Protection of dauricine and daurisoline on PC12 cells damaged by glutamate or Aβ25-35

Glutamate (Glu) excitotoxicity and amyloid-β (Aβ) deposition are significant factors in the occurrence and development of Alzheimer's disease (AD). Dauricine and daurisoline are two alkaloid components of Menispermum dauricum DC. that have a protective effect on the nervous system. The protection of dauricine and daurisoline on Glu-injured PC12 cells and the protection dauricine on Aβ_25-35-injured PC12 cells were investigated in this study. The results of the study demonstrated that on PC12 cells damaged by Glu (20 mM), dauricine and daurisoline (3 and 10 μM) increased the cell viability, reduced cell apoptosis, and enhanced mitochondrial membrane potential (MMP) levels. Dauricine and daurisoline can also reduce the levels of intracellular ROS and free Ca²⁺, and suppression the expression of CaM, p-CaMKII, and p-Tau in Glu-damaged PC12 cells. In addition, on PC12 cells damaged by Aβ_25-35 (30 μM), dauricine (3 and 10 μM) can also significantly increase the cell viability and MMP levels, inhibit cell apoptosis, reduce intracellular ROS and free Ca²⁺ levels, and down-regulate protein expression of CaM, p-CaMKII, and p-Tau. This study demonstrates for the first time that dauricine and daurisoline may inhibit the excessive phosphorylation of Tau protein and subsequent cell apoptosis by suppressing the Ca²⁺-CaM/CaMKII pathway, thereby protecting PC12 cells damaged Glu or Aβ_25-35in vitro. Dauricine and daurisoline have the potential to treat AD effectively and have further research value.

Metabolomics combined with biochemical analyses revealed phenolic profiles and antioxidant properties of rapeseeds

Phenolic compounds, one of the most crucial lipid concomitants in rapeseed, have garnered heighten attention due to their numerous health benefits. Therefore, efficiently characterizing the phenolic profile of rapeseed is paramount for discerning their potential bioactivities. This study employed untargeted metabolomics in conjunction with molecular networking to trace the phenolic composition across three rapeseed genotypes. A total of 117 phenolic compounds were identified in rapeseed by mass spectrometry under positive and negative ionization modes, including 36 flavonoids, 23 coumarins, 12 phenolic acids, 10 lignans, 4 stilbenes, 4 diarylheptanes, 1 tannin, and several other phenolic constituents. Biochemical analyses revealed that Brassica napus rapeseed typically exhibited the highest total phenolic content and total flavonoid content as well as the strongest antioxidant capacity among three rapeseed genotypes. Through correlation analysis, 17 potential antioxidant phenolic compounds were tentatively screened from rapeseed, supporting the development and utilization of natural antioxidants from rapeseed.

Study of the multitarget mechanism of Astragalus (HUANGQI) in the treatment of Alzheimer’s disease based on network pharmacology and molecular docking technology

CONTEXTIn China, HUANGQI is widely used for the treatment of Alzheimer's disease (AD). However, a comprehensive understanding of its mechanism of anti-AD effects is lacking.OBJECTIVETo explore the active ingredients of HUANGQI and its potential targets and mechanisms of action in AD.MATERIALS AND METHODSThe active ingredients and targets of HUANGQI were screened from databases (TCSMP, ETCM, and BATMan), and AD-related genes were obtained from DrugBank and GeneCards. The same target genes were screened, and a drug-target disease network was constructed. The PPI network was constructed and GO and KEGG pathway enrichment analyses of the targets. The Cell Counting Kit-8 (CCK-8) assay was used to determine suitable HUANGQI treatment concentrations for HT-22 cells between 0-480 μg/mL. CCK-8, FITC-phalloidin and propidium iodide (PI) assays were used to examine the protective effect of (0, 60, 120, 240 μg/mL) of HUANGQI on 20 μM Aβ1-42-induced HT-22 cell cytotoxicity.RESULTSTwelve active ingredients of HUANGQI were selected, with 679 common targets associated with AD. GO and KEGG analysis revealed that the therapeutic mechanisms of HUANGQI involve TNF, AGE, the NF-κB pathway, and nuclear receptor activity-related processes. The CCK-8 assay indicated that HUANGQI was not cytotoxic to HT-22 cells at concentrations less than 240 μg/mL and was able to attenuate Aβ1-42-induced cellular damage (EC₅₀ = 83.46 μg/mL). FITC-phalloidin and PI assays suggested that HUANGQI could alleviate 20 μM Aβ1-42-induced neuronal cell cytotoxicity in a dose-dependent manner.CONCLUSIONHUANGQI has a protective effect on Aβ1-42-induced nerve cell injury; further mechanism research was needed.
