NUCLEAR TRANSPORT FACTOR 2‐LIKE improves drought tolerance by modulating leaf water loss in alfalfa (Medicago sativa L.)

SUMMARYDrought is a major environmental factor that limits the production of alfalfa (Medicago sativa). In the present study, M. sativa NUCLEAR TRANSPORT FACTOR 2‐LIKE (MsNTF2L) was identified as a nucleus‐, cytoplasm‐, and plasma membrane‐localized protein. Its transcriptional expression was highly induced by ABA and drought stress. Overexpression of MsNTF2L in Arabidopsis resulted in hypersensitivity to ABA during both the seed germination and seedling growth stages. However, transgenic Arabidopsis plants exhibited enhanced tolerance to drought stress by reducing the levels of reactive oxygen species (ROS) and increasing the expression of stress/ABA‐inducible genes. Consistently, analysis of MsNTF2L overexpression (OE) and RNA interference (RNAi) alfalfa plants revealed that MsNTF2L confers drought tolerance through promoting ROS scavenging, a decrease in stomatal density, ABA‐induced stomatal closure, and epicuticular wax crystal accumulation. MsNTF2L highly affected epicuticular wax deposition, as a large group of wax biosynthesis and transport genes were influenced in the alfalfa OE and RNAi lines. Furthermore, transcript profiling of drought‐treated alfalfa WT, OE, and RNAi plants showed a differential drought response for genes related to stress/ABA signaling, antioxidant defense, and photosynthesis. Taken together, these results reveal that MsNTF2L confers drought tolerance in alfalfa via modulation of leaf water loss (by regulating both stomata and wax deposition), antioxidant defense, and photosynthesis.

Genetic characterization of nuclear export factor NXT1 and its paralog NXT2 in primates and murine rodents

Translocation of RNA across the nuclear envelope relies on transport receptors. Receptor nuclear transport factor 2 (NTF2)-like export protein 1 (NXT1 [also called p15 or p15-1]) shuttles between the nucleus and cytoplasm of metazoan cells and contributes to the nuclear export of a diverse spectrum of RNAs. NXT2 (also called p15-2), a paralog of NXT1 in eutherians, also has implications for RNA nuclear export. A comprehensive description is currently lacking as to the genetic signature of these molecules. In this study, we analyzed genetic changes in the NXT1 and NXT2 genes in primates and murine rodents, including the commonly used model organisms Macaca spp., Mus musculus, and Rattus norvegicus. The results show that NXT1 has been subject to functional constraints in both phylogenetic lineages. Conversely, NXT2 exhibits discrepant patterns of genetic changes between these taxa. Murine NXT2 has evolved conservatively; by contrast, adaptive selection has frequently contributed to genetic changes in primate NXT2. The genetic discrepancy of the NXT2 orthologs leads to the suggestion that they had experienced quite different evolutionary fates potentially constituting different functional implementations in these taxa. These findings raise awareness of further study on different organisms to comprehensively understand their functional characteristics.

AbstractCancer dependency maps, which use CRISPR/Cas9 depletion screens to profile the landscape of genetic dependencies in hundreds of cancer cell lines, have identified context-specific dependencies that could be therapeutically exploited. An ideal therapy is both lethal and precise, but these depletion screens cannot readily distinguish between gene effects that are cytostatic or cytotoxic. Here, we use a diverse panel of functional genomic screening assays to identify NXT1 as a selective and rapidly lethal in vivo relevant genetic dependency in MYCN-amplified neuroblastoma. NXT1 heterodimerizes with NXF1, and together they form the principal mRNA nuclear export machinery. We describe a previously unrecognized mechanism of synthetic lethality between NXT1 and its paralog NXT2: their common essential binding partner NXF1 is lost only in the absence of both. We propose a potential therapeutic strategy for tumor-selective elimination of a protein that, if targeted directly, is expected to cause widespread toxicity.Significance:We provide a framework for identifying new therapeutic targets from functional genomic screens. We nominate NXT1 as a selective lethal target in neuroblastoma and propose a therapeutic approach where the essential protein NXF1 can be selectively eliminated in tumor cells by exploiting the NXT1–NXT2 paralog relationship.See related commentary by Wang and Abdel-Wahab, p. 2129.This article is highlighted in the In This Issue feature, p. 2113
