Sodium and pyrimidine nucleoside symporter of the plasma membrane that imports uridine, thymidine and cytidine into cells by coupling their transport to the transmembrane sodium electrochemical gradient. Also transports adenosine, an atypical substrate transported with high apparent affinity, but low maximum velocity. Identification and validation of SLC16A8 as a prognostic biomarker in clear cell renal cell carcinoma: a six-gene solute carrier signature

Solute carrier (SLC) proteins are essential for nutrient transport, influencing tumor metabolism and growth while preserving cellular homeostasis. Despite the critical biological functions of these transporters, their applicability as therapeutic targets in clear cell renal cell carcinoma (ccRCC) remains largely unexplored. In the current study, we analyzed transcriptomic data and discovered 77 differentially expressed SLC genes in ccRCC, with 24 demonstrating predictive potential. Using Lasso regression, we developed a prognostic signature comprising six key genes: SLC2A3, SLC11A1, SLC14A1, SLC16A8, SLC22A6, and SLC28A1. This signature demonstrated strong diagnostic performance and served as an independent predictor of patient survival. Further analysis integrating clinical variables and risk scores enabled the construction of nomograms, which exhibited high predictive accuracy for patient outcomes. Immune profiling revealed distinct infiltration patterns between risk groups: high-risk patients showed elevated levels of memory B cells, activated CD4+ T cells, regulatory T cells (Tregs), M0 macrophages, and neutrophils. In contrast, their low-risk counterparts showed M1 macrophages, resting dendritic cells, and resting mast cells. Validation experiments confirmed that SLC16A8 was significantly overexpressed in ccRCC tissues compared to normal samples, correlating with poor prognosis. Functional studies demonstrated that SLC16A8 knockdown impaired tumor progression in vitro. Consistent with these findings, in vivo experiments demonstrated reduced tumor growth upon SLC16A8 knockdown. Mechanistically, decreased SLC16A8 attenuated PI3K/AKT signaling, suggesting a potential regulatory pathway in ccRCC progression. In summary, we established a six-gene SLC signature with significant prognostic value in ccRCC. Among these genes, SLC16A8 emerged as a promising biomarker and therapeutic target, warranting further investigation.

Human equilibrative nucleoside transporter 1 and concentrative nucleoside transporter 1 in colorectal cancer: What do we know? A systematic review.

Colorectal cancer (CRC) remains a major global health challenge despite advances in screening, diagnosis, and treatment. This systematic review examines the roles of Human Equilibrative Nucleoside Transporter 1 (hENT1) and Human Concentrative Nucleoside Transporter 1 (hCNT1) in CRC, focusing on their expression, regulation, and impact on chemotherapeutic efficacy, particularly with nucleoside analogues like 5-fluorouracil (5-FU). We conducted a comprehensive literature search following PRISMA guidelines, yielding 29 studies that met our inclusion criteria. The review reveals variable expression of hENT1 and hCNT1 in CRC tissues compared with normal tissues, with implications for treatment response and development of resistance. Increased hENT1 expression is associated with poor outcomes and resistance to 5-FU, suggesting its potential as a biomarker for predicting treatment response. Conversely, hCNT1's role appears more complex, with its expression influencing the efficacy of other chemotherapeutic agents like gemcitabine and capecitabine. The review also highlights the lack of robust, standardised methods for assessing mRNA and protein levels, which complicates the interpretation of data and the establishment of these transporters as reliable clinical markers. Key findings include the potential therapeutic benefits of modulating hENT1 and hCNT1 expression to enhance drug efficacy and overcome resistance. The study underscores the need for further research using standardised and advanced methodologies, such as 3D cell culture assays, to better understand the mechanistic pathways and clinical implications of nucleoside transporter expression in CRC. Future research should aim to clarify the roles of hENT1 and hCNT1 in CRC and chemoresistance to develop targeted therapies and improve patient outcomes.

Functional Validation of Doxorubicin-Induced Cardiotoxicity-Related Genes

BackgroundGenome-wide association studies and candidate gene association studies have identified more than 180 genetic variants statistically associated with anthracycline-induced cardiotoxicity (AIC). However, the lack of functional validation has hindered the clinical translation of these findings.ObjectivesThe aim of this study was to functionally validate all genes associated with AIC using human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs).MethodsThrough a systemic literature search, 80 genes containing variants significantly associated with AIC were identified. Additionally, 3 more genes with potential roles in AIC (GSTM1, CBR1, and ERBB2) were included. Of these, 38 genes exhibited expression in human fetal heart, adult heart, and hiPSC-CMs. Using clustered regularly interspaced short palindromic repeats/Cas9-based genome editing, each of these 38 genes was systematically knocked out in control hiPSC-CMs, and the resulting doxorubicin-induced cardiotoxicity (DIC) phenotype was assessed using hiPSC-CMs. Subsequently, functional assays were conducted for each gene knockout on the basis of hypothesized mechanistic implications in DIC.ResultsKnockout of 26 genes increased the susceptibility of hiPSC-CMs to DIC. Notable genes included efflux transporters (ABCC10, ABCC2, ABCB4, ABCC5, and ABCC9), well-established DIC-associated genes (CBR1, CBR3, and RAC2), and genome-wide association study-discovered genes (RARG and CELF4). Conversely, knockout of ATP2B1, HNMT, POR, CYBA, WDR4, and COL1A2 had no significant effect on the in vitro DIC phenotype of hiPSC-CMs. Furthermore, knockout of the uptake transporters (SLC28A3, SLC22A17, and SLC28A1) demonstrated a protective effect against DIC.ConclusionsThe present findings establish a comprehensive platform for the functional validation of DIC-associated genes, providing insights for future studies in DIC variant associations and potential mechanistic targets for the development of cardioprotective drugs.
