Introduction

ABCG5 and ABCG8 form an obligate heterodimer that mediates Mg(2+)- and ATP-dependent sterol transport across the cell membrane (PubMed:27144356). Plays an essential role in the selective transport of dietary plant sterols and cholesterol in and out of the enterocytes and in the selective sterol excretion by the liver into bile (PubMed:11099417, PubMed:11138003, PubMed:27144356, PubMed:15054092). Required for normal sterol homeostasis (PubMed:11099417, PubMed:11138003, PubMed:15054092). The heterodimer with ABCG8 has ATPase activity (PubMed:16893193, PubMed:20210363, PubMed:27144356).

Mechanism Analysis of Increased Erucic Acid Content in Brassica napus L. Seeds Resulting Low Nighttime Temperature

Brassica napus L. (B. napus L.), a crop of Brassica in the family Cruciferae, is a major sources of edible vegetable oil and one of the four most widely grown. Oil accumulation is determined by genetic and environmental factors, including temperature, light, humidity, edatope, latitude, and altitude. Temperature also plays a crucial part in the maturation stage. A highly temperature-sensitive line (STSL, DH0729DH0815) and weakly temperature-sensitive line (WTSL, DH0729) were used to express LOC106368911 and its effect on the erucic acid content of seeds under low nighttime temperatures. Condition of 20/18 °C (±0.5 °C, daytime/nighttime temperature, CK) and 20/13 °C (low nighttime temperature, LNT) were used in tests. Under LNT, the erucic acid content in STSL seeds increased significantly, whereas the WTSL change was not significant. The relative expression of LOC106368911 was significantly increased in STSL at 27, 35 and 43 days after flowering (DAF), whereas the change in WTSL was not significant. The CDS sequence of the LOC106368911 was cloned. Compared to the protein sequence encoded by the reference gene, STSL changed the 96th amino acid sequence from I (leucine) to L (isoleucine). However, there was no difference in the secondary and tertiary structures. Based on this, we cloned the LOC106368911 promoter sequence and found that the mutation of C to T at position -790 in WTSL caused the loss of LTR (cis-acting element involved in low-temperature responsiveness, -791 to -786) element function in response to low temperature and increased erucic acid content. The LOC106368911 promoter had 2 LTR elements in STSL (-791 to -786 and -588 to -583). A GUS reporter vector was constructed to study the transient expression in tobacco leaf transformations. GUS gene expression at 13 ℃ after 2 h was significantly higher than that at 18 ℃. Base and number differences in the LTR element were found in the LOC106368911 promoter sequence of STSL and WTSL. Base mutations occurred in the LTR element in WTSL, which resulted in decrease or loss of erucic acid content in response to low nighttime temperatures.

Caveolin-1 mitigates the advancement of metabolic dysfunction-associated steatotic liver disease by reducing endoplasmic reticulum stress and pyroptosis through the restoration of cholesterol homeostasis

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease worldwide, which has the potential to advance to fibrosis. CAV1 has the effects of improving liver lipid deposition in MASLD, however, the potential mechanism is largely unknown. Here, we establish a MASLD mouse model in CAV1 knockout (KO) mice and perform transcriptome analysis on livers from mice to investigate the effects of CAV1 in MASLD progression. In addition, we evaluated the expression of CAV1 in human liver samples, and also conducted assays in vitro to investigate the molecular role of CAV1 in MASLD progression. The results illustrate that the expression of liver CAV1 in the decreases during MASLD progression, which aggravates the accumulation of cholesterol in the liver, leading to more severe endoplasmic reticulum (ER) stress and pyroptosis. Mechanistically, CAV1 regulates the expression of FXR/NR1H4 and its downstream cholesterol transporter, ABCG5/ABCG8, suppressing ER stress and alleviating pyroptosis. Our study confirms CAV1 is a crucial regulator of cholesterol homeostasis in MASLD and plays an important role in disease progression.

Evaluation of the Decreased Cholesterol Potential of Levilactobacillus brevis M-10 Isolated from Spontaneously Fermented Sour Porridge in Mice with High-Cholesterol Levels

Excessive cholesterol levels can lead to hypercholesterolemia, which is related to cardiovascular diseases (CVDs), and CVDs are a serious threat to human health. Therefore, lowering cholesterol levels is necessary, and diet intervention is safer than drugs are. The cholesterol-lowering effect of Levilactobacillus brevis M-10 isolated from spontaneously fermented millet sour porridge was investigated in fifty C57BL/6N male mice. After a 4-week intervention, the food intake, weight gains and organ indices were calculated; the lipid contents in the serum, liver, and feces were determined; the histopathology of the liver tissues was observed; the expression of metabolism-related genes was determined; and short-chain fatty acid (SCFA) levels in the droppings were monitored. The results showed that administration of a high dose of L. brevis M-10 (1 × 10¹⁰ CFU/mL) significantly reduced food intake, suppressed weight gain; prevented excessive liver growth; and reduced the total serum cholesterol, triglycerides, low-density lipoproteins; and total hepatic cholesterol and triglyceride contents (P < 0.05) in high-cholesterol mice. Moreover, a high dose of L. brevis M-10 significantly promoted the fecal excretion of cholesterol and triglycerides (P < 0.05) and alleviated liver damage induced by a high-cholesterol diet. Furthermore, a high dose of L. brevis M-10 significantly downregulated the cholesterol metabolism-related gene expression of NPC1L1, ACAT2, HMG-CoA, and SREBP2 but upregulated the gene expression of ABCG5, CYP7A1, and LXR-α (P < 0.05). Additionally, a high dose of L. brevis M-10 significantly increased SCFA contents, including those of acetic acid, propionic acid and n-butyric acid (P < 0.05). These findings could provide support for the use of L. brevis M-10 in the application of functional foods to alleviate hypercholesterolemia.
