Exploration of the Relationship between Polycystic Ovary Syndrome and Recurrent Pregnancy Loss Based on Bioinformatics

Background:Recurrent Pregnancy Loss (RPL) and Polycystic Ovary Syndrome (PCOS) are both common diseases involving women of childbearing age, and their pathogenesis is still not sufficiently known.Objective:This study aimed to explore the relationship between RPL and PCOS in bioinformatics.Methods:Two expression chips, GSE86241 (obtained from 8 PCOS patients and 9 healthy controls) and GSE73025 (obtained from 5 RPL patients and 5 healthy controls), were downloaded from the Gene Expression Omnibus (GEO) database. We used the GEO database to analyze the gene expression profiles of PCOS and RPL to identify the intersection of abnormal miRNA expression, predicted the target genes of the intersecting miRNAs from miRDB, miRTarBase, and TargetScan databases, and then incorporated the miRNA-mRNA modulation network. By using the string database, the PPI network was built, which could screen the Hub genes and enrich them for analysis. Ultimately, the critical miRNA-mRNA regulatory network was set on the basis of the relationship between hub genes and miRNA.Results:A total of 39 significantly altered miRNAs of PCOS and 137 significantly altered miRNAs of RPL were obtained, three miRNAs (miR-767-5p, miR-3196, and miR-187-3p), five signaling pathways (PI3K-Akt, p53, Toll-like receptor, C-type lectin receptor, and TNF signaling pathways), and six Hub genes (CASP8, PIK3R1, ADAMTS2, ADAMTS3, COL3A1, and MDM2) were found to be related to the development and progression of two diseases. More importantly, all Hub genes were regulated by miR-767-5p.Conclusion:This research clarifies the possible relationship between miRNA and mRNA with PCOS and RPL for the first time. It provides a basis for illustrating the pathogenic mechanism and a target of therapies for these two diseases.

Cumulative Impact Loading and Cartilage Synthesis Biomarkers May Be Associated With Injury Risk in Female Collegiate Basketball Players

PurposeTo evaluate whether cumulative impact load and serum biomarkers are related to lower-extremity injury and to determine any impact load and cartilage biomarker relationships in collegiate female basketball athletes.MethodsThis was a prospective longitudinal study evaluating lower-extremity impact load, serum cartilage biomarkers, and injury incidence over the course of a single collegiate women's basketball season. Data were collected from August 2022 to April 2023; no other follow-up after the cessation of the season was conducted in this cohort. Inclusion criteria for the study included collegiate women's basketball athletes, ages 18 to 25 years, who were noninjured at the start of the study time frame (August 2024). Cartilage synthesis (procollagen II carboxy propeptide and aggrecan chondroitin sulfate 846 epitope) and degradation (collagen type II cleavage) biomarkers were evaluated at 6 season timepoints. Impact load metrics (cumulative bone stimulus, impact intensity) were collected during practices using inertial measurement units secured to the distal medial tibiae. Injury was defined as restriction of participation for 1 or more days beyond day of initial injury. Cumulative impact load metrics were calculated over the week before any documented injury and blood draws for analysis. Point biserial and Pearson product moment correlations were used to determine the relationship between impact load metrics, serum biomarkers, and injury.ResultsEleven collegiate women's basketball athletes (height: 1.86 meters, mass: 82.0 kg, age: 20.54 years) participated. Greater medium-range (6-20 g) cumulative impact intensities during week 5 and 6 for both limbs (r = 0.674, P = .023) and high-range (20-200 g) during week 8 for both limbs (0.672, P = .024) were associated with injury. Greater cumulative bone stimulus was associated with increased procollagen II carboxy propeptide levels before conference playoffs for right (r = 0.694, P = .026) and left (r = 0.747, P = .013) limbs. Greater chondroitin sulfate 846 epitope levels at off-season-1 (r = 0.729, P = .017), and at the beginning of the competitive season (r = 0.645, P = .044) were associated with season-long injury incidence.ConclusionsIn this study, we found that moderate-to-high intensity impacts (6-200 g) early in the season were associated with subsequent injury among female collegiate basketball players. Increased cartilage synthesis at various time points was correlated with increased cumulative bone stimulus metrics and season-long injury incidence in this population.Level of EvidenceLevel IV, prognostic case series.

Genetic background of hematological parameters in Holstein cattle based on genome-wide association and RNA sequencing analyses

Hematological parameters refer to the assessment of changes in the number and distribution of blood cells, including leukocytes (LES), erythrocytes (ERS), and platelets (PLS), which are essential for the early diagnosis of hematological system disorders and other systemic diseases in livestock. In this context, the primary objectives of this study were to investigate the genomic background of 19 hematological parameters in Holstein cattle, focusing on LES, ERS, and PLS blood components. Genetic and phenotypic (co)variances of hematological parameters were calculated based on the average information restricted maximum likelihood method and 1,610 genotyped individuals and 5,499 hematological parameter records from 4,543 cows. Furthermore, we assessed the genetic relationship between these hematological parameters and other economically important traits in dairy cattle breeding programs. We also carried out genome-wide association studies and candidate gene analyses. Blood samples from 21 primiparous cows were used to identify candidate genes further through RNA sequencing (RNA-seq) analyses. Hematological parameters generally exhibited low-to-moderate heritabilities ranging from 0.01 to 0.29, with genetic correlations between them ranging from -0.88 ± 0.09 (between mononuclear cell ratio and lymphocyte cell ratio) to 0.99 ± 0.01 (between white blood cell count and granulocyte cell count). Furthermore, low-to-moderate approximate genetic correlations between hematological parameters with one longevity, 4 fertility, and 5 health traits were observed. One hundred ninety-nine significant SNP located primarily on the Bos taurus autosomes (BTA) BTA4, BTA6, and BTA8 were associated with 16 hematological parameters. Based on the RNA-seq analyses, 6,687 genes were significantly downregulated and 4,119 genes were upregulated when comparing 2 groups of cows with high and low phenotypic values. By integrating genome-wide association studies (GWAS), RNA-seq, and previously published results, the main candidate genes associated with hematological parameters in Holstein cattle were ACRBP, ADAMTS3, CANT1, CCM2L, CNN3, CPLANE1, GPAT3, GRIP2, PLAGL2, RTL6, SOX4, WDFY3, and ZNF614. Hematological parameters are heritable and moderately to highly genetically correlated among themselves. The large number of candidate genes identified based on GWAS and RNA-seq indicate the polygenic nature and complex genetic determinism of hematological parameters in Holstein cattle.
