Introduction

Facilitative hexose transporter that mediates the transport of glucose, fructose and galactose (PubMed:8027028, PubMed:16186102, PubMed:23396969, PubMed:28083649, PubMed:8457197). Likely mediates the bidirectional transfer of glucose across the plasma membrane of hepatocytes and is responsible for uptake of glucose by the beta cells; may comprise part of the glucose-sensing mechanism of the beta cell (PubMed:8027028). May also participate with the Na(+)/glucose cotransporter in the transcellular transport of glucose in the small intestine and kidney (PubMed:3399500). Also able to mediate the transport of dehydroascorbate (PubMed:23396969).

Chemical characterization and DPP IV inhibitory capacity of purified adzuki bean β-vignin digest in comparison to soybean β-conglycinin and in vitro effect of β-vignin on diabetic-related outcomes

Adzuki bean (AB) is a legume with a low glycemic index and is traditionally used in Asian cultures to modulate type 2 diabetes (T2D). Our objectives were to characterize the functional peptides from purified AB β-vignin after simulated gastrointestinal digestion in comparison to soybean β-conglycinin, to evaluate their DPP IV inhibitory capacity, and to determine in vitro the effect of digested AB β-vignin on diabetic-related outcomes using HepG2 cells in healthy and insulin-resistant states. Five peptides (215-742 Da) from AB β-vignin and five peptides (215-447 Da) from β-conglycinin were identified to exhibit bioactivity as DPP IV inhibitors. Molecular docking demonstrated peptides could bind to DPP IV at the same binding site as a diabetic medication, linagliptin. Digested AB β-vignin significantly increased (p < 0.05) hepatic glucose uptake (> 290 %) via DPP IV inhibition (> 40 %) in healthy and insulin-resistant states. IRS-1, Akt-1, and Glut 2 increased after treating cells with digested AB β-vignin in healthy and insulin-resistant states.

The effect of carbohydrates with different levels of digestibility on energy metabolism in vivo under hypobaric hypoxic conditions

Current strategies for improving energy supply in hypobaric hypoxic environments are limited. Therefore, this study investigates the effects of four carbohydrates with different levels of digestibility on energy metabolism in vivo in hypobaric hypoxic environments. First, we characterized the four types of carbohydrates. Subsequently, reverse transcription quantitative polymerase chain reaction (RT-qPCR) was used to characterize the expression of GLUT1, GLUT2, and SGLT1 in the glucose transport pathway in vivo. In addition, the effects of different levels of carbohydrate digestibility on energy expenditure were evaluated in vivo. The results showed that pre-gelatinized corn starch significantly increased GLUT1 gene expression in the hypobaric hypoxic conditions (1.58 times, compared to normobaric normoxic). In addition, pre-gelatinized corn starch increased energy expenditure in the hypobaric hypoxic conditions and performed better in terms of glycogen accumulation and glucose transport. Therefore, pre-gelatinized corn starch administration may be a promising strategy for long-term energy supplementation in hypobaric hypoxic.

MicroRNAs bta-novel-miR-117, bta-novel-miR-234 and bta-novel-miR-417 have adverse effects on blastocyst formation

In a previous study we found that the levels of the novel microRNAs (miRNAs) bta-novel-miR-117 bta-novel-miR-234 and bta-novel-miR-417 (P < 0.001) are significantly up-regulated in extracellular vesicles (EVs) in the culture medium of degenerating embryos compared to blastocysts. Because the functions of these novel miRNAs are still unknown, we investigated their regulatory roles during bovine blastocyst development by adding their mimics and inhibitors to the culture medium. The addition of mimics for bta-novel-miR-117, bta-novel-miR-234 and bta-novel-miR-417 resulted in a decreased blastocyst rate, and supplementation of bta-novel-miR-234 inhibitors increased the cleavage rate significantly (P < 0.001). Low-input transcriptome analysis and RT-qPCR results revealed that bta-novel-miR-117, bta-novel-miR-234 and bta-novel-miR-417 co-target genes such as ANKEF1, HAND2 and SLC2A2, downregulated their expression significantly (P < 0.001). These genes associated with glucose transmembrane transport and plasma membrane raft metabolism play crucial roles in embryonic development. The results suggest that overexpressing of these three novel miRNAs impairs embryonic development, and they might serve as biomarkers to detect failing bovine embryos.
