Introduction

Soluble frizzled-related proteins (sFRPS) function as modulators of Wnt signaling through direct interaction with Wnts. They have a role in regulating cell growth and differentiation in specific cell types. SFRP5 may be involved in determining the polarity of photoreceptor, and perhaps, other cells in the retina.

Association of Serum Secreted Frizzled-Related Protein 5 Levels With Coronary Artery Disease

Background Coronary artery disease (CAD) is the leading cause of death and premature disability worldwide. Secreted frizzled-related protein 5 (SFRP5), a recently identified adipokine, acts as an antagonist of Wingless-type family member 5A (WNT5A) signaling. Since WNT5A triggers inflammation in endothelial cells, the anti-inflammatory properties of SFRP5 play a crucial role in counteracting this effect. Lower SFRP5 levels contribute to the chronic inflammatory conditions associated with CAD by promoting WNT5A signaling. Aim This study aimed to assess serum SFRP5 levels in CAD patients and compare them with those in non-CAD patients at a tertiary care hospital. Materials and methods The study sample included 40 angiographically confirmed CAD patients as cases and 40 non-CAD patients as controls. Serum SFRP5 levels and lipid profiles were measured and compared between the two groups. Data analysis was performed using IBM SPSS Statistics for Windows, Version 26.0 (Released 2019; IBM Corp., Armonk, NY, USA), employing descriptive statistics, normality tests, t-tests, chi-square tests, and correlation analyses. A p-value of <0.05 was considered statistically significant. Results Serum SFRP5 levels were significantly lower in CAD patients (1.25 ± 0.35) compared to controls (2.46 ± 0.96) (p < 0.01). Additionally, SFRP5 showed a significant negative correlation with triglycerides and low-density lipoprotein cholesterol (p < 0.05). Conclusions This study demonstrated that serum SFRP5 levels were significantly lower in CAD patients compared to controls. Therefore, serum SFRP5 may serve as a novel biomarker for the early prediction of CAD.

Expression, prognosis, immunological infiltration, and DNA methylation of members of the SFRP gene family in colorectal cancer: a comparative bioinformatic and experimental analysis

AbstractThis study aimed to investigate the expression, prognostic significance, methylation, and immune invasion levels of secreted frizzled-related proteins (SFRP1-5) in colorectal cancer (CRC). Additionally, the relationship between SFRP1/2 methylation and immune infiltration in CRC was explored. The expression of SFRP1-5 was analyzed using several databases, including GEO, TCGA, TIMER, STRING, and GEPIA. Molecular interactions with SFRPs were examined via Cytoscape software. Gene Ontology (GO) and Kyoto Encyclopedia of Genes, and Genomes (KEGG) pathway analyses were conducted using the DAVID database. Methylation levels of SFRP1/2 in CRC were assessed through methylation-specific PCR (MSP) and bisulfite sequencing PCR (BSP) experiments. Apoptosis and proliferation in CRC cells following the knockdown of SFRP1/2 expression were evaluated using flow cytometry and CCK-8 assays. The TISIDB database was used to analyze the relationship between SFRP1/2 methylation levels and immune infiltration. The expression of SFRP1, SFRP2, and SFRP5 was significantly lower in CRC patients, while SFRP4 expression was higher compared to that in healthy individuals. Elevated mRNA expression of SFRP2 was significantly associated with improved overall survival (OS), disease-specific survival, and progression-free intervals. SFRP1/2 expression was also linked to immune invasion, with higher levels correlating with increased immune infiltration. Both SFRP1 and SFRP2 showed hypermethylation in CRC. Knockdown of SFRP1/2 expression resulted in increased proliferation of CRC cells, and their methylation levels were inversely correlated with immune cell presence. The expression, methylation, and immune cell infiltration patterns of the SFRP family in CRC differed markedly from those in healthy individuals. These findings suggest that SFRPs may serve as potential therapeutic targets and key genes associated with immune cell infiltration in CRC.

Identification of differentially expressed genes and polymorphisms related to intramuscular oleic‐to‐stearic fatty acid ratio in pigs

AbstractThe intramuscular oleic‐to‐stearic fatty acid ratio (C18:1n‐9/C18:0) is an important indicator of the biosynthesis and desaturation of fatty acids in muscle. By using an RNA‐Seq approach in muscle samples from 32 BC1_DU (25% Iberian and 75% Duroc) pigs with divergent values (high: H and low: L) of C18:1n‐9/C18:0 fatty acids ratio, a total of 81 differentially expressed genes (DEGs) were identified. Functional analyses of DEGs indicate that mainly peroxisome proliferator‐activated receptor signaling pathway (associated genes: PPARG, SCD, PLIN1, and FABP3) was overrepresented. Notably, SCD is directly involved in the conversion of C18:0 to C18:1n‐9, and PPARG is a transcription factor regulating lipid metabolism genes, including SCD. However, other DEGs (e.g., ACADVL, FADS3, EPHB2, HGFAC, NGFR, NR0B2, MDH1, MMAA, PPP1R1B, SFRP5, RAB30, and TRARG1) are plausible candidate genes to explain the phenotypic differences of the C18:1n‐9/C18:0 ratio. Interestingly, seven genetic variants within the SCD (including the well‐known AY487830:g.2228T>C SNP and other novel genotyped polymorphisms) are associated with two haplotypes. Although the haplotypes are segregating at different frequencies in the H and L groups, they do not fully explain the desaturation ratios or the SCD expression levels. A more complex model, including polyunsaturated fatty acids such as C18:2n‐6, C20:4n‐6, and C18:3n‐3, is suggested to explain the regulation of the C18:1n‐9/C18:0 desaturation ratio in porcine muscle.
