Introduction

Proton-coupled monocarboxylate symporter. Catalyzes the rapid transport across the plasma membrane of monocarboxylates such as L-lactate, pyruvate and ketone bodies, acetoacetate, beta-hydroxybutyrate and acetate (PubMed:32415067, PubMed:9786900). Dimerization is functionally required and both subunits work cooperatively in transporting substrate (PubMed:32415067).

Effects of decrease in Klotho protein expression on insulin signaling and levels of proteins related to brain energy metabolism

Mutations in Klotho have been associated with premature ageing and cognitive dysfunction. Although highly expressed in specific regions of the brain, the actions of Klotho in the central nervous system (CNS) remain largely unknown. Here, we show that animals with a mutated hypomorphic Klotho gene have altered glycaemic regulation, suggesting higher insulin sensitivity. In the CNS, pathways related to insulin intracellular signalling were found to be up-regulated in the hippocampus, with higher activation of protein kinase B and mammalian target of rapamycin and inactivation of the transcription factors forkhead box O (FOXO)-1 and FOXO-3a. In addition, the present study showed that in the hippocampi of wild-type aged mice, where Klotho is naturally downregulated, the levels of some proteins related to energy metabolism and metabolic coupling between neurones and astrocytes, such as monocarboxylate transporter 2 and 4, 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase isoform 3 and lactate dehydrogenase enzymes isoforms A and B were altered. These findings suggest that Klotho plays an essential role in regulating proteins and genes related to metabolic coupling in the brain.

Muscle‐Derived miR‐200a‐3p Through Light‐Intensity Exercise May Contribute to Improve Memory Dysfunction in Type 2 Diabetic Mice

ABSTRACTMemory dysfunction associated with type 2 diabetes mellitus (T2DM) poses a threat to well‐being. Engaging in light‐intensity exercise has favorable effects on hippocampal function and molecular profiles, including Mct2 mRNA and miR‐200a‐3p. Here, we investigated the involvement of exosomal miR‐200a‐3p secretion from gastrocnemius muscles in T2DM mice undergoing light‐intensity exercise intervention, focusing on its potential to ameliorate memory dysfunction. We initially assessed the effects of light‐intensity exercise over a 4‐week period on memory function, the secretion of gastrocnemius muscle‐derived exosomal miR‐200a‐3p, and hippocampal mRNA. Subsequently, the impact of a daily intraperitoneal injection of the mmu‐miR‐200a‐3p mimic over a 4‐week duration on hippocampal dysregulation in ob/ob mice was investigated. The light‐intensity exercise intervention improved gastrocnemius muscle‐derived and plasma exosomal miR‐200a‐3p levels in ob/ob mice, concomitant with improved memory dysfunction. Intriguingly, the daily intraperitoneal injection of the mmu‐miR‐200a‐3p mimic also improved memory function in ob/ob mice. Notably, both the exercise intervention and miR‐200a‐3p mimic treatment induced downregulation in hippocampal Keap1 and upregulation in Hsp90aa1 and Mct2 mRNA in ob/ob mice. These results imply that the augmentation of peripherally derived miR‐200a‐3p contributes to ameliorating memory dysfunction in T2DM mice undergoing light‐intensity exercise, with a possible contribution from gastrocnemius muscle‐derived exosomal miR‐200a‐3p to these exercise effects.

RPE-specific MCT2 expression promotes cone survival in models of retinitis pigmentosa

Retinitis pigmentosa (RP) is the most common cause of inherited retinal degeneration worldwide. It is characterized by the sequential death of rod and cone photoreceptors, the cells responsible for night and daylight vision, respectively. Although the expression of most RP genes occurs only in rods, there is a secondary degeneration of cones. One possible mechanism of cone death is metabolic dysregulation. Photoreceptors are highly metabolically active, consuming large quantities of glucose and producing substantial amounts of lactate. The retinal pigment epithelium (RPE) mediates the transport of glucose from the blood to photoreceptors and, in turn, removes lactate, which can influence the rate of consumption of glucose by the RPE. One model for metabolic dysregulation in RP suggests that following the death of rods, lactate levels are substantially diminished causing the RPE to withhold glucose, resulting in nutrient deprivation for cones. Here, we present adeno-associated viral vector-mediated delivery of monocarboxylate transporter 2 (MCT2, Slc16a7 ) into the eye, with expression limited to RPE cells, with the aim of promoting lactate uptake from the blood and encouraging the passage of glucose to cones. We demonstrate prolonged survival and function of cones in rat and mouse RP models, revealing a possible gene-agnostic therapy for preserving vision in RP. We also present the use of fluorescence lifetime imaging-based biosensors for lactate and glucose within the eye. Using this technology, we show changes to lactate and glucose levels within MCT2-expressing RPE, suggesting that cone survival is impacted by changes in RPE metabolism.
