The prevalence of obesity in Mexico is 35.4%, and it is considered a risk factor for the development of diabetes, systemic arterial hypertension and dyslipidemia. Obesity due to the increased distribution and growth of adipose tissue creates a pro-inflammatory state induced by molecules secreted by the adipocytes themselves. Netrin-1 is a cell migration protein, which directs the recruitment, migration and entrapment of macrophages in different tissues, within adipose tissue the entrapment of macrophages induces the release of pro-inflammatory cytokines, which increase the secretion of pro-inflammatory adipokines. It has been found in high concentration in patients with obesity, insulin resistance and type 2 diabetes. Thymol is a phytopharmaceutical derived from oregano oil that has shown powerful anti-inflammatory and antioxidant effects through the stimulation of PPAR-gamma, adiponectin and inhibition of the NF-κB pathway mediated by the JNK pathway, pathways in which netrin-1 is involved in macrophage entrapment and recruitment.

An intelligent thymol/alizarin-loaded polycaprolactone/gelatin/zein nanofibrous film with pH-responsive and antibacterial properties for shrimp freshness monitoring and preservation

Improper storage methods cause food resources to be wasted, and the development of multifunctional intelligent packaging can realize freshness monitoring and extend the shelf life. In this study, an intelligent alizarin/thymol-loaded polycaprolactone/gelatin/zein nanofibrous film was prepared and achieved the dual functions of pH-responsive and antibacterial properties. The film was fabricated using electrospun technology and characterized by SEM, FT-IR, WCA, TGA, DSC, and mechanical property tests, which had good antioxidant properties (81.40 ± 0.74 % free radical scavenging). The film could effectively inhibit the growth of Escherichia coli, Staphylococcus aureus, and Shewanella putrefaciens within 12 h and exhibited light yellow-light purple-dark purple color change at pH 2-12, with good color stability and color responsiveness to ammonia in the environment. In the shrimp application, shelf life was extended by 2 days and freshness could be monitored by the color change. This scientific research opens up new possibilities for realizing multifunctional food packaging alternatives.

Combining antibacterial and wound healing features: Xanthan gum/guar gum 3D-printed scaffold tuned with hydroxypropyl-β-cyclodextrin/thymol and Zn2+

Biofilm formation on biological and material surfaces represents a heavy health and economic burden for both patient and society. To contrast this phenomenon, medical devices combining antibacterial and pro-wound healing abilities are a promising strategy. In the present work, Xanthan gum/Guar gum (XG/GG)-based scaffolds were tuned with thymol and Zn²⁺ to obtain wound dressings that combine antibacterial and antibiofilm properties and favour the healing process. The tuning process preserved the 3D extrusion-based printability of the XG/GG ink. Scaffolds swelling profile was assessed in PBS pH 7.4, and the resistance to compressive forces was studied using a texturometer. The scaffolds microarchitectures were analyzed by SEM, while ATR-FTIR spotlighted the chemical modifications of the customized materials. Thymol and Zn²⁺ release was analyzed in biologically relevant media, showing a burst release in the first hours. The antibacterial properties were confirmed against S. aureus, P. aeruginosa, and S. epidermidis by isothermal microcalorimetry and biofilm viable cell counting. Incorporation of hydroxypropyl-β-cyclodextrin (HPβCD) improved thymol loading (7- and 14-times higher thymol content) and enhanced the antimicrobial and antioxidant performances of the dressing, while the presence of Zn²⁺ strongly potentiated the antimicrobial activity, showing a potent antibiofilm response in both Gram-positive and Gram-negative strains of clinical concern. The thymol and Zn²⁺ combination led to a reduction of 99.95 %, 99.99 %, and 98.26 %, of biofilm formation against S. aureus, P. aeruginosa, and S. epidermidis, respectively. Furthermore, the scaffolds demonstrated good hemocompatibility, cytocompatibility, tissue integration and pro-angiogenic features in an in ovo CAM model.

Neuroprotective Effects of Thymol and p-Cymene in Immobilized Male rats through Alterations in Molecular, Biochemical, Histological, and Behavioral Parameters

The research was conducted to examine the neuroprotective effect of thymol and its precursor p-cymene on chronic immobility stress in adult male Wistar rats. The rats were subjected to 2.5 h of stress every day for 14 consecutive days by placing them inside a restrainer. Thymol (10 mg/kg) and p-cymene (50 mg/kg) were given to the rats during the same period. The results showed that thymol and p-cymene prevented the increase of MDA level, decline of GSH level, and decrease of SOD and GPx activity in the hippocampus of rats exposed to stress. These monoterpenes also prevented the increase in the expression of Tnfa, Il1b, Tlr4, and Nfkb, and the decrease in the expression of Nrf2, Ho1, and Bdnf. In addition, thymol and p-cymene inhibited the increase in the expression and activity of acetylcholinesterase in the hippocampus of animals exposed to immobility and enhanced the expression of A7nachr. They also reduced neuronal death in the CA1 region of stressed animals and improved their performance in the Morris water maze and elevated plus maze tests. Based on these findings, thymol and p-cymene may be effective in preventing neurodegenerative diseases as they reduce oxidative stress and neuroinflammation, strengthen ACh signaling, and stimulate Bdnf expression.
