Covalent fragment-based drug discovery

Fragment-based drug discovery (FBDD) has delivered roughly fifty drugs to the clinic, five of which have already been approved. In contrast to traditional drug discovery, where millions of larger mols. are often screened, FBDD starts with libraries of just a few thousand very small mols., or fragments. This approach has multiple advantages over conventional screening methods including increased coverage of chem. space and the ability to target "difficult" proteins.<br/>Another relatively new phenomenon in drug discovery is targeted covalent drug discovery. Although covalent drugs such as aspirin and penicillin have been used for decades, only in recent years have researchers prospectively pursued covalent drugs. Most covalent drugs in development started with a high-affinity non-covalent binder onto which a "warhead" was grafted.<br/>Frontier Medicines is combining fragment-based and covalent drug discovery. Like putting a computer in a phone, we believe the combination of these two techniques will create new opportunities, particularly for previously intractable targets. Indeed, the only covalent fragment-based drug to have been approved targets a mutant form of KRAS - a protein that had previously been thought to be undruggable. Frontier is continuously building and expanding our covalent fragment library and applying this in a variety of assays, from isolated proteins to cell-based screens. This presentation will provide an overview of our approach and learnings.
