Introduction

Electrogenic proton-coupled amino-acid transporter that transports oligopeptides of 2 to 4 amino acids with a preference for dipeptides. Transports neutral and monovalently charged peptides with a proton to peptide stoichiometry of 1:1 or 2:1 (By similarity) (PubMed:15521010, PubMed:18367661, PubMed:19685173, PubMed:26320580, PubMed:7896779, PubMed:8914574, PubMed:9835627). Primarily responsible for the absorption of dietary di- and tripeptides from the small intestinal lumen (By similarity). Mediates transepithelial transport of muramyl and N-formylated bacterial dipeptides contributing to recognition of pathogenic bacteria by the mucosal immune system (PubMed:15521010, PubMed:9835627).

Maternal peptidoglycan overexposure during late pregnancy alters neurodevelopment and behavior in juvenile offspring

Emerging evidence suggests that maternal gut microbiota-derived metabolites and components influence fetal brain development and subsequent neurodevelopment. This study investigates the effects of maternal overexposure to muramyl dipeptide (MDP)-a bacterial peptidoglycan (PGN) motif recognized by Nod2 receptors-on offspring neurodevelopment and behavior. Time-mated C57BL/6J female mice received MDP via drinking water from gestational days 16-19. Nod2 activation in amniotic fluid was assessed using a Nod2 cell-based reporter assay, showing a significant increase in males 24 h after MDP exposure. Gene expression analysis revealed upregulation of PGN transporters in fetal brains, with males showing higher levels of Slc15a1/PepT1, Slc15a2/PepT2, and Slc46a2. No changes in inflammatory or microglia-related markers were found. Behavioral assessments during the juvenile period revealed sex-specific effects: prenatally exposed males showed reduced social interaction, while females exhibited reduced novelty-induced locomotion and impaired social recognition. These behavioral changes were linked to altered expression of synaptic (Dlg4, Ppp1r9b, Darpp-32) and microglial (Trem-2, Cx3cr1) genes in the prefrontal cortex. Our findings underscore the sex-specific effects of maternal PGN overexposure on offspring neurodevelopment, highlighting the potential role of the maternal microbiome in the neurobiology of neurodevelopmental disorders, even in the absence of infection or robust inflammation.

Molecular mechanism of melatonin-mediated mitophagy regulating proline production to ameliorate skin aging

Collagen loss is one of the major contributor to signs of skin aging such as dryness, roughness, and wrinkle formation, which is closely linked to a decline in the amount of proline produced in mitochondria. Melatonin has been shown to improve several clinical signs of skin aging, while the mechanism is unclear. In our study, we found that mitophagy, proline synthesis key enzyme NADK2 and proline and collagen levels were significantly reduced, while oxidative stress levels increased in aging skin, and melatonin supplementation could effectively up-regulate mitophagy level and restore proline synthesis and further improved skin aging. However, proline supplementation could also exert an anti-aging effect, while it had no effect on the mitochondrial dysfunction. Moreover, our study indicated that melatonin enters the cell by binding to the MT1 receptor and then enters the mitochondria via the PEPT1 transporter to exert its mitochondrial protective effects. This study helps to elucidate the mechanism of mitochondrial dysfunction-induced skin aging, and provides new theoretical guidance for revealing the mechanism of skin aging and rationally utilizing endocrine hormones to improve skin aging, which has a broad application prospect.

Yellow mealworm (Tenebrio molitor) meal replacing dietary fishmeal alters the intestinal microbiota, anti-oxidation and immunity of large yellow croaker (Larimichthys crocea)

The present study investigated the impact of dietary fishmeal replacement with yellow mealworm (Tenebrio molitor) meal (TM) on the anti-oxidation, immunity and intestinal microbiota of large yellow croaker (Larimichthys crocea), with an initial body weight of 189.18 ± 0.13 g. Seven isonitrogenous and isolipidic diets were made using TM replacing the fishmeal at 0 % (TM0), 15 % (TM15), 30 % (TM30), 45 % (TM45), 60 % (TM60), 75 % (TM75) and 100 % (TM100), respectively. Each experimental diet was randomly assigned to three replicate groups of large yellow croaker (100 fish per group). After an 80-day feeding trial, it was showed that the activities of maltase, lysozyme and acid phosphatase in intestine, as well as serum albumin concentration and total anti-oxidative capacity in serum of fish fed diets with TM replacing fishmeal more than 60 % (P < 0.05). The intestinal muscularis thickness and perimeter-to-diameter ratio decreased significantly in TM75 and TM100 groups compared to the control (P < 0.05). At the transcriptional level, the mRNA expression of occludin, zonula occludens-1 (zo-1), oligopeptide transporter 1 (pept1), and oligopeptide transporter 2 (pept2) in the intestine, along with nuclear factor erythroid 2-related factor 2 (nrf2) and interleukin-10 (il-10) in both the intestine and liver, were linearly downregulated as the fishmeal replacement level increased (P < 0.05). While the relative expression levels of kelch-like ECH-associated protein 1 (keap-1), nuclear factor-κB (nf-κb) and tumor necrosis factor-α (tnf-α) in the intestine and liver were linearly upregulated with increasing dietary fishmeal replacement levels (P < 0.05). Besides, increasing dietary fishmeal replacement levels maintained intestinal microbiota alpha diversity (P > 0.05), while altering the intestinal microbial composition (P < 0.05). In conclusion, replacing 45 % of fishmeal with TM (equivalent to 25.57 % TM in diet) had no significant negative effects on the intestinal microflora, anti-oxidation and immunity of large yellow croaker.
