Computational Modeling of the Enzymatic Achmatowicz Rearrangement by Heme-Dependent Chloroperoxidase: Reaction Mechanism, Enantiopreference, Regioselectivity, and Substrate Specificity

The chloroperoxidase from Caldariomyces fumago (CfCPO) catalyzes the oxidative ring expansion of α-heterofunctionalized furans via the Achmatowicz rearrangement, providing an elegant tool to convert furan rings into complex-prefunctionalized scaffolds. However, the mechanism of this transformation remains unclear. Herein, the CfCPO-catalyzed reaction of rac-1-(2-furyl)ethanol (1a) is studied by quantum chemical calculations and molecular dynamics simulations. The calculations reveal that the conversion follows the general mechanism of the Achmatowicz reaction. Notably, the binding of 1a to the enzyme's active site influences the Compound I (Cpd I) formation, and the (R)-1a enantiomer binding results in a lower barrier compared to (S)-1a, explaining the observed (R)-enantiopreference toward a racemic substrate. Additionally, due to the weaker steric hindrance between the porphyrin ring and substrate, the nucleophilic attack of Cpd I on the furan core of 1a is preferred at the less-substituted C4=C5 bond, providing a rationale for the experimentally observed regioselectivity. Finally, the bottleneck residues in the substrate delivery channel and also the active site surroundings are proposed to be responsible for the substrate specificity of CfCPO. This study lays a theoretical foundation for the rational design of new CPOs that catalyze the Achmatowicz rearrangement with a broader substrate spectrum or specific stereopreference.

Temporal dynamics of extended-spectrum β-lactamase-producing Escherichia coli and carbapenemase-producing Gram-negative bacteria in hospital wastewater

Hospital wastewater is a reservoir for the environmental spread of clinically relevant antimicrobial-resistant bacteria and resistance genes. The aim of this study was to quantify total Escherichia coli, extended-spectrum β-lactamase (ESBL)-producing E. coli, and carbapenemase-producing organisms (CPOs) and perform whole-genome sequencing-based characterization of these bacterial isolates in hospital wastewater samples collected bimonthly in Japan from January to November 2021. Total E. coli counts were 8.1 × 10³-8.8 × 10⁴ colony-forming units (CFU)/mL. ESBL-producing E. coli were detected in the sampling months of January, March, May, and July, with the ratio of ESBL-producing E. coli to total E. coli being remarkably highest (95 %) in July. In contrast, DHA-1 Ambler class C β-lactamase (AmpC)-producing E. coli was detected in September and November, accounting for 28 % and 3 % of total E. coli counts, respectively. All 140 ESBL-producing E. coli isolates harbored the bla_CTX-M genes, with bla_CTX-M-14 being the most common genotype (94.3 %), the vast majority of which were associated with the human virulent B2-O25b: H4-ST131-fimH30R/non-Rx. In September, E. coli clade I-O8:H33-ST3910-fimH1074 was primarily associated with bla_DHA-1. Among 26 representative CPO isolates, Aeromonas caviae (34.6 %) and A. hydrophila subsp. hydrophila (30.8 %) were dominant. The most frequently detected carbapenemase gene was bla_IMP-1 (57.7 %), followed by bla_GES-24 (34.6 %) and bla_GES-4 (7.7 %). Estimated bacterial counts of CPOs ranged from 4.0 × 10^-1 to 4.7 × 10³ CFU/mL over the six sampling months. bla_IMP-1-positive A. hydrophila subsp. hydrophila ST860, which was repeatedly detected over the five sampling months, accounted for the highest total number of this bacterial clone (79 %). Overall, this study provides insights into the overwhelming presence and persistence of E. coli B2-O25b:H4-ST131-H30R/non-Rx with bla_CTX-M-14 and Aeromonas spp. with bla_IMP-1 in hospital wastewater, and the change in the dynamics of resistance gene prevalence from bla_CTX-M-positive E. coli to bla_DHA-1-positive E. coli.

Neutrophil‐Mimicking Nanozyme with Cascade Catalytic Releasing Nitric Oxide and Signet Oxygen Property for Synergistic Bimodal Therapy of Methicillin‐Resistant Staphylococcus Aureus Infections

AbstractRecently, chloroperoxidase (CPO)‐mediated enzyme dynamic therapy (EDT) by mimicking the antipathogen function of neutrophils via generating highly active signet oxygen (1O2) has attracted great interest in biomedical applications. However, the therapeutic efficiency of EDT is largely restricted by the low CPO delivery efficiency and insufficient hydrogen peroxide (H2O2) supply. In the present work, a neutrophil‐mimicking nanozyme of MGBC with high CPO delivery efficiency, H2O2 self‐supply, and enzyme‐cascade catalytic properties is designed for high‐efficient treatment of methicillin‐resistant Staphylococcus aureus (MRSA) infections. In the infection microenvironment, MGBC can effectively catalyze glucose to self‐supply substantial H2O2, which enables long‐lasting 1O2 generation via the CPO‐mediated catalytic reaction. At the meantime, MGBC can also catalyze H2O2 to sustainably release NO for gas therapy (GT), which synergistically strengthens the therapeutic effect of EDT. As a result, MGBC displayed effective MRSA‐killing and MSRA biofilms‐eradicating properties, and high efficiency in treating both MRSA infected full‐thickness excision wounds and subcutaneous MRSA infection by exerting the synergistic bimodal EDT/GT therapeutic effects. In‐depth mechanism study revealed that the synergistic EDT/GT antibacterial effects of MGBC can attenuate the drug resistance and toxicity of MRSA by significantly downregulating quorum sensing, multidrug efflux, virulence, and biofilm formation‐related genes.
