Introduction

Plays a key role in the sexual differentiation of germ cells by promoting the male fate but suppressing the female fate. Represses the female fate pathways by suppressing meiosis, which in turn results in the promotion of the male fate. Maintains the suppression of meiosis by preventing STRA8 expression, which is required for premeiotic DNA replication, after CYP26B1 is decreased. Regulates the localization of the CCR4-NOT deadenylation complex to P-bodies and plays a role in recruiting the complex to trigger the degradation of mRNAs involved in meiosis. Required for the maintenance of the spermatogonial stem cell population. Not essential for the assembly of P-bodies but is required for the maintenance of their normal state (By similarity).

Unlocking New Pharma/Nutraceutical Frontiers With Neuroprotective Properties of Three Hypericum Species: A Study Combination With In Vitro and In Silico Methodologies

ABSTRACTThis study investigates the phytochemical composition and biopharmacological potential of three Hypericum species (H. scabrum, H. lysimachioides, and H. uniglandulosum) from Turkey. Aqueous and hydroalcoholic extracts were analyzed for their total phenolic content (TPC), total flavonoid content (TFC), and individual components (by the UHPLC–HRMS technique). Antioxidant activities were investigated by DPPH, ABTS, CUPRAC, FRAP, phosphomolybdenum, and metal chelating assays. The inhibition effects of the tested extracts on acetylcholinesterase (AChE), butyrylcholinesterase (BChE), tyrosinase, amylase, and glucosidase were examined. One hundred compounds were identified in the chemical composition, and specific compounds for the genus Hypericum, such as hyperoside, hypericin, and pseudohypericin, were detected. The highest TPC was detected in the ethanol/water extract of H. lysimachioides with 69.21 mg GAE/g. Furthermore, the ethanol/water extract showed the strongest free radical and reducing effect. The ethanol/water extracts of the tested Hypericum species were more active in tyrosinase, amylase, and glucosidase than the water extracts. Neuroprotective assessments indicated downregulation of COX‐2 and NOS‐2 genes in LPS‐stimulated mouse cortex models, alongside modulation of SERT and NET expression, suggesting reduced neuroinflammation and enhanced neurotransmitter release. Molecular docking and dynamics analyses highlighted strong binding interactions, especially in the NET_hyperoside and NET_myricitrin complexes. The results indicate significant therapeutic potential for these extracts, supporting their development as natural agents against oxidative stress, neuroinflammation, and related neurodegenerative diseases.

Examination of immunohistochemical of the effects of flattened and unflattened radiotherapy beams in nude mice breast cancer xenografts

PURPOSEThe aim of this study was to investigate the radiobiological effects underlying the inhibition of breast cancer (BCa) following radiotherapy in nude mice models, and to evaluate the impact of changes in immunohistochemical parameters induced by FF and FFF beams.MATERIALS AND METHODSThe study included thirty-six adult nude mouse models, which were randomly assigned to five groups: control (G1), breast cancer (BCa) (G2), FF-400 MU/min (G3), FFF-1100 MU/min (G4), and FFF-1800 MU/min (G5). The control group received neither radiation nor treatment, while the BCa group had a cancer model without radiation. The BCa models were subjected to a single dose of 20 Gy of radiotherapy at varying dose rates. Twenty days after the implantation of the MCF-7 cancer cell line, the nude mice were irradiated and sacrificed 48 h later for ER, PR, HER-2, Ki-67, CD-133, Caspase-3, APAF-1, NOS-2 and NOS-3 IHC analysis.RESULTSA statistically significant decrease in IHC staining values for ER, Ki-67 and NOS-2 was observed in the FF-400 MU/min, FFF-1100 MU/min and FFF-1800 MU/min groups due to radiotherapy compared to the BCa group. The FFF beams demonstrated superior efficacy in the treatment of BCa. The significant differences in Caspase-3 and APAF-1 levels were found between BCa and control groups, while CD-133, NOS-3, HER-2, and PR staining showed no differences between groups.CONCLUSIONSIt was concluded that FFF beam was more effective than FF beam for BCa, especially on ER, Ki-67 and NOS-2 IHC parameters.

Dietary organic acid blend modulates hemato-immunological parameters, digestive and reproductive performances in red tilapia (Oreochromis niloticus × O. mossambicus) broodstock

AbstractThis study evaluated the effects of dietary organic acid (OA) blend on hemato-immunological responses, reproduction capacity, gene expression, and histological features of red tilapia (Oreochromis niloticus × O. mossambicus) broodstock. Four diets were formulated, containing 0 (control), 2, 3, and 4 g OAs kg−1. The diets were fed to triplicate groups (n = 3) of red tilapia broodstock (75 ± 5.56 g( (mean ± standard deviation (SD)) stocked in 10-m3 concrete tanks at a male to female ratio of 1:3, to satiation, twice a day, for 8 weeks. At the end of the feeding trial, fish in each tank were collected, counted, and weighed. Blood samples were collected from five fish from each tank and used for the determination of hematological and biochemical parameters. The fish were then dissected to study the reproductive performance and reproduction-related genes. The red blood cells (RBCs), hemoglobin (Hb), packed cell volume (PCV%), mean corpuscular volume (MCV), and lysozyme activity were significantly increased (P < 0.05) with increasing dietary OAs to 4 g kg−1. Mean corpuscular hemoglobin (MCH), white blood cells (WBCs), total protein, albumin, and globulin), and digestive enzyme activity values leveled off or slightly decreased (P > 0.05) at OA levels above 3 g kg−1. Optimum liver enzyme activity was obtained at 2 g kg−1 OA. The reproductive hormones: testosterone (T), follicle-stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E2), and progesterone (Prog), organo-somatic index (GSI), reproductive performance, and the expression of reproductive genes (vasa, nanos1a, nanos2, dnd1, pum1, amh, and VTG) exhibited dose-dependent responses (P < 0.05), suggesting that 4 g OA kg−1 boosted the optimum reproductive performance. In conclusion, about 2–3 g OA kg−1 diet can improve the hemato-biochemical parameters, immune response, antioxidant status, and digestive enzyme activity in red tilapia broodstock, whereas 4 g kg−1 could accelerate their reproductive performance through upregulation of reproductive genes.
