Enzymes acting on inulin (I) or sucrose (II) were studied.Three enzymes, fructosyltransferase (EC 2.4.1.99) (III), endo-inulinase (EC 3.2.1.7) (IV), and exo-inulinase (EC 3.2.1.80) (V) were separated from the fermentation liquor of A. ficuum.III acted on II to transfer its fructosyl residue to the fructosyl moiety of acceptor II-producing fructooligosaccharides.It showed maximum action at pH 5.0 and 50°.III could be used for producing Neo-sugar, a mixture of 1-kestose, nystose, etc.).IV converted I to a mixture of fructooligosaccharides, mainly inulotriose and inulotetraose.It had a optimum pH at 5.0 and optimum temperature of 55-60°.When used in combination with V, it provides an efficient system for producing fructose from I.

Cloning of a maltogenic alpha-amylase from Bacillus stearothermophilus

A novel maltogenic α-amylase from B. stearothermophilus was cloned and expressed on a plasmid in B. subtilis.Originally, the plasmid was very unstable in the absence of selection, but was stabilized due to a spontaneous, copy number reducing mutation.The promoter region and the extension of the gene were analyzed, and a provisional DNA sequence was determinedThe N-terminus of the mature amylase was determined and shown to possess a typical gram-pos. signal sequence of 33 amino acids which may be processed by signal peptidase.

Safety evaluation of lipase fromMucor mieheifor inter‐esterification of edible oils and fats

A new lipase of modification of edible oils and fats was subjected to toxicol. investigations.M. miehei Was classified as a weak pathogen to mice, with no evidence of pathogenic potential in healthy persons.The toxicol. studies on the enzyme included subchronic 13-wk feeding studies in rats and dogs, teratogenicity and 2-generation reproduction studies in rats, and mutagenicity studies with bacteria and rats.The no-effect levels from subchronic toxicity studies were approx. 1 g/kg body weight/day for rats and 4 g/kg body weight/day for dogs.There was no evidence of teratogenic or mutagenic potential and no effect on fertility and general reproductive performance.This lipase is suitable for the production of oils and fats for human consumption.
