The aim is to optimize extraction process of picroside I and picroside II from Picrorhiza scrophulariiflora Pennell by orthogonal design method. Conditions for the percolation were studied by orthogonal design as guided by total content of picroside I and picroside II. The orthogonal test was used in studying the effects of the particle, soaking time, solvent amount and flow rate on extraction technol. with 95% ethanol as solvent. The best condition for the permeating extraction is, pulverizing herb to 65 mesh, moistening with 95% ethanol, soaking for 12h, percolation extracting with 15 fold 95% ethanol at flow rate of 1ml/min. The extraction process is stable and reliable, and suitable for extraction of picroside I and picroside II in Picrorhiza scrophular ii flora Pennell, and can be used as reference for industrial production
