Computational Screening of Microbial Metabolites as Erythropoietin (EPO) Mimetics for the Treatment of Anemia

Anemia remains a critical global health burden, often driven by impaired erythropoietin (EPO) signaling, which reduces red blood cell production. While recombinant EPO therapy is effective, its high cost and associated safety concerns limit its accessibility. This study explores microbial metabolites as affordable and safe alternatives that act as EPO mimetics that can bind and activate the erythropoietin receptor (EPOR). A computational screening of 90 microbial bioactive compounds was conducted, and from those, 16 were selected for detailed analysis. The extracellular domain of EPOR (PDB: 1EBP) was used as the target protein. Molecular docking was performed using AutoDock, followed by ADMET (Absorption, Distribution, Metabolism, Excretion, and Toxicity) profiling with SwissADME and ProTox-III. Protein-protein interaction (PPI) networks were also analyzed in Cytoscape, and the stability of the top complexes was validated via 100 ns molecular dynamics (MD) simulations. Docking results identified Abyssomicin W, Abyssomicin C, and Camptothecin as the top candidates with strong binding affinities (−7.60 kcal/mol) to EPOR. ADMET predictions confirmed their favorable drug-likeness and safety profiles, with Abyssomicin W exhibiting the most promising characteristics, including high gastrointestinal absorption, and no predicted hepatotoxicity or carcinogenicity. PPI network analysis underscored the functional relevance of EPOR in erythropoietic pathways, while molecular dynamics (MD) simulations revealed that Abyssomicin W and Camptothecin formed highly stable complexes with the receptor, whereas the Abyssomicin C complex was unstable. The integrated computational pipeline successfully identified Abyssomicin W as the most stable and promising EPO mimetic candidate. In conclusion, this study identifies Abyssomicin W as a potential and stable EPO mimetic candidate, highlighting the potential of microbial metabolites as cost-effective therapeutics for anemia. Further experimental validation, including direct binding and functional cell-based assays is recommended to confirm its efficacy and safety in biological systems.

Aim: Preexisting anti-polyethylene glycol (PEG) antibodies (APAs) may affect the efficacy and safety of PEGylated compounds. Omontys^® and Krystexxa^® withdrawal and SARS-CoV-2 RNA vaccine anaphylaxis have all been linked to APAs. This project aimed to develop and validate a method to detect total antibodies against PEG, pre- and post-dose. Materials & methods: The repetitive, linear PEG structure prevented the use of a bridging homogenous format, hence the requirement to use a solid-phase extraction and acid dissociation assay coupled with the Meso Scale Discovery^® platform. Results & conclusion: Using singlicate analysis, the method was validated to successfully detect APA pre- and post-dose, with a crucial aspect of the method being the preparation of an appropriate negative control.

Adeno-associated viral (AAV) capsids are an emerging vector technology for a number of novel gene therapy modalities (including transgene delivery and CRISPR gene editing). In this commentary, the proper approach to managing uncertainty (described by Rosenberg et al., 2012) when determining critical quality attributes is stated and applied retrospectively to Peginesatide and prospectively to AAV drug product integrity. With Peginesatide, the omission of advanced analytical techniques (for particles) led to a severe safety risk that appeared post marketing. Peginesatide was withdrawn from the market. One of the critical quality attributes of AAV capsid products is drug product integrity. Drug product integrity is critical because it is related to measuring the active dose of product and because the effects of empty, aggregated, particulate, and partial capsids on efficacy and safety are uncertain. The dose of an AAV capsid vector is typically measured as genomes per milliliter. Regulatory agencies have already recommended digital PCR (dPCR) methods because traditional real-time PCR methods were not precise enough for drug product characterization. However, even with dPCR methods, the practice of characterizing AAV product with a simple empty-full ratio is problematic because simplex dPCR methods count partial genomes as full genomes. The proper management of uncertainty requires a robust quantitative measurement of AAV drug product integrity to ensure control of efficacy and safety. Multiplex dPCR and SV-AUC are promising technologies that together have the potential to enable the development of assays fit for the purpose of measuring AAV drug product integrity more precisely.

Nektar Therapeutics (CA) Announces Initiation of Investigator-Initiated Trial Evaluating Etirinotecan Pegol (NKTR-102) as a Third-Line Treatment in Patients With Metastatic and Recurrent Non-Small Cell Lung Cancer (NSCLC)

SAN FRANCISCO, Feb. 5, 2013 /PRNewswire/ -- Nektar Therapeutics (NASDAQ:NKTR) announced today the start of a Phase 2 investigator-initiated study of etirinotecan pegol in patients with metastatic and recurrent NSCLC. The study is being conducted at the Abramson Cancer Center of the University of Pennsylvania under the direction of Charu Aggarwal, M.D., M.P.H., Assistant Professor of Oncology and Corey Langer, M.D., Professor of Oncology. "Patients with metastatic or recurrent non-small cell lung cancer currently have no standard treatment options after failure of second-line treatment," said Dr. Aggarwal. "In Phase 1 studies, etirinotecan pegol demonstrated encouraging anti-tumor activity in a broad range of solid tumors including non-small cell lung cancer. We are highly interested in evaluating etirinotecan pegol as a potential treatment option for this patient population." The primary endpoint of the Phase 2 study is overall response rate (ORR). Secondary endpoints include progression free survival (PFS), overall survival (OS), median duration of response (DoR) and the safety pro etirinotecan pegol in patients with NSCLC after failure of second-line therapy. The open label, single-arm trial is expected to enroll approximately 37 patients who will receive etirinotecan pegol once every three weeks as monotherapy. About NSCLC Lung cancer is the leading cause of cancer-related mortality in the US with NSCLC accounting for approximately 85% of all lung cancer diagnoses [1]. Most NSCLC patients present with loco-regionally advanced or metastatic disease. Platinum-based chemotherapy is the mainstay of treatment for NSCLC in the first-line setting for metastatic disease. Even after response to first-line therapy, patients eventually develop progressive disease and require additional treatment. Currently, pemetrexed, docetaxel, and erlotinib are the only FDA approved agents in the second line setting. Erlotinib is the only agent approved for use in the third-line setting, however, progression-free survival and overall survival remain suboptimal and there are no cytotoxics formally approved to treat patients in the third-line setting. Consequently, there is a need for new therapies that are safe and effective, which may improve overall survival and quality of life. About Etirinotecan Pegol Etirinotecan pegol is a unique, targeted topoisomerase I inhibitor designed for prolonged tumor cell exposure. Etirinotecan pegol is believed to penetrate the vasculature of the tumor environment more readily than normal vasculature, increasing the concentration of active drug within tumor tissue to enhance anti-tumor activity. The BEACON study is a Phase 3 clinical study currently evaluating etirinotecan pegol for the treatment of locally recurrent or metastatic breast cancer. Nektar Therapeutics (CA) Reports Third Quarter 2011 Financial Results

SAN FRANCISCO, Calif., Nov. 2, 2011 /PRNewswire/ -- Nektar Therapeutics (Nasdaq: NKTR) today reported its financial results for the third quarter ended September 30, 2011. Cash, cash equivalents, and investments at September 30, 2011 were $458.0 million as compared to $315.9 million at December 31, 2010. Revenue for the third quarter of 2011 was $27.1 million, a decrease as compared to $37.9 million in the third quarter of 2010 primarily as a result of the completion as of December 31, 2010 of the amortization of the $125.0 million upfront payment received in 2009 from AstraZeneca for the NKTR-118 and NKTR-119 license agreement. "Nektar is highly focused on continuing to advance our important proprietary drug candidates in pain and cancer," said Howard W. Robin, President and Chief Executive Officer of Nektar. "AstraZeneca's Phase 3 KODIAC program for NKTR-118 for opioid-induced constipation is continuing on-track with AZ targeting regulatory filing in 2013.
