In vitro antibody display and screening technologies geared toward the discovery and engineering of clinically applicable antibodies have evolved from screening artificial antibody formats, powered by microbial display technologies, to screening of natural, full-IgG molecules expressed in mammalian cells to readily yield lead antibodies with favorable properties in production and clinical applications. Here, we report the development and characterization of a novel, next-generation mammalian cell-based antibody display and screening platform called Transpo-mAb Display, offering straightforward and efficient generation of cellular libraries by using non-viral transposition technology to obtain stable antibody expression. Because Transpo-mAb Display uses DNA-transposable vectors with substantial cargo capacity, genomic antibody heavy chain expression constructs can be utilized that undergo the natural switch from membrane bound to secreted antibody expression in B cells by way of alternative splicing of Ig-heavy chain transcripts from the same genomic expression cassette. We demonstrate that stably transposed cells co-express transmembrane and secreted antibodies at levels comparable to those provided by dedicated constructs for secreted and membrane-associated IgGs. This unique feature expedites the screening and antibody characterization process by obviating the need for intermediate sequencing and re-cloning of individual antibody clones into separate expression vectors for functional screening purposes. In a series of proof-of-concept experiments, we demonstrate the seamless integration of antibody discovery with functional screening for various antibody properties, including binding affinity and suitability for preparation of antibody-drug conjugates.

Sortase enzyme-mediated generation of site-specifically conjugated antibody drug conjugates with high in vitro and in vivo potency

Antibody drug conjugates (ADCs) have recently been proven to be highly potent anti-tumor drugs, typically exceeding the efficacy of conventional monoclonal antibodies (mAbs). ADCs are currently produced by chemical conjugation of a small-molecule toxin to the mAb through lysine or cysteine side chains. This leads to heterogeneous mixtures of ADCs in which variable numbers of drugs are conjugated to individual antibodies and in which the site of conjugation cannot be defined. Consequently, there is currently significant interest in further development of drug conjugation technologies, with a particular focus on site-specific payload conjugation. Here, we present an enzymatic conjugation platform based on the S. aureus sortase A-mediated transpeptidation reaction, allowing the efficient generation of ADCs with toxins conjugated to pre-defined sites at pre-defined drug-to-antibody ratios. For this, two modifications were introduced: first, immunoglobulin heavy (IgH) and light (IgL) chains were modified at their C-termini by addition of the sortase A recognition motif LPETG, and second, the small molecule tubulin polymerization inhibitors monomethylauristatin E (MMAE) and maytansine were modified by addition of a pentaglycine peptide, thus making them suitable substrates for sortase A-mediated transpeptidation. We demonstrate efficient generation and characterization of the anti-CD30 ADC Ac10-vcPAB-MMAE, an enzymatically conjugated counterpart of brentuximab vedotin (Adcetris), as well as several anti-HER-2 ADCs including trastuzumab-maytansine, the counterpart of trastuzumab emtansine (Kadcyla). ADCs generated in this manner were found to display in vitro cell killing activities indistinguishable from the classic conjugates. Further, when tested in vivo in a HER-2-overexpressing ovarian cancer xenograft mouse model, enzymatically generated trastuzumab-maytansine was found to lead to complete regression of established tumors, similar to Kadcyla.

Ipsen joins ADC race via $900M biobuck deal with Sutro, chasing same target as Merck, Boehringer

STRO-003 will become the first antibody-drug conjugate in Ipsen’s collection, and the company will focus on getting the candidate into the clinic. Ipsen has become the newest entrant in the antibody-drug conjugate (ADC) race, handing over around $90 million in near-term payments to Sutro Biopharma in return for the global rights to a preclinical candidate. The asset in question, dubbed STRO-003, targets the receptor tyrosine kinase-like orphan receptor 1 (ROR1) tumor antigen. ROR1 is overexpressed in various cancers—including solid tumors and blood cancers—and the target is also being chased by the likes of Boehringer Ingelheim and Merck & Co. STRO-003 will become the first ADC in Ipsen’s armory, and the French company said the focus will be on getting the candidate into the clinic. The deal is worth around $900 million in combined upfront, development, regulatory and commercial milestone payments and an equity investment in Sutro. As a result, Ipsen will take on responsibility for preparing STRO-003 for phase 1 trials and beyond. “The potential for ADCs in oncology is well-documented and we are excited by the addition of STRO-003, Ipsen’s first ADC candidate with best-in-class potential,” Mary Jane Hinrichs, senior vice president and head of early development at Ipsen, said in an April 2 release. “STRO-003 is a next-generation ROR1 ADC, leveraging Sutro’s site-specific technology to generate a highly stable conjugate, coupled with exatecan payloads, that have shown significant potential in solid tumors,” Hinrichs added. Capitalizing on Ipsen's interest, Sutro announced plans this morning to bring in $75 million from an underwritten offering of 14.4 million shares of its common stock. Ipsen isn’t the only biopharma that’s spotted potential in ROR1. The target was at the center of Boehringer Ingelheim’s $1.5 billion acquisition of NBE-Therapeutics back in 2020, the same year that Merck bought VelosBio for $2.8 billion for a ROR1 ADC. In 2022, Astellas also bought into Sutro’s vision, handing over $90 million upfront to the South San Francisco-based biotech for the rights to three immunostimulatory ADCs, a modality designed to combine the best features of ADCs and personalized vaccine biology. The ADC race heated back up toward the end of last year, with everyone from GSK to Eli Lilly and Bristol Myers Squibb fronting serious cash to pad out their pipelines over the space of a few months.

Ipsen jumps on ADC bandwagon with deal for Sutro’s ROR1-targeting therapy

Ipsen added the first antibody-drug conjugate (ADC) to its pipeline through a deal with Sutro Biopharma potentially worth up to $900 million. Mary Jane Hinrichs, head of early development at Ipsen, suggested that Sutro’s STRO-003, which targets the ROR1 tumour antigen, has “best-in-class potential.”Ipsen will assume responsibility for Phase I preparation activities for STRO-003 as well as all subsequent clinical development and global marketing activities. In return, Sutro is eligible to receive around $90 million in near-term payments, including an equity investment, along with further milestone payments and tiered sales royalties.Hinrichs noted that STRO-003 utilises Sutro’s site-specific technology to generate a highly stable conjugate, coupled with exatecan payloads, that have shown significant potential in solid tumours. The ADC is in the final stages of preclinical development.Both Boehringer Ingelheim and Merck & Co. splashed out in 2020 to add ADCs targeting ROR1 to their pipelines. Merck’s asset, gained via the purchase of VelosBio, is currently in mid-stage development, but Boehringer, which added its therapy through the acquisition of NBE-Therapeutics, halted work on the drug last year.
