Anti-inflammatory and antioxidative effects of Perilla frutescens-derived extracellular vesicles: Insights from Zebrafish models

Plant-derived extracellular vesicles have recently been extracted and recognized as promising bioactive molecules, owing to their distinctive biological properties and inherent therapeutic activities. In this study, we investigated the physicochemical characteristics, bioactive properties, and therapeutic potential of Perilla frutescens-derived exosome-like nanoparticles (PELNs). Transmission electron microscopy (TEM) revealed that PELNs exhibited a cup-shaped morphology, with a lipid bilayer and a size distribution ranging from 40 to 200 nm (mean: 68.4 ± 13.0 nm). The cargoes in PELNs were analyzed through multi-omics and small RNA sequencing. In vivo studies on zebrafish demonstrated that PELNs are non-toxic at experimental concentrations. A reduction in neutrophil migration to injured fins evidenced the anti-inflammatory properties of PELNs. Furthermore, a meta-analysis of transcriptomic data identified hundreds of differentially expressed genes (DEGs) across 12 samples of three experimental groups. These DEGs were annotated into three categories following gene ontology (GO) enrichment analysis. Additionally, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis revealed that these DEGs were involved in immune-related pathways, including complement and coagulation cascades, systemic lupus erythematosus, PPAR signaling pathways, and antigen processing and presentation. Twelve selected DEGs were validated by quantitative real-time PCR (qRT-PCR), with particular confirmation of the mpx and lcp1 genes via in situ hybridization. Furthermore, PELNs demonstrated antioxidative effects by mitigating reactive oxygen species (ROS) levels, as evidenced by measurements of four oxidative stress (OS) indicators (i.e., SOD, CAT, GSH, and MDA) in zebrafish larvae subjected to H₂O₂-induced OS. In summary, PELNs exhibit substantial anti-inflammatory and antioxidant properties, underscoring their potential as therapeutic agents for treating various inflammatory diseases.

Microglia are essential for tissue contraction in wound closure after brain injury in zebrafish larvae

Wound closure after brain injury is crucial for tissue restoration but remains poorly understood at the tissue level. We investigated this process using in vivo observations of larval zebrafish brain injury. Our findings show that wound closure occurs within the first 24 h through global tissue contraction, as evidenced by live-imaging and drug inhibition studies. Microglia accumulate at the wound site before closure, and computational models suggest that their physical traction could drive this process. Depleting microglia genetically or pharmacologically impairs tissue repair. At the cellular level, live imaging reveals centripetal deformation of astrocytic processes contacted by migrating microglia. Laser severing of these contacts causes rapid retraction of microglial processes and slower retraction of astrocytic processes, indicating tension. Disrupting the lcp1 gene, which encodes the F-actin–stabilising protein L-plastin, in microglia results in failed wound closure. These findings support a mechanical role of microglia in wound contraction and suggest that targeting microglial mechanics could offer new strategies for treating traumatic brain injury.

Expanding frontiers in liquid biopsy-discovery and validation of circulating biomarkers in renal cell carcinoma and bladder cancer

Renal cell carcinoma (RCC) and Bladder cancer (BC) are two lethal urological cancers that require diagnosis at their earliest stage causing decreasing survival rates in case of aggressive disease. However, there is no reliable circulating marker in blood or urine for their less or non-invasive diagnosis. Our objective was to review the potential circulating biomarkers, namely proteins, micro-RNA (miRNA), long non-coding RNA (lncRNA), and circulating tumour cells (CTCs) for which we performed a PubMed-based literature search of biomolecules (protein, miRNA, lncRNA and CTCs) found as circulating biomarkers in blood and urine for the early detection of RCC and BC. Among the numerous studies, certain biomolecules represent promising early-stage biomarkers such as proteins (NNMT, LCP1, and NM23A; KIM1), mi-RNAs (5-panel: miR-193a-3p, miR-362, miR-572, miR-378, and miR-28-5p; miR-200a) and lncRNAs (5-panel: LET, PVT1, PANDAR, PTENP1 and linc00963; GIHCG) for RCC. Similarly, proteins (APOA1), miRNAs (7-panel: miR-7-5p, miR-22-3p, miR-29a-3p, miR-126-5p, miR- 200a-3p, miR-375, and miR-423-5p; miRNA 181a, miRNA 30c, and miRNA 570) and lncRNAs (3-panel: MALAT1, MEG3, and SNHG16; exosomal derived 3-panel: PCAT-1, UBC1 and SNHG16; H19) were reported in BC subjects. Notably, the majority of the biomarkers presented for early detection in RCC cases were found in blood, while in urine for BC. Our results reveal that though a plethora of circulating biomarkers show early diagnostic ability, all of them are still bench-only biomarkers and require further validation. Adequate clinical trials/studies testing which of these potential markers individually or in combination, will become clinically applicable still remain elusive.
