Molecular characteristics of oligomeric protein complexes AIM2 and TM4SF19 and their association with the pathogenesis of oral squamous cell carcinoma: Potential biomarkers

The aim of this study was to reveal the molecular characteristics of AIM 2 and TM4SF19 oligomeric protein complexes in oral squamous cell carcinoma (OSCC) and to evaluate their potential as potential biomarkers. The expression levels of AIM 2 and TM4SF19 in tumor and normal oral mucosa tissues were determined by immunohistochemistry, and their mRNA and protein levels were quantified by quantitative real-time PCR (qRT-PCR) and Western blot. Cell models for overexpression and knockdown of AIM 2 and TM4SF19 were constructed to investigate their effects on the proliferation, migration and invasion of OSCC cells. Finally, the function of AIM 2 and TM4SF19 in OSCC cells was further investigated by the apoptosis assay and cell cycle analysis. It was found that compared with normal oral mucosa, the expression of AIM2 in OSCC was significantly down-regulated, while the expression of TM4SF19 showed the opposite trend, that is, it was up-regulated in tumor tissues. qRT-PCR and Western blot results further confirmed this finding and showed that AIM2 and TM4SF19 expression levels were closely correlated with clinical stage and lymph node metastasis in OSCC patients. The overexpression of TM4SF19 promoted the malignant behavior of tumor cells. In contrast, the knock down of AIM2 and the knock down of TM4SF19 produced the opposite effect. Therefore, AIM2 and TM4SF19 are expected to be potential biomarkers for OSCC, providing new ideas for the early diagnosis and treatment of OSCC.

Psychiatric disorders are highly inheritable, and most psychiatric disorders exhibit genetic overlap. Recent studies associated the 3q29 recurrent deletion with schizophrenia (SCZ) and autism spectrum disorder (ASD). In this study, we investigated the association of genes in the 3q29 region with SCZ and ASD. TM4SF19 and PAK2 were chosen as candidate genes for this study based on evidence from previous research. We sequenced TM4SF19 and PAK2 in 437 SCZ cases, 187 ASD cases and 524 controls in the Japanese population. Through targeted sequencing, we identified 6 missense variants among the cases (ASD & SCZ), 3 missense variants among controls, and 1 variant common to both cases and controls; however, no loss-of-function variants were identified. Fisher's exact test showed a significant association of variants in TM4SF19 among cases (p=0.0160). These results suggest TM4SF19 variants affect the etiology of SCZ and ASD in the Japanese population. Further research examining 3q29 region genes and their association with SCZ and ASD is thus needed.

TM4SF19 controls GABP-dependent YAP transcription in head and neck cancer under oxidative stress conditions

Tobacco and alcohol are risk factors for human papillomavirus–negative head and neck squamous cell carcinoma (HPV − HNSCC), which arises from the mucosal epithelium of the upper aerodigestive tract. Notably, despite the mutagenic potential of smoking, HPV − HNSCC exhibits a low mutational load directly attributed to smoking, which implies an undefined role of smoking in HPV − HNSCC. Elevated YAP (Yes-associated protein) mRNA is prevalent in HPV − HNSCC, irrespective of the YAP gene amplification status, and the mechanism behind this upregulation remains elusive. Here, we report that oxidative stress, induced by major risk factors for HPV − HNSCC such as tobacco and alcohol, promotes YAP transcription via TM4SF19 (transmembrane 4 L six family member 19). TM4SF19 modulates YAP transcription by interacting with the GABP (Guanine and adenine-binding protein) transcription factor complex. Mechanistically, oxidative stress induces TM4SF19 dimerization and topology inversion in the endoplasmic reticulum membrane, which in turn protects the GABPβ1 subunit from proteasomal degradation. Conversely, depletion of TM4SF19 impairs the survival, proliferation, and migration of HPV − HNSCC cells, highlighting the potential therapeutic relevance of targeting TM4SF19. Our findings reveal the roles of the key risk factors of HPV − HNSCC in tumor development via oxidative stress, offering implications for upcoming therapeutic approaches in HPV − HNSCC.
