Introduction

Transcription factor involved in developmental processes such as cell fate determination, gene transcriptional regulation and progenitor cell regulation in a number of organs. Plays a critical role in embryonic development and functions as a key regulatory protein in neurogenesis and the development of the heart, eye lens, liver, pancreas and the lymphatic system. Involved in the regulation of the circadian rhythm. Represses: transcription of the retinoid-related orphan receptor RORG, transcriptional activator activity of RORA and RORG and the expression of RORA/G-target genes including core clock components: BMAL1, NPAS2 and CRY1 and metabolic genes: AVPR1A and ELOVL3.

The role of exercise in promoting lymphangiogenesis and extracellular matrix synthesis in lymphedema-induced tissue injury

BACKGROUNDSecondary lymphedema is a progressive condition caused by lipid- and protein-rich interstitial fluid accumulation resulting from compromised lymphatic function. It commonly occurs in cancer patients following surgical lymph node ablation and radiation treatment. This study aims to elucidate the effects of exercise on the myokine interleukin (IL)-6 and the molecular changes involved in lymphangiogenesis and extracellular matrix (ECM) synthesis using a lymphedema mouse model.METHODS AND RESULTSWe induced lymphedema in male ICR mice by excising the inguinal, popliteal, and iliac lymph nodes and occluding the femoral lymphatic vessel in the right hind leg. We then conducted a time course analysis of swelling and targeted gene expression analysis of the affected leg tissue. We began the mice on involuntary wheel running exercise 2 days post-surgery. Over the subsequent 2 weeks, the swelling initially increased and then gradually declined. In contrast, the genes associated with lymphangiogenesis and ECM synthesis showed progressive upregulation, even after the swelling had subsided. Exercise reduced limb swelling and increased the genetic expression of IL-6 and key lymphangiogenesis markers, including Prox1, VEGF-C, VEGFR-3, and LYVE-1, in the affected limb. Exercise also increased the expression of FABP4 and αSMA, which are markers of active adipocytes and fibroblasts.CONCLUSIONSOur results suggest that exercise reduces lymphedema by promoting lymphangiogenesis and ECM synthesis, facilitating recovery. The insights obtained into the molecular changes underpinning exercise-induced improvements in lymphedema may contribute to the development of targeted therapeutic strategies.

Construction of a prognostic risk model for clear cell renal cell carcinomas based on centrosome amplification-related genes

Clear cell renal cell carcinoma (ccRCC) is the urological malignancy with the highest incidence, centrosome amplification-associated genes (CARGs) have been suggested to be associated with carcinogenesis, but their roles in ccRCC are still incompletely understood. This study utilizes bioinformatics to explore the role of CARGs in the pathogenesis of ccRCC and to establish a prognostic model for ccRCC related to CARGs. Based on publicly available ccRCC datasets, 2312 differentially expressed genes (DEGs) were identified (control vs. ccRCC). Disease samples were classified into high and low scoring groups based on CARG scores and analysed for differences to obtain 345 DEGs associated with CARG scores (S-DEGs). 137 candidate genes were obtained by taking the intersection of DEGs and S-DEGs. Six prognostic genes (PCP4, SLN, PI3, PROX1, VAT1L, and KLK2) were then screened by univariate Cox, LASSO, and multifactorial Cox regression. These genes exhibit a high degree of enrichment in ribosome-associated pathways. Both risk score and age were independent prognostic factors, and the Nomogram constructed based on them had a good predictive performance (AUC > 0.7). In addition, immunological analyses identified 6 different immune cells and 23 immune checkpoints between the high- and low-risk groups, whereas mutational analyses identified frequent VHL mutations in both high- and low-risk groups. Finally, 93 potentially sensitive drugs were identified. In conclusion, this study identified six CARGs as prognostic genes for ccRCC and established a risk model with predictive value. These findings provide insights for prognostic prediction of ccRCC, optimisation of clinical management and development of targeted therapeutic strategies.

Lymphangiogenesis in mouse embryonic and early postnatal ovaries

BACKGROUNDThe critical importance and remodeling capacity of the blood vasculature within the ovary have been extensively analyzed, while the lymphatic vasculature has received limited attention, and its characteristics were reported by only a few studies.OBJECTIVESTo investigate the morphological patterns of lymphatic vessel formation in mouse embryonic and early postnatal ovary.MATERIAL AND METHODSImmunostaining was performed on ovarian tissues from Prox1-EGFP transgenic mice and wild-type mice to investigate the lymphangiogenesis, and the CD31 was employed to label vascular endothelium and LYVE1 and Prox1 as co-markers for lymphatic endothelium.RESULTProx1⁺ cells, LYVE1⁺ cells, and Prox1-EGFP⁺/LYVE1⁺ vessels were absent in the medulla near the ovarian hilum until P7, and YAP1 was expressed in the nucleus and cytoplasm of granulosa cells, alongside the observation of scattered primary follicles within the ovary. By P10, these markers were abundantly expressed in the ovarian medulla, while YAP1 was localized primarily in the cytoplasm of granulosa cells, alongside the occurrence of secondary follicles. By P14, Prox1⁺ cells, LYVE1⁺ cells, and Prox1-EGFP⁺/LYVE1⁺ vessels extended to the cortex, and YAP1 shifted to the nuclei of granulosa cells, alongside the reaching of follicles to the antral stage. From P14 to P28, these markers exhibited a further increasing trend, while YAP1 localized in the nucleus of granulosa cells, alongside the progress of follicles to the preovulatory stage.CONCLUSIONOvarian lymphangiogenesis in the mouse began on postnatal day 7 and subsequently developed from the medulla to the cortex. Additionally, ovarian lymphangiogenesis occurred in synchrony with follicular maturation, during which YAP1 exhibited an ectopic expression pattern.
