Repare Therapeutics announced that a combination of two targeted drugs resulted in confirmed responses of 19% and 17% in endometrial cancer and ovarian cancer, respectively, and now plans to take the regimen forward in one but not both diseases. In a Phase 1 study, Repare looked at an ATR inhibitor camonsertib plus a PKMYT1 inhibitor lunresertib in patients with endometrial cancer and platinum-resistant ovarian cancer harboring certain mutations, the company announced Thursday. Of 27 endometrial cancer patients, 5 had a confirmed response with 2 additional unconfirmed responses. And in 24 ovarian cancer patients, 4 had a response with 5 additional unconfirmed responses. Stifel analyst Benjamin Burnett said the data “came in below expectations.” Repare’s shares $RPTX fell 47% Friday morning to about $2 a share. It plans to start a Phase 3 study of lunresertib combined with camonsertib in endometrial cancer in the latter half of 2025. But will hold off in ovarian cancer. “While we’ve decided to put our initial focus and capital behind the [endometrial cancer] opportunity where we saw greater unmet need, [ovarian cancer] represents the first of several life cycle expansions,” Repare CEO Lloyd Segal said in an analyst call. Camonsertib inhibits ATR, which plays a role in the DNA damage response pathway, and lunresertib inhibits PKMYT1, which Repare says is what’s called a synthetic lethal partner to multiple cancer-driving mutations. Synthetic lethality, which Repare built its platform around, describes a phenomenon where changes to two genes together lead to cell death, while a mutation in either alone does not. Across 67 patients treated across tumor types at the Phase 2 dose, the most common adverse event was anemia, and 26.9% of patients had grade 3 anemia.
