A tyramine receptor gene LsTAR2 is involved in reproduction and feeding in the small brown planthopper Laodelphax striatellus

Tyramine (TA) is an important biogenic amine present in the central nervous system of insects, and mediates a variety of physiological and behavioral functions via tyramine receptors (TARs). However, TARs have not yet been characterized in planthoppers, and their physiological functions remain poorly understood in rice pests. Here, we cloned a tyramine receptor gene (LsTAR2) from the small brown planthopper Laodelphax striatellus, one of the most destructive rice pests. LsTAR2 shares high sequence identity with its orthologous receptors, and is closely related to its corresponding receptor groups. LsTAR2 transcript was expressed mostly in the egg stage and brain. RNAi-mediated knockdown of LsTAR2 significantly prolonged the preoviposition period and decreased the fecundity in females. Furthermore, LsTAR2 knockdown reduced the expression levels of vitellogenin (LsVg) in the fat body and ovary of L. striatellus, and changed the expressions of juvenile hormone (JH) and 20-hydroxyecdysone (20E) pathway genes. In addition, LsTAR2 knockdown significantly decreased the honeydew excretion of the adults, and affected the transcript levels of feeding-related neuropeptide signaling genes. These results provide critical information concerning the role of LsTAR2 in reproduction and feeding behavior in L. striatellus, and open the way for further investigations into novel strategies targeting TARs for pest control.

Wall teichoic acid glycosylation of bovine-associated Staphylococcus aureus strains

Staphylococcus aureus (S. aureus) is one of the major causes of bovine mastitis, a disease with detrimental effects on health and wellbeing. Current control measures are costly, laborious and not always effective in eradicating S. aureus. The cell wall-linked polysaccharide wall teichoic acid (WTA) is highly immunogenic in humans and is considered as a prospective vaccine antigen based on promising pre-clinical studies in animals. WTA consist of polymerized ribitol-phosphate backbone that is modified with N-acetylglucosamine (GlcNAc) moieties in different configurations by the glycosyltransferases TarS (β-1,4-GlcNAc), TarM (α-1,4-GlcNAc) and TarP (β-1,3-GlcNAc). This study aimed to characterize the presence and genetic variation in tarS, tarM and tarP in bovine-associated S. aureus strains and how this impacts WTA-glycoprofile. Bioinformatic analyses of a whole genome sequence database consisting of 1047 S. aureus, 10 S. schweitzeri, and 6 S. argenteus strains showed that over 99% of strains contained tarS, 34 % also contained tarM, while 5 % of the strains encoded tarP in addition to tarS. The distribution of WTA-glycosyltransferase genes was similar to what has been reported for human-associated S. aureus strains. Phenotypic analysis of WTA glycosylation by flow cytometry corroborated with tarS/tarM/tarP gene presence. The WTA glycoprofile was variable between bovine-associated strains and the levels and ratios of GlcNAcylation were affected by growth conditions. Interestingly, a divergent tarM allele was present in strains of clonal complexes (CC) 49 and the mastitis-associated CC151, but its function was similar to canonical tarM. In conclusion, we demonstrated that bovine-associated S. aureus strains show similar variation in WTA GlcNAc decoration as human S. aureus strains, despite the presence of a divergent tarM allele.

A Mutation in the CACNA1F Gene Found by Whole Exome Sequencing (WES) and In Silico Analysis in an Iranian Family with Consanguineous Relationships

Background:X-linked mutations are highly important in clinical diagnosis, and at least 533 disorders are related to the genes located on the X chromosome.Case Presentation:A 21-year-old Caucasian woman with a 24-year-old Caucasian man as her fiancé referred Clinical genetic lab for premarital genetic counseling (carrier screening). None of them had any abnormal manifestations. Following genetic counseling, Whole Exome Sequencing (WES) test performed to find the possible pathogenic mutations. Also, after drawing the couple's pedigree, candidate mutations were examined in the woman's parents as well as her uncles. Additionally, in silico investigations were performed through SWISS-MODEL, MolProbity, ProSA, Py- Mol, and FATCAT tools. The most important mutation diagnosed in the woman (R1362Q in the 35th exon of CACNA1F), was observed in her mother and her two uncles. The mutation was also screened in both her father and her fiancé, but they had no mutations. After medical examinations of carriers, there was no sign of any eye impairment. Other mutations were TCTN2 (c.1613-2A>G), TARS (p.K319E), SPEG (p.E3020K), CPS1 (p.A1180V), MYO3A (p.I736M), NNT (p.R968Q), MED23 (p.K406T). Bioinformatics analyses indicated no alteration in the mutant structure of CACNA1F (Q1362) compared with the normal structure (R1362).Conclusion:Conclusively, the current study emphasizes the non-pathogenic effect of missense mutation R1362Q in the 35th exon of CACNA1F in association with ocular diseases. This will ensure the reports of this mutation as healthy instead of uncertain in the literature and databanks.
