Structural characterization and hypolipidemic activity of a hetero-galactan purified from Sanghuangporus vaninii based on modulation of TLR4/NF-κB pathway

Sanghuangporus vaninii showed great activities of anti-inflammation and anti-tumor, due to its bioactive macromolecules. However, the hypolipidemic properties of polysaccharides isolated from S. vaninii have not been systematically reported. In this research, a polysaccharide of S. vaninii was obtained and its hypolipidemic activity was investigated. SVP3, a neutral hetero-galactan from S. vaninii, has a →6)-α-Galp-(1→ backbone with partial H-2 branches of α-Manp-(1→ or α-Manp-(1→2)-α-Fucp-(1→. In a hyperlipidemia mouse model, SVP3 significantly inhibited body weight gain and suppressed serum levels of total cholesterol, triglycerides, and low-density lipoprotein cholesterol. SVP3 inhibited the expansion of adipocytes in three types of white adipose tissues and attenuated hepatic injury and hepatic lipid deposition in the mice. The combined analysis of gut microbiota, serum metabolomics, and liver proteomics revealed that SVP3 effectively regulated the abundance of specific gut microbiota and serum metabolites and mediated the inhibitory effect on inflammation-associated toll-like receptor 4/nuclear factor kappa-B pathway by regulating the expression levels of glutathione S-transferase P1, stromal cell derived factor 2-like 1, ribosomal protein L10, thiosulfate sulfurtransferase, and biliverdin reductase A in liver, ultimately realizing the hypolipidemic activity. The results of the present study provide experimental evidence for the development of clinical adjuvant therapeutic drugs to treat hyperlipidemia.

Assemble and comparative analysis of the mitochondrial genome of Rhododendron delavayi: Insights into phylogenetic relationships and genomic variations

Rhododendron delavayi, a notable ornamental plant primarily found in regions of China like Yunnan and Guizhou provinces, holds substantial horticultural value. To elucidate the systematic phylogenetic relationships and organelle genomic differences within R. delavayi and related Rhododendron species, we conducted sequencing and assembly of the complete mitochondrial genome of R. delavayi. The full-length mitochondrial genome of it was a singular circular molecule spanning 1,009,263 bp, comprising 53 protein-coding genes, including 18 transfer RNA (tRNA) genes, 3 ribosomal RNA (rRNA) genes, and 32 protein-coding genes. A total of 1,182 simple sequence repeats (SSRs) loci were identified in the R. delavayi mitochondrial genome, primarily consisting of single nucleotide, dinucleotide, and trinucleotide repeats. Nucleotide diversity analysis highlighted five genes (atp6, atp9, cox2, nad1, and rpl10) with the highest diversity within the mitochondrial genomes of Rhododendron genus. Comparative analysis of the mitochondrial genome of R. delavayi with those of four other Rhododendron species indicated complex rearrangements in 21 genes, including rps4, nad6, rps3, atp6, cob, atp9, nad7, among others. The mitochondrial phylogenetic tree revealed a close relationship between R. delavayi and R. decorum, forming a sister clade to R. × pulchrum and R. simsii. Furthermore, 126 plastid-to-mitochondrial gene transfers in R. delavayi were identified, ranging from 30 bp to 19,385 bp. These fragments collectively constituted 47.54 % and 9.52 % of the chloroplast and mitochondrial genomes (202,169 bp), respectively. Complex mitochondrial-to-mitochondrial transfers were also observed, with 843 identified fragments totaling 312,036 bp (30.92 % of the mitochondrial genome). Segments exceeding 10 kb may mediate homologous recombination within the mitochondrial molecules. Remarkably, our study underscores that the mitochondrial genome of R. delavayi was the largest reported within the Rhododendron genus to date. The intricate rearrangements observed in the mitochondrial genomes of Rhododendron species, alone with the identification of five potential molecular marker sites, provided valuable insights for species classification and parentage identification within the Rhododendron genus.

Analysis of the lymph node metastasis distribution in patients with invasive resectable non-small cell lung cancer

BackgroundThe current standard for the surgical management of lung cancer involves anatomic lung resection combined with systemic lymph node dissection/sampling. The purpose of this study was to investigate the patterns of pathological lymph nodes in invasive non-small cell lung cancer (NSCLC), explore the occurrence in lymph node metastasis (LNM), and provide recommendations for optimal lymph node resection/sampling in lung cancer operation.MethodsThere were 1,678 patients with NSCLC who underwent lobectomy between 2018 and 2021 at the Taizhou Hospital of Zhejiang Province were reviewed retrospectively. The location and incidence of LNM and postoperative pathological findings were studied. We analysed the metastasis rates of lymph node dissection stations using Pearson's χ² and Fisher's exact tests.ResultsThere were 1,308 patients assessed as eligible and included in the study. The median number of lymph nodes cleared in the cohort was 11.2±5.1. In patients with lung adenocarcinoma, the rate of LNM was significantly higher in central than in peripheral lung cancer, especially in 2R/2L, L7, L9, L10, L11, and L12. Lung cancer patients with tumors ≤1 cm had no N2 lymph node metastases but few (2/191, 1.1%) N1 lymph node metastases. The likelihood of N2 metastasis increased (T1a, 0%, 0/191; T1b 3.5%, 22/625; T1c, 5.6%, 14/249; T2 and above, 18.9%, 46/243) with increasing tumor diameter. Thirty-four patients with stage N2 lung adenocarcinoma and 1-3 cm tumors displayed lobe-specific lymph node metastases in the mediastinum. In patients diagnosed with squamous cell carcinoma, no significant differences were observed in mediastinal LNM across various parameters (central versus peripheral location, tumor site, and tumor size).ConclusionsOur study proposes recommendations for lymph node resection according to the pathological type of lung cancer, tumor location, lung lobes affected and tumor size, which may provide a certain reference value for the clinical work.
