Introduction

Ribosome-binding protein involved in ribosome hibernation, a process during which ribosomes are stabilized in an inactive state and preserved from proteasomal degradation (By similarity). Acts via its association with eiF5a (EIF5A and EIF5A2) at the polypeptide exit tunnel of the ribosome, preventing mRNA translation (By similarity). Involved in ribosome hibernation in the mature oocyte by preventing mRNA translation, leading to ribosome inactivation (By similarity). Ribosomes, which are produced in large quantities during oogenesis, are stored and translationally repressed in the oocyte and early embryo (By similarity). Also acts as a negative regulator of autophagy (PubMed:20537536). Involved in mediating interferon-gamma-induced cell death (PubMed:7828849).

Cysteine-assisted synthesis of copper nanoclusters for construction of FRET-based ratiometric sensor for visual detection of alkaline phosphatase

Abnormal alkaline phosphatase (ALP) levels in the human body are closely associated with various diseases, particularly hepatobiliary diseases and bone diseases. Herein, we constructed a ratiometric sensor based on Förster resonance energy transfer (FRET) using strongly photoluminescent copper nanoclusters (Cu NCs) for the detection of ALP with high sensitivity and specificity. The cysteine-stabilized Cu NCs (Cys-Cu NCs) were synthesized through a ligand-exchange reaction and core-size etching focusing, which displayed bright photoluminescence (PL) with a quantum yield (QY) of 10.5 %. Multispectral characterization indicated that zwitterionic cysteine ligands without obvious steric hindrance could significantly enhance intra/inter-ligand-involved charge transfer, leading to a significant increase in fluorescence emission (∼14 folds) compared to precursor Cu NCs. A FRET-based ratiometric sensor for ALP detection was constructed by combining Cys-Cu NCs with a Cu²⁺-assisted oxidation reaction of o-phenylenediamine (OPD) to generate fluorescent 2,3-diaminophenazine (DAP). The strong coordination interaction between the ALP substrate and Cu²⁺ significantly affected the FRET process between Cys-Cu NCs and DAP, thereby altering the fluorescence ratio. Based on the specific response of ALP to its substrate, the ratiometric sensors showed good linear relationship within the range of 0.1-50 U/L, with a detection limit (LOD) of 0.075 U/L. Furthermore, the FRET-based ratiometric sensor was integrated with a polymer hydrogel to fabricate a portable hydrogel sensor for simple and visual detection of ALP.

A novel dual-mode colorimetric and ratiometric fluorescence method for detecting isocarbophos based on S, N-CDs inhibiting Fe3O4@Cu-BTC nanozyme activity

Although many colorimetric methods have been developed for on-site pesticide detection, most exhibit low sensitivity and limited resistance to interference. To address these issues, this study developed a novel dual-mode colorimetric-ratiometric fluorescence method for isocarbophos (ICP) detection using sulfur and nitrogen co-doped carbon nanodots (S, N-CDs) to inhibit catalytic Fe₃O₄@Cu-BTC nanozyme activity. The synthesized Fe₃O₄@Cu-BTC nanozymes with higher peroxidase-like activity were inhibited by S, N-CDs. The inhibitory mechanism was deeply investigated and was ascribed to (1) the restriction of the reaction between Fe₃O₄@Cu-BTC and hydrogen peroxide (H₂O₂), which decreased the ·OH production, and (2) the ·OH scavenging ability of the S, N-CDs, forming the basis of the dual-mode method. The Fe₃O₄@Cu-BTC/Apt-complementary strand (CS)/S, N-CDs hybridization complexes demonstrated minimal to no peroxidase-like activity, preventing H₂O₂ from oxidizing o-phenylenediamine (OPD) to produce yellow fluorescent 2,3-diaminophenazine (DAP). Meanwhile, the detection system demonstrated a low fluorescence intensity ratio (I₅₇₅/I₄₆₅) at an excitation wavelength of 390 nm. However, the presence of ICP caused the CS/S, N-CDs to separate from the Fe₃O₄@Cu-BTC/Apt due to the high affinity between Apt and ICP. Then, the Fe₃O₄@Cu-BTC/Apt-ICP complexes recovered peroxidase-like activity, which promoted DAP production, accompanied by a pronounced fluorescence intensity ratio (I₅₇₅/I₄₆₅). In optimum conditions, the detection limits (LODs) were 2.79 ng/mL (colorimetric) and 0.26 ng/mL (ratiometric fluorescence), respectively. The feasibility of the proposed method was verified via three spiked samples, yielding recovery rates of between 86.08 % and 112.64 %, indicating the significant potential for the highly sensitive, accurate on-site detection of ICP in actual samples.

Proteomic profiling of kars knockout zebrafish larvae

BACKGROUNDKARS encodes both mitochondrial and cytoplasmic lysyl-tRNA synthetase, which is one of the aminoacyl-tRNA synthetases (ARSs) necessary for protein translation. Pathogenic variants in KARS have been reported to be involved in hearing loss, visual disorders, neuropathology, and diseases combined with multisystem phenotypes. In vitro studies have shown that KARS mutations cause a decrease in aminoacylation. However, the pathogenetic mechanisms underlying the complex neurological phenotypes remain largely unknown.METHODSWe developed kars knockout zebrafish and proteomic analyses on larvae with different genotypes at five days post-fertilization were performed using isobaric tags for relative and absolute quantitation (iTRAQ). Then the differentially abundant proteins (DAPs) analyzed by iTRAQ were validated by parallel reaction monitoring (PRM).RESULTS420 differentially abundant proteins were identified between the knockout and wildtype groups, of which, 138 were up-regulated and 282 down-regulated. The Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways analyses showed the greatest DAP cluster enrichment in ribosome (P = 2.1 × 10^-6, 28 genes), aminoacyl-tRNA biosynthesis (P = 7.34 × 10^-6, 13 genes), and hypertrophic cardiomyopathy (P = 7.45 × 10^-6, 28 genes). A further PRM-based analysis identified changes in nars, mybphb, atp2a1l, col6a1 and rps3a that were specially linked to kars-deficency.CONCLUSIONSThis work provides new valuable in vivo data for understanding the molecular mechanism of KARS deficiency-associated diseases, and will give us comprehensive insights into ARS-related disorders.
