A series of homotyrosine-substituted pneumocandin B₀ analogs, e.g. I [R = H, OH, R¹ = H, iodo, NO₂, NH₂, N₃; R = H, R¹ = CO₂Me, (E)-CH:CHCO₂Me, CH:CH₂, CH₂CH:CH₂, Et, n-Pr, CHO, OH, CH₂CHO, CH₂CH₂OH, (E)- and (Z)-CH₂CH:CHCO₂Me, CH₂CH₂NH₂; R = OH, R¹ = NHAc, NHCOCH₂CH₂CO₂H, NHCOCH₂SH, NHCOCH₂NH₂, NHCOCH₂CH₂NH₂], substituted at the 3'-position of the homotyrosine residue have been prepared and evaluated for their inhibition of 1,3-β-D-glucan synthesis and for their antifungal activity against C. albicans.Cationic analogs displayed enhanced antifungal properties.The phenolic hydroxyl is involved in a critical hydrogen-bond at the binding site of the enzyme.
