Acsbg1 regulates differentiation and inflammatory properties of CD4+ T cells

AbstractEpigenetic modifications are critical for the regulation of CD4+ T cell differentiation and function. Previously, we identified Acyl-CoA Synthetase Bubble Gum 1 (Acsbg1), a gene involved in fatty acid metabolism, as part of an epigenetic signature that was selectively demethylated in ex vivo isolated T helper 17 (TH17) cells. However, its functional relevance for CD4+ T cells remains incompletely understood. Here, we used in vitro differentiation assays and the adoptive transfer colitis model to investigate the role of Acsbg1 in the differentiation and function of TH1, TH17, and regulatory T (Treg) cells. In vitro, Acsbg1 was expressed in both TH17 and in vitro-induced Treg (iTreg) cells, whereas TH1 cells lacked Acsbg1 expression. Accordingly, Acsbg1 deficiency resulted in impaired TH17 and iTreg differentiation, whereas TH1 differentiation was unaffected. In vivo, upon adoptive transfer of Acsbg1⁻/⁻ Tnaïve cells, immunodeficient recipient mice exhibited an exacerbated colitis, characterized by an altered balance of TH17 and Treg cells, indicating that Acsbg1 expression is essential for optimal TH17 and Treg cell differentiation and function. Our findings highlight the importance of fatty acid (FA) metabolism in maintaining immune homeostasis by regulating T cell differentiation and provide novel insights into the metabolic targeting of inflammatory diseases.

Consensus gene co-expression analysis across multiple intestinal tissues to identify key genes and pathways associated with abdominal fat deposition in broilers

1. Abdominal fat deposition (AFD) is regulated by multiple intestinal tissues, and changes in the function of intestinal tissues are associated with AFD. Currently, integration of transcriptomic data across multiple intestinal tissues to explore excessive AFD has rarely been reported in broilers.2. In this study, a consensus gene co-expression network across the duodenum, jejunum, ileum and caecum of high- and low-abdominal fat broiler lines (HL and LL) was constructed using a publicly available transcriptomic data set. Combining the results of functional enrichment analyses and differential gene expression analyses, this investigated the genes and biological pathways across the four intestinal tissues that might influence AFD.3. In one expression module, NDUFA5, NDUFS6, NDUFA4, NDUFS4, ATP5H, ATP5J and ATP5C1 were significantly enriched in the oxidative phosphorylation pathway, with GPX2 and GSR significantly enriched in the glutathione metabolism pathway. These genes were significantly downregulated in the four intestinal tissues of the HL compared to LL chickens, which may be associated with AFD by increasing intestinal permeability.4. Lipid metabolism relevant genes were identified in other modules (ALDH7A1, ACSBG1, THEM4 and DECR1), which may be linked to AFD through regulation of lipid metabolism. Interestingly, in the first module, 12 genes were significantly enriched in the proteasome pathway and significantly downregulated in the four intestinal tissues in HL birds compared to LL birds, indicating a link between the proteasome and AFD.

The role of molecular weight on chitosan and chitosan oligosaccharides in sleep regulation: Integrating network pharmacology and multi-omics analysis in Drosophila

This study aims to investigate the sleep-regulating effects of chitosan (COS) with different molecular weights (1 kDa, 3 kDa and 30 kDa). Network pharmacology predicted the sleep-regulating effects of COS and COS oligosaccharides and identified sleep-related targets. Drosophila sleep analysis demonstrated that COS ameliorated caffeine-induced insomnia, and its efficacy was critically influenced by molecular weight. Specifically, COS30K exhibited superior effects compared to COS1K and COS3K in improving the nighttime sleep time, duration of sleep episode and number of sleep episodes in flies. These differential effects were primarily related to acetate-producing bacteria Acetobacter and polysaccharide-degrading bacteria (Bacteroides and Ruminococcus). Their influence on tryptophan metabolism (serotonin and kynurenine) may contribute to the molecular weight-dependent differences in sleep-regulating effects of COS. Integration of network pharmacology and transcriptomic analyses identified five crucial targets (Insc, Aox1, bgm, Acer and hll). This study provides a strategy for predicting novel biological activity of COS and COS oligosaccharides.
