主要目的：观察中国健康受试者在空腹/餐后状态下单次口服受试制剂二甲双胍格列本脲片（规格：500mg/2.5mg）和参比制剂二甲双胍格列本脲片（商品名：Glucovance®；规格：500mg/2.5mg；持证商：Merck SANTE S.A.S）后的药代动力学特征，评价两种制剂在空腹/餐后状态下的生物等效性。次要目的：观察受试制剂二甲双胍格列本脲片和参比制剂二甲双胍格列本脲片（Glucovance®）在中国健康受试者中的安全性。

Primary objective: To observe the pharmacokinetic characteristics of the test preparation metformin glibenclamide tablets (specification: 500mg/2.5mg) and the reference preparation metformin glibenclamide tablets (trade name: Glucovance®; specification: 500mg/2.5mg; licensee: Merck SANTE S.A.S) after a single oral administration in the fasting/postprandial state in healthy Chinese subjects, and to evaluate the bioequivalence of the two preparations in the fasting/postprandial state. Secondary objective: To observe the safety of the test preparation metformin glibenclamide tablets and the reference preparation metformin glibenclamide tablets (Glucovance®) in healthy Chinese subjects.

Optimization of purification process for alkaloids from Strychnos nux-vomica in Yisui Tongjing prescription

The purification technol. conditions of alkaloids from Strychnos nux-vomica in Yisui Tongjing prescription by macroporous adsorption resin were optimized.With the contents of brucine and strychnine as indexes, which were determined by HPLC, macroporous adsorption resins were screened by static adsorption and desorption test, single factor test was used to investigate adsorption and elution conditions, such as adsorption speed, adsorption time, elution concentration, elution speed, the amount of elution and so on, to determine purification technol. of alkaloids from S. nux-vomica in Yisui Tongjing prescription.The optimum purification technol. was: type of macroporous resin was X-5, the concentration of sample solution 0.1 g·mL^-1, loading speed 1.0 mL·min^-1, sample volume 4 BV, adsorption time 2 h, washed with 1 BV water for eluting impurity, eluted with 6 BV 50% ethanol, elution rate 2.0 mL·min^-1.The optimized technol. was simple and economical, and it was feasible for taking macroporous adsorption resin to purify alkaloids from S. nux-vomica.

Quality control of Yisuitongjing release granules

Quality control of Yisuitongjing release granules was established.Antler and Glycyrrhiza uralensis were qual. identified by TLC; and content of strychnine and strychnine was determined by HPLC.Antler and Glycyrrhiza uralensis had clear spot by TLC and without neg. interference.Strychnine had good linear relationship within range of 0.007284 mg·mL^-1-0.182 100 mg·mL^-1 (r=0.9999), average recovery rate was 101.05% and RSD was 1.50%.Strychnine had good linear relationship within range of 0.00744 mg·mL^-1-0.18600 mg·mL^-1 (r=0.9996), average recovery rate was 100.87% and RSD was 1.25%.The method was simple, feasible, reproducible, stable and accurate, and it could be used in quality control of Yisuitongjing release granules.

Study on formalizing process and drug-releasing mechanism of Yisuitongjing sustained-release granules

The optimum process and drug-releasing mechanism of Yisuitongjing sustained-release granules were investigated.The forming process was selected using wet granulation technol. by single factor experiment to fit the drug releasing curve with math. model and judge the drug-releasing mechanism with the fitting result.The best molding process of Yisuitongjing sustained-release granules was: prescription herbs extract 81.25%, HPMC_K15M18.75%, with 80% ethanol as wetting agent, the granules was prepared by wet granulation; release behavior of brucine and strychnine in Yisuitongjing sustained release granules fit well with the Hi guchi model, indicating that the release behaviors of two alkaloids were in line with the Higuchi model, belonging to the diffusion release mechanism.The optimum process of Yisuitongjing sustained-release granules was simple and reasonable, and suitable for big industrial production
