The antinociceptive effects of matrine were identified in rats via intraperitoneal (i.p.) injection using the tail-immersion and formalin tests. Pharmacodynamic parameters for matrine against formalin-induced pain were calculated with nonlinear regression analysis. Inductively coupled plasma mass spectrometry (ICP-MS) technology was utilized to detect contents of the ionome in CSF and spinal cord. Variations in ionomic metabolism in different treated groups were examined using Pearson's correlation coefficients and principal component analysis (PCA).

RESULTS:

In the tail-immersion test, matrine significantly prolonged tail-flick latency in rats. Matrine also dose-dependently yielded analgesia against formalin-induced biphasic pain, with an onset at around 10 min post-injection and a duration of 100 min. The ED₅₀ and E_max values were 19.01 mg/kg and 71.86% for phase I and 40.30 mg/kg and 81.51% for phase II, respectively. Pearson's correlation coefficient study and PCA revealed significant reprogramming of ionomic metabolism in the CSF and the spinal cord in the NM (normal saline + matrine), NF (normal saline + formalin), and FM (formalin + matrine) groups, compared to the NN (normal saline + normal saline) group.

DISCUSSION AND CONCLUSIONS:

These findings broaden the known analgesic spectrum of matrine and provide novel insights into the involvement of ionomic metabolism in its analgesic efficacy.

In-depth characterization of quinolizidine alkaloids via highly efficient enrichment and multi-stage mass spectrometry

The identification of new natural products is a critical step in the discovery of lead compounds, and is a prerequisite for the development of novel pharmaceuticals. Traditional Chinese medicines (TCMs) are renowned for their diverse pharmacological activities, and represent a valuable source of novel chemicals. The quinolizidine alkaloids are abundant in plants of the Fabaceae family and exhibit significant bioactivities, including anticancer and anti-inflammatory effects. However, the structural diversity of quinolizidine alkaloids and the chemical complexity associated with TCMs pose great challenges in the discovery of novel quinolizidine alkaloids. In the present study, Sophora flavescens Ait. was used as a model to establish a comprehensive strategy for characterizing quinolizidine alkaloids. A simple and selective method was developed using an FC8HL column for the efficient enrichment of polar quinolizidine alkaloids. Through the integration of high-resolution multi-stage mass spectrometry and feature-based molecular networking, the enriched alkaloids were thoroughly characterized, including matrine-type, cystine-type, aloperine-type, anagyrine-type, lupinine-type, and dimeric species. A total of 186 quinolizidine alkaloids were identified, including 131 newly discovered compounds. A series of novel substituents was also identified for the first time. The findings of this study not only deepen our understanding of the structural diversity of quinolizidine alkaloids, but they also offer a novel research strategy for the comprehensive characterization of quinolizidine alkaloids in other plants, potentially facilitating the discovery of new drug candidates.

Matrine attenuates cerebral ischemia-reperfusion injury via UCP2/AMPKα-mediated suppression of oxidative stress, apoptosis, and neuroinflammation

Ischemic stroke remains a major contributor to disability and mortality, emphasizing the urgent need for new therapies to enhance reperfusion treatment outcomes. In this study, we systematically investigated the neuroprotective effects of matrine, an alkaloid derived from Sophora flavescens, using both computational and experimental approaches. Initially, a network pharmacology strategy was employed to identify 69 potential targets of matrine relevant to ischemic stroke by integrating data from Swiss Target Prediction, Similarity Ensemble Approach, PharmMapper, and GeneCards. Subsequent protein-protein interaction analyses, along with GO and KEGG pathway enrichment, highlighted the AMPK signaling pathway as a critical mediator. To validate these findings, male C57BL/6J mice were subjected to middle cerebral artery occlusion (MCAO) to induce stroke. Matrine treatment in this model resulted in significant improvements in neurological function, reductions in brain damage, and marked decreases in oxidative stress and apoptosis. Furthermore, molecular docking and molecular dynamics (MD) simulations studies revealed a strong binding affinity between matrine and key targets such as AMPK, CHRNA4, and SLC6A3, with Western blot analyses confirming the involvement of AMPK and its upstream regulator UCP2. Collectively, our results demonstrate that matrine confers neuroprotection in cerebral ischemia-reperfusion injury by modulating the AMPK pathway, suggesting its potential as a novel therapeutic agent for ischemic stroke. The proposed mechanisms of matrine in ischemic stroke are summarized in the graphical abstract.

Devisol Salmiakki has been on sale in Finland only starting from December 2021. Consumers are requested to return both opened and unopened Devisol Salmiakki products to the nearest pharmacy at the latest on 13 October 2022. The products will be reimbursed for the consumers. Empty packages or free samples will not be reimbursed. The recall does not concern Orion’s other Devisol products. The recall is due to levels of matrine in the licorice extract used in the Devisol Salmiakki product which have been found to exceed the maximum level allowed by the EU. Matrine occurs naturally in certain plants and is also used as a pesticide. Regarding matrine, there is no EU-level risk assessment and approval, and there is therefore no certainty about the safety of foodstuffs containing it. As a precaution, all batches of Devisol Salmiakki on the market are withdrawn. The daily recommended dose of Devisol Salmiakki is one tablet, in which case the exposure to matrine through this preparation is assumed to be low. Orion has not received any reports of adverse effects related to the Devisol Salmiakki product, and Orion is not aware of any consumer reports related to the safety of the Devisol Salmiakki product.
