Evaluation Value of Preoperative Serum Transaminase Ratio Combined with Serum Tumor Markers for Prognosis of Patients with Stage III Colorectal Cancer: A Retrospective Study.

OBJECTIVEThis study utilized data from two hospitals to investigate the relationship between the preoperative serum alanine aminotransferase to aspartate aminotransferase ratio (LSR) combined with preoperative serum tumor markers (TM) and the prognosis of patients with stage III colorectal cancer (CRC).METHODSAn analysis was performed on the clinical data of 211 patients diagnosed with stage III colorectal cancer (CRC), treated surgically at the Department of Gastrointestinal Surgery of the Second Affiliated Hospital of Nanchang University and the General Surgery Department of the First Affiliated Hospital of Nanchang University between January 2016 and December 2016. Laboratory data were also compiled for these patients. The main follow-up indicators were the five-year disease-free survival (DFS) and the five-year overall survival (OS). Survival analysis was carried out using the log-rank test, and Cox regression analysis was applied to ascertain independent prognostic factors for survival. Rank sum tests were utilized to compare categorical variables, with a p-value of less than 0.05 considered statistically significant.RESULTBetween January 2016 and December 2016, a total of 211 patients were enrolled in the two hospitals, including 113 males and 98 females. Multivariate analysis showed that the independent prognostic factors for five-year disease-free survival of stage III colorectal cancer were LSR (HR: 0.45,95% CI:0.31-0.67, p<0.001) and CEA (HR: 2.11,95%CI:1.42-3.13, p<0.001). Independent prognostic factors for five-year overall survival of stage III colorectal cancer were LSR (HR:0.48, 95%CI:0.31-0.75, p=0.001) and CA199 (HR:1.73, 95%CI:1.11-2.68, p=0.015). Kaplan-Meier survival analysis showed that LSR combined with TM could evaluate the five-year DFS and five-year OS of stage III colorectal cancer (DFS, p<0.001, OS: p=0.002).CONCLUSION1. The prognosis of patients with high preoperative LSR and low CEA, CA199 in perioperative period is better than that of other stage III colorectal cancer patients. 2. Perioperative preoperative serum transaminase ratio combined with CEA, CA199 has a certain reference value in the prognosis assessment of stage III colorectal cancer.

Lipolysis-stimulated lipoprotein receptor is involved in fatty acid binding protein 4-mediated prostate cancer cell growth in bone

Obesity-induced excess adipokine production is associated with malignancy and mortality in prostate cancer. We previously showed that fatty acid binding protein 4 (FABP4), a major adipokine of mature adipocytes, promotes the progression of prostate cancer cell growth and invasion. In this report, we present lipolysis-stimulated lipoprotein receptor (LSR) as a newly identified binding partner for FABP4. Their binding induced Akt phosphorylation, whereas LSR knockdown (KD) failed to phosphorylate Akt. Intraosseous injection of LSR-KD prostate cancer cells showed smaller areas of intraosseous tumor, lower Ki-67 labeling indices, and lower numbers of phospho-Akt-positive cancer cells compared with control prostate cancer cells. Moreover, the contact coculture of prostate cancer cells with bone marrow stromal cells (BMSCs) promoted FABP4 secretion by BMSCs. Our findings indicated that FABP4-mediated prostate cancer cell progression was regulated by cellular signaling via FABP4-LSR binding in the bone microenvironment. J. Med. The role of the tricellular junction protein ILDR2 in glomerulopathies: Expression patterns and functional insights

The tricellular tight junctions are crucial for the regulation of paracellular flux at tricellular junctions, where tricellulin (MARVELD2) and angulins (ILDR1, ILDR2, or LSR) are localized. The role of ILDR2 in podocytes, specialized epithelial cells in the kidney, is still unknown. We investigated the role of ILDR2 in glomeruli and its influence on blood filtration. Western blots, single-cell RNA sequencing (scRNA-seq), and superresolution microscopy showed a strong expression of ILDR2 in podocytes that colocalized with the podocyte-specific claudin CLDN5. Co-immunoprecipitation revealed that ILDR2 interacts with CLDN5. In glomerulopathies, induced by nephrotoxic serum and by desoxycorticosterone acetate (DOCA)-salt heminephrectomy, ILDR2 was strongly up-regulated. Furthermore, Ildr2 knockout mice exhibited glomerular hypertrophy and decreased podocyte density. However, they did not develop effacement of podocyte foot processes or proteinuria. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) proteomic analysis of isolated glomeruli showed an increase in matrix proteins, such as fibronectin and collagens. This suggests a protective role of ILDR2 in glomerulopathies.
