The regulatory effect of chitooligosaccharides on islet inflammation in T2D individuals after islet cell transplantation: the mechanism behind Candida albicans abundance and macrophage polarization

Islet cell transplantation (ICT) represents a promising therapeutic approach for addressing diabetes mellitus. However, the islet inflammation during transplantation significantly reduces the surgical outcome rate, which is related to the polarization of macrophages. Chitooligosaccharides (COS) was previously reported which could modulate the immune system, alleviate inflammation, regulate gut microecology, and repair the intestinal barrier. Therefore, we hypothesized COS could relieve pancreatic inflammation by regulating macrophage polarization and gut microbiota. First, 18S rDNA gene sequencing was performed on fecal samples from the ICT population, showing abnormally increased amount of Candida albicans, possibly causing pancreatic inflammation. Functional oligosaccharides responsible for regulating macrophage polarization and inhibiting the growth of Candida albicans were screened. Afterwards, human flora-associated T2D (HMA-T2D) mouse models of gut microbiota were established, and the ability of the selected oligosaccharides were validated in vivo to alleviate inflammation and regulate gut microbiota. The results indicated that ICT significantly decreased the alpha diversity of gut fungal, altered fungal community structures, and increased Candida albicans abundance. Moreover, Candida albicans promoted M1 macrophage polarization, leading to islet inflammation. COS inhibited Candida albicans growth, suppressed the MyD88-NF-κB pathway, activated STAT6, inhibited M1, and promoted M2 macrophage polarization. Furthermore, COS-treated HMA-T2D mice displayed lower M1 macrophage differentiation and higher M2 macrophage numbers. Additionally, COS also enhanced ZO-1 and Occludin mRNA expression, reduced Candida albicans abundance, and balanced gut microecology. This study illustrated that COS modulated macrophage polarization via the MyD88/NF-κB and STAT6 pathways, repaired the intestinal barrier, and reduced Candida albicans abundance to alleviate islet inflammation.

Isatidis root polysaccharides ameliorates post-weaning diarrhea by promoting intestinal health and modulating the gut microbiota in piglets

This study aimed to investigate the effects of dietary isatidis root polysaccharide (IRP) on diarrhea, immunity, and intestinal health in weanling piglets. Forty healthy piglets were randomly assigned to five groups receiving varying dosages of IRP. The findings indicated that different concentrations of IRP significantly reduced diarrhea scores (p < 0.01). Notably, the serum levels of immunoglobulin A and immunoglobulin G increased linearly and quadratically (p < 0.01), while immunoglobulin M also showed a linear increase (p < 0.05) in IRP-fed piglets. The secretory immunoglobulin A levels in ileal contents were significantly higher compared to control piglets (p < 0.01). Key intestinal health parameters, including villus height, villus height-to-crypt depth ratio, and goblet cell numbers, showed linear and quadratic increases in both the jejunum and ileum (p < 0.05), while crypt depth decreased significantly (p < 0.01). Additionally, the expression of IL-10, ZO-1, occludin, and mucin2 was upregulated linearly and quadratically in IRP-fed piglets (p < 0.05). In cultured IPEC-J2 cells, ZO-1 and occludin expression levels significantly increased upon exposure to 400 μg/mL IRP (p < 0.01). Furthermore, the relative abundances of Escherichia coli, Ralstonia pickettii, and Desulfovibrio fairfieldensis decreased linearly with increasing dietary IRP concentration. In conclusion, IRP shows promise as an effective dietary supplement for mitigating diarrhea and enhancing intestinal health in early weaned piglets.

3'-Sialyllactose and B. infantis synergistically alleviate gut inflammation and barrier dysfunction by enriching cross-feeding bacteria for short-chain fatty acid biosynthesis

Ulcerative colitis (UC) poses significant threats to human health and quality of life worldwide, as it is a chronic inflammatory bowel disease. 3'-sialyllactose (3'-SL) is a key functional component of milk oligosaccharides. This study systematically evaluates the prebiotic effects of 3'-SL and its therapeutic potential in combination with Bifidobacterium infantis (B. infantis) for UC. The findings reveal that 3'-SL and B. infantis synergistically mitigate intestinal inflammation and barrier dysfunction by promoting the production of short-chain fatty acids (SCFAs) through cross-feeding mechanisms among gut microbiota. Individually, 3'-SL, B. infantis, and the synbiotic treatment all effectively alleviated UC symptoms, including reduced weight loss, improved disease activity scores, and prevention of colon shortening. Histopathological and immunofluorescence analyses further demonstrated that the synbiotic treatment significantly ameliorated colonic injury, enhanced barrier function, restored goblet cell counts, increased glycoprotein content in crypt goblet cells, and upregulated the expression of tight junction proteins (ZO-1, occludin, and claudin-1). Notably, the synbiotic treatment outperformed the individual components by better restoring gut microbiota balance, elevating SCFA levels, and modulating serum cytokine profiles, thereby reducing inflammation. These findings provide mechanistic insights into the protective effects of the synbiotic and underscore its therapeutic potential for UC and other intestinal inflammatory disorders.
