Effect of Nutritional Supplementation With Turmeric on the Cognitive Performance of Subjects With Metabolic Syndrome

The EPICURO study aims to demonstrate the beneficial effects of a 6-month dietary supplementation with an improved bioavailable turmeric (MERIVA®) on inflammatory, oxidative and metabolic parameters together with cognitive performance, potentially resulting in the reduction of the risk of cognitive decline in subjects, male and female, with Metabolic Syndrome. Evaluation of a Computational Model That Predicts the Effects of Quercetin on Metabolic Health Using Biological Age and Other Lipid-related Blood Metrics

Quercetin is a natural flavonoid compound widely found in vegetables, fruits, and nuts. It has a long history of use as a dietary supplement. This study aims to assess results from a computational model suggesting that quercetin may offer novel benefits to metabolic health. Participants will take quercetin (as Quercetin Phytosome, a proprietary formulation with enhanced absorption properties) for 90 days while keeping lifestyle habits consistent throughout to estimate net effects of quercetin as much as possible. Blood samples will be collected at the beginning and end of the study to assess the effects of quercetin supplementation on metabolic health metrics such as LDL cholesterol, blood sugar, liver, kidney and immune function and calculated metabolic age. In addition, questionnaires will be completed to ensure compliance with study requirements and assess potential quercetin benefits in the participants' quality of life. This work will provide a proof-of-concept from a computational model of nutraceutical compounds and proposes a new application of quercetin in support of healthy human metabolism.

The aim of the present investigation was to better understand the pharmacokinetic profile of bilberry (Vaccinium Myrtillus) anthocyanins and the role of glucose transporters (sGLT1 and GLUT2) on their absorption. In particular, the absorption of 15 different anthocyanins contained in a standardized bilberry extract (Mirtoselect^®) was measured in rats by a validated LC-ESI-MS/MS approach. The plasma concentration peak (Cmax) of 11.1ng/mL was reached after 30min and fasting condition significantly increased the bioavailability of anthocyanins by more than 7 fold in respect to fed rats. Glucose co-administration did not interfere with the overall anthocyanin uptake. Bioavailability of each anthocyanin was then estimated by comparing the relative content in plasma vs extract. The 15 anthocyanins behaved differently in term of bioavailability and both the aglycone and the sugar moiety were found to affect the absorption. For instance, arabinoside moiety was detrimental while cyanidin enhanced bioavailability. Computational studies permitted to rationalize such results, highlighting the role of glucose transporters (sGLT1 and GLUT2) in anthocyanins absorption. In particular a significant correlation was found for the 15 anthocyanins between sGLT1 and GLUT2 recognition and absorption.

At the HHS Black Health Forum 2024, held on 28 February, the secretary announced a project for gene therapies, beginning with work towards access to sickle cell disease treatments. “We now have, within our grasp, not just a treatment, but a cure for sickle cell disease,” added Becerra. However, he highlighted monetary barriers to treatment access. On 8 December, the US Food and Drug Administration (FDA) approved two gene therapies for the treatment of sickle cell disease in patients 12 and older. Vertex Pharmaceuticals and CRISPR Therapeutics ’ CRISPR/Cas-9 therapy, Casgevy (exagamglogene autotemcel), and Bluebird Bio’s cell-based gene therapy, Lyfgenia (lovotibeglogene autotemcel), became the first US-approved gene therapies for sickle cell disease. Becerra declared that the US Centers for Medicare and Medicaid Services (CMS) will soon begin an initiative to collaborate with as many states as possible to negotiate a deal with the pharmaceutical manufacturers of the two therapies. Headed by CMS administrator Chiquita Brooks La-Sure, the organisation plans to reach a deal on a reasonable price for gene therapies to increase access for patients. “If we do it together, we can bargain because we have the power of numbers behind us and we can negotiate the best deal,” says Becerra. Sickle cell disease is the name of a group of diseases caused by dysfunction in the red blood cells. Symptoms of the disease include anaemia, increased risk of infection and, in some cases, stroke and death. The US Centers for Disease Control and Prevention (CDC) reports that the disease affects approximately 100,000 Americans, disproportionately occurring in one in every 365 Black or African-American births. Becerra called the audience to action saying: “Make sure that your states are joining in this voluntary effort …to drive down the price and get the best bargain for those who have sickle cell disease and who are on Medicaid.” At the forum, the HHS secretary raised other examples of the department’s health equity actions, highlighting the extension of Medicaid postpartum coverage. HHS currently has a programme, in partnership with Congress, that allows it to partner with states to deliver 365 days of postpartum care to new mothers. Becerra says 44 states have joined the programme since it launched, but he encouraged the remaining states to join the initiative. Resultingly, batches of crofelemer API manufactured by Milan, Italy-based Indena S.P.A. can now be used by Jaguar for further development work. About Cancer Therapy-related Diarrhea A significant proportion of patients undergoing cancer therapy experience diarrhea, and diarrhea has the potential to cause dehydration, potential infections, and non-adherence to treatment in this population. Novel "targeted cancer therapy" agents, such as epidermal growth factor receptor (EGFR) antibodies and tyrosine kinase inhibitors (TKIs), with or without cycle chemotherapy agents, may cause increased electrolyte and fluid content in the gut lumen, which results in passage of loose/watery stools (i.e., diarrhea). Diarrhea has been reported as one of the most common side effects of TKIs and may result in cancer therapy drug holidays or reductions from therapeutic dose, potentially impacting patient outcome.
