. Melanoma fluorescence in situ hybridization (FISH) Probe Kit targeting RREB1, MYC, CDKN2A, and CCND1, was negative for copy number variation in 4 cases tested, including 2 cases with complete p16 nuclear loss on immunohistochemistry. Array comparative genomic hybridization (aCGH) performed in 3 lesions detected numerous segmental chromosomal imbalances in 2 cases diagnosed SM. DNA and RNA sequencing detected several further genomic alterations, including POU2F3 overexpression in 3 highly pigmented lesions. Further studies are needed to confirm possible correlations between the microscopic features and a particular fusion partner (or additional genetic events) in RET-fused Spitz neoplasms.
