Monitoring, Detoxifying, and Rebalancing Metals During Acute Myeloid Leukemia (AML) and Myelodysplastic Syndrome (MDS) Therapy

This phase I trial studies the side effects and best dose of edetate calcium disodium or succimer in treating patients with acute myeloid leukemia or myelodysplastic syndrome undergoing chemotherapy. Edetate calcium disodium or succimer may help to lower the level of metals found in the bone marrow and blood and may help to control the disease and/or improve response to chemotherapy.

Sulfur metabolism and response to light in Ulva prolifera green tides

The outbreak of Ulva prolifera blooms causes significant changes in the coastal sulfur cycle due to the high production of dimethylsulfoniopropionate (DMSP) and the emission of dimethylsulfide (DMS). However, the sulfur metabolism mechanism of U. prolifera has not been thoroughly investigated. In this study, we examined the levels of intracellular and extracellular sulfate (SO₄^2--S), total sulfur (TS), DMSP, and DMS in fresh U. prolifera under different light intensity conditions (54, 108 and 162 μmol photons m^-2 s^-1) during algal growth. We also conducted transcriptome analyses to investigate sulfur uptake and metabolism. When the light intensity increased by 50% (from 108 to 162 μmol photons m^-2 s^-1), the amount of absorbed SO₄^2--S increased by 3.5 times after 24 h, while the fresh weight of U. prolifera increased by 16%, and the average release rates of DMS and DMSP increased by 136% and 100%, respectively. However, the expression of sulfate transporter and assimilation-related genes did not show significant up- or down-regulation in response to the light intensity changes. Therefore, it is speculated that the key gene responsible for DMSP synthesis in U. prolifera has not yet been identified. The sulfate metabolic pathway of U. prolifera was established, and four Alma genes, including DMSP lyase, were identified. During the bloom period, it is estimated that U. prolifera releases a maximum of approximately 0.4 tons of sulfur and 0.3 tons of carbon in the form of DMS into the atmosphere per day. Additionally, biogenic sulfur dissolved in seawater or within algae could potentially impact the regional climate and environment.

Nano-alkaline ion-excited NETs ablative eye drops promote ocular surface recovery

Neutrophil extracellular traps (NETs) promote neovascularization during the acute phase after ocular chemical injury, while the local inflammatory acidic environment delays post-injury repair. Currently, the mechanism of NETs promoting neovascularization has not been fully elucidated, and there is a lack of therapeutic strategies to effectively improve the local microenvironment for corneal repair. In this study, we validated the NETs-M2-angiogenic pathway after injury. Using transcriptomics sequencing and liquid-phase microarray assays, the intrinsic immune cascade mechanism of NETs inducing macrophage M2 polarization and releasing VEGF via PI3K/AKT was identified. Based on this pathology and the physiological need to improve the local inflammatory acidic environment and promote corneal repair, we organically integrated the alkaline ion-rich bioglass with the highly transmissive and highly adhesive filipin protein, and constructed NETs ablative gel eye drops (DMS) that can release DNase I and alkaline ions in a sustained manner. The eye drops restricted the inflammatory interaction of NETs with macrophages from the source, adhered to the corneal surface and continuously released alkaline ions to improve the local acidic inflammatory environment, providing a favorable immune microenvironment for corneal recovery. We established a cell co-culture system and a corneal alkali burn model to further validate the role of DMS in modulating the intrinsic immune cascade of neovascularization for corneal repair and the related mechanisms. In conclusion, based on the biological mechanism of NETs-M2-VEGF after corneal chemical injury, the present study designed eye drops for dual regulation of intrinsic immunity and the inflammatory acid environment, which not only further supplemented and improved the pathophysiological mechanism of corneal neovascularization after chemical injury, but also provided a new way of thinking about corneal regeneration after injury.

Evaluating the impact of dose monitoring software alerts on radiation dose reduction in computed tomography: A systematic review

INTRODUCTIONRadiation Dose Monitoring Software (DMS) tools have been developed to monitor doses and alert computed tomography (CT) users of high radiation exposure. However, the causal factors for alerts and the impact of DMS in dose optimisation are poorly understood.AIMThis review aims to identify high-dose CT examinations triggering alerts and their determinants, and to assess if the alerts from DMS help to reduce CT dose levels.METHODSTo identify relevant articles published to December 2023, an electronic search of Medline, Scopus, CINAHL, Embase, and the Web of Science was undertaken. Reference lists of published articles were also assessed to identify further articles. The Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) methodology was employed to evaluate articles for relevance. Articles were included if they used the DMS tool to detect high-dose events that issued alerts in CT and were published in English.RESULTSThe search resulted in 83 articles, of which, nine were included after a thorough screening process. High dose alerts most often occurred in CT chest, CT head/brain, CT Chest/abdomen/pelvis, and CT abdomen/pelvis with alert percentages ranging from 1.45% to 10.21%, 1.54% to 4.18%, 4.48% to 6.60%, and 4.47% respectively. Alerts were mainly triggered by overweight patients, scan repetition, miscentering of the patients, extra CT study added, orthopaedic hardware in the scanning area and scanning beyond the desire anatomy. Most of the studies reviewed show that DMS tools played a role in reducing the number of high-dose events that trigger alerts.CONCLUSIONDMS tools are valuable in automatically identifying high-dose CT protocols, enabling quick investigation and dose optimisation. The high-dose events occurred due to patient and technical factors, which can be mitigated through proper monitoring and investment in both technological resources and staff training.
