Introduction

A syndrome characterized by recurring fever, rash, and arthralgias occurring days to weeks after a rat bite. The causative agents are either Streptobacillus moniliformis or Spirillum minus.

Multicenter Retrospective Study on Streptobacillus Moniliformis Infections in France

French data on Streptobacillus moniliformis infections (zoonosis transmitted by rat bites) are rare and there are no case series in the literature to date. This is a pathology presenting atypical clinical manifestations for a bacterial infection and which therefore remains underdiagnosed. Streptobacillus moniliformis infections can be fatal without appropriate antibiotic treatment. This study could improve diagnosis, develop early treatment strategies and contribute to the overall understanding of zoonotic diseases

Molecular survey of vector-borne agents in lowland tapirs (Tapirus terrestris) from Brazil reveals a new Anaplasma genotype

Although vector-borne agents have been detected in different species of wild animals, studies involving tapirs (Tapirus terrestris), the largest land mammals in Brazil, are scarce. The aim of the present study was to investigate the occurrence and molecular identity of Anaplasmataceae agents, Coxiella burnetii and Hepatozoon spp. in blood samples of wild T. terrestris from two biomes (Cerrado and Pantanal) in Brazil. A total of 122 blood samples from 99 tapirs were analyzed. Sixty-one tapirs were sampled in Pantanal, whereas 38 were from Cerrado biome. DNA was extracted from blood samples and subjected to conventional and/or quantitative PCR assays for molecular screening and characterization of DNA from Anaplasmataceae agents (Anaplasma, Ehrlichia, and Neorickettsia), C. burnetii and Hepatozoon spp. None of the samples were positive for Ehrlichia, C. burnetii or Hepatozoon spp. Twenty-two samples (22/122; 18%) amplified fragments from the expected size for the Anaplasma 16S rRNA fragment tested herein. Out of these samples, 2 (9.1%) presented amplification for the Anaplasma ITS 23S-5S. Nine positive samples for the 16S rRNA assay were selected for cloning and sequencing. Phylogenetically, distance and haplotype analyses based on large fragments (>1,200 bp) of the 16S rRNA suggest that tapir-related Anaplasma and Anaplasma odocoilei are genetically similar species. Moreover, 31 (25.4%) samples were positive for Neorickettsia based on amplification of partial 16S rRNA. Phylogenetic assessment of the three obtained sequences demonstrated relatedness to Neorickettsia risticii, the causative of Potomac fever in horses. This is the first report of Neorickettsia sp. and description of a new Anaplasma genotype in tapirs.

Investigating Xiaochaihu Decoction's fever-relieving mechanism via network pharmacology, molecular docking, dynamics simulation, and experiments

Xiaochaihu Decoction（XCHD）is a classic prescription for the treatment of fever, but the mechanism is not clear. In this study, We elucidated the mechanism of action through network pharmacology and molecular docking. A rat fever model was established to verify the prediction results of network pharmacology. The analysis revealed that 120 intersection targets existed between XCHD and fever. The TP53, STAT3, RELA, MAPK1, AKT1, TNF and MAPK14 as potential core targets of XCHD in fever treatment. GO and KEGG pathway enrichment analyses indicated that XCHD may act through pathways such as the AGE-RAGE signaling pathway in diabetic complications, TNF signaling pathway, IL-17 signaling pathway. Molecular docking results demonstrated that quercetin, kaempferol, β-sitosterol, stigmasterol and baicalein exhibited strong binding activity to key targets. Animal experiments showed that XCHD significantly reduced body temperature and levels of IL-1β, IL-6, TNF-α, NO, PGE2, and cAMP in rats with fever. Importantly, no significant difference was observed between the XCHD self-emulsifying nano phase plus suspension phase and XCHD group. XCHD exerts its therapeutic effects on fever through a multi-ingredient, multi-target, and multi-pathway approach.

[Chemical profiling of Xiaochaihu Granules and characterization of absorbed components in rat plasma after oral administration].

The chemical components of Xiaochaihu Granules and absorbed components in rats after oral administration were identified by using ultra performance liquid chromatography-quadrupole orbitrap mass spectrometry(UPLC-Q-Exactive-Orbitrap-MS)and UPLC-triple quadrupole mass spectrometry(UPLC-MS/MS). Separation was performed on a CORTECS UPLC C~+_(18)(2.1 mm×100 mm, 1.6 μm)column with gradient elution using acetonitrile-0.1% formic acid aqueous solution as the mobile phase. Data on the chemical components were collected in positive and negative ion modes and identified based on the retention time, precise molecular weight, fragment ion information in comparison with the reference substance, and literature report. The rat fever model was established by subcutaneous injection of dry yeast. Subsequently, the normal and model rats received oral administration of Xiaochaihu Granules. Blood samples were taken from the orbital vein at different time points after administration, and the plasma was isolated for scanning and identification of absorbed components using the multi reaction monitoring mode(MRM).A total of 112 chemical components were identified in Xiaochaihu Granules, including 63 flavonoids, 31 saponins, 6 organic acids, 4 phenylpropanoids, 3 amino acids and 5 other compounds. Additionally, 18 prototypical components were identified in rat plasma. This study lays the foundation for further study of the therapeutic material and quality control of Xiaochaihu Granules.
