Introduction

Required for normal renal development and establishment of left-right axis. Probably acts as a molecular switch between different Wnt signaling pathways. Inhibits the canonical Wnt pathway by targeting cytoplasmic disheveled (DVL1) for degradation by the ubiquitin-proteasome. This suggests that it is required in renal development to oppose the repression of terminal differentiation of tubular epithelial cells by Wnt signaling. Involved in the organization of apical junctions in kidney cells together with NPHP1, NPHP4 and RPGRIP1L/NPHP8 (By similarity). Does not seem to be strictly required for ciliogenesis (By similarity).

Haplotype‐resolved genome assembly provides new insights into the genomic origin of purple colour in Prunus mume

SummaryPrunus mume, an important ornamental woody plant in the Rosaceae family, contains many interspecific hybridizations. Purple colour is a breeding trait of aesthetic value for P. mume, but little is known about the origin and genetic architecture of this trait. Here we address these issues by producing a haplotype‐resolved genome from an interspecific hybrid cultivar of P. mume (M) and P. cerasifera ‘Pissardii’ (C), named P. mume ‘Meiren’, followed by a detailed molecular characterization. The final length of the diploid genome is 499.47 Mb, with 250.66 Mb of haplotype M (HM) and 248.79 Mb of haplotype C (HC). Approximately 95.42% (476.61 Mb) of the phased assembly is further anchored to 16 homologous chromosomes. Based on the genomic variation, we identify a 1.8 Mb large‐fragmented inversion (INV) on chromosome 1b of HC, which co‐segregates with purple colour traits of ‘Meiren’ inherited from its purple C parent ‘Pissardii’. We find that a MYB transcription factor, PmmMYB10.5b, resides at the distal breakpoint of the INV, which displays consistent allele‐specific expression (ASE). By directly binding to the promoter of anthocyanin synthetic alleles, PmmMYB10.5b serves as a co‐activator to promote anthocyanin accumulation in ‘Meiren’ organs. Notably, the INV identified in ‘Meiren’ is generated from ‘Pissardii’ rather than P. cerasifera, which alters the promoter sequence of PmmMYB10.5b, activates its expression and results in the purple colour trait. Results from this study shed light on the evolutionary origin of purple colour in ‘Meiren’ and could potentially provide guidance on the genetic improvement of colour traits in ornamental woody plants.

ATG9A vesicles are a subtype of intracellular nanovesicle

ABSTRACTCells are filled with thousands of vesicles, which mediate protein transport and ensure homeostasis of the endomembrane system. Distinguishing these vesicles functionally and molecularly represents a major challenge. Intracellular nanovesicles (INVs) are a large class of transport vesicles that likely comprise multiple subtypes. Here, we define the INV proteome and find that it is molecularly heterogeneous and enriched for transmembrane cargo molecules, including integrins, transporters and ATG9A, a lipid scramblase associated with autophagy. ATG9A is known to reside in ‘ATG9A vesicles’ – small vesicles that contribute to autophagosome formation. Here, using in-cell vesicle capture assays, we found that ATG9A, as well as other ATG9A vesicle cargoes, are in INVs. Quantitative analysis showed that virtually all ATG9A vesicles are INVs, but that only ∼20% of INVs are ATG9A vesicles, suggesting that ATG9A vesicles are in fact a subtype of INV, which we term ATG9A-flavor INVs. Finally, we show that perturbing ATG9A-flavor INVs impairs the autophagy response induced by starvation.

Recycled wheat straw biochar enhances nutrient-poor soil: Enzymatic kinetics of carbon, nitrogen, and phosphorus cycling

The recycling of waste wheat straw into biochar for soil improvement is a promising, sustainable strategy to enhancing nutrient-poor soils. Biochar application to soil has been shown to enhances the activity of key enzymes involved in nutrient cycling, such as invertase (INV), urease (URE), and alkaline phosphatase (ALP), which play roles in carbon (C), nitrogen (N), and phosphorus (P) cycling, as well as catalase (CAT), an enzyme with redox properties. However, the kinetic behavior of these enzymes remains largely unexplored. A 36-week laboratory study was conducted to evaluate the effects of biochar on enzyme kinetics in grey desert and aeolian soils including Michaelis-Menten constant (K_m), maximum reaction velocity (V_max), and catalytic efficiency (V_max/K_m). Results indicated that biochar application significantly increased soil pH, available phosphorus (AP), ammonium nitrogen (NH₄⁺-N), dissolved organic carbon (DOC), and microbial biomass carbon (MBC) while reducing nitrate nitrogen (NO₃^--N), which were identified as dominant factors influencing enzyme kinetic parameters. Enzyme activities related to C, N, and P cycling (excluding catalase) increased significantly at a biochar application rate of 4 % by mass. Changes in K_m and V_max for URE, INV, ALP, and CAT suggest that biochar influences enzyme kinetics through mechanisms such as adsorption, microenvironmental shifts, and allosteric modulation. An economic assessment identified 4 % biochar application as the optimal rate, yielding soil quality index (SQI) values of 0.53 and 0.65 for grey desert and aeolian soils, respectively. These findings suggest that biochar-amended soils exhibit improved fertility, highlighting the potential of biochar to enhance soil health.
