The identification of synthetic drug laboratories based on the detection of precursors

The Netherlands is a small country but large in terms of production and trade in synthetic drugs, especially amphetamines and MDMA. In fact, Dutch criminals are global leaders, with networks and business practices all over the world. It's a billion dollar business, that forms a shadow economy with great appeal. Production of these drugs takes place at remote locations, in barns connected to farmhouses but also in residential areas where production locations are hidden in cellars and attics. In this study we developed a method that may be helpful to detect such illegal synthetic drug laboratories in suspected areas. An SPME sampler is used to passively collect air samples which are then analyzed for benzyl methyl ketone (BMK) and piperonyl methyl ketone (PMK), the precursors of respectively amphetamines and MDMA. The detection limit was 0.01 ng absolute for both compounds which means that BMK and PMK can be detected at concentrations comparable to other airborne VOCs with a sampling time of less than 10 min. Environmental conditions have only a limited influence on the uptake of BMK and PMK. In measurements in and around a simulated synthetic drug laboratory BMK and PMK could easily be detected inside the facility and even outside at distances up to 5 m. In a training laboratory facility of the police, BMK as well as PMK could be detected even while these compounds were no longer present in this facility. The identification of BMK and PMK was selective which was confirmed by measurements at 40 unsuspected locations where BMK and PMK were not detected in the presence of other compounds. The method can help to confirm the identification of illegal synthetic drug laboratories or a storage facility in suspected locations. While analysis in this study was done in the laboratory, analysis in practice may be possible using a mobile GC/MS system.

Endophytic Bacteria Enterobacter cloacae PN7 Promotes Biosynthesis and Accumulation of Saponins in Panax notoginseng

Panax notoginseng is an important Chinese medicinal plant. Saponins are the major bioactive secondary metabolites with a wide range of medicinal and commercial value in P. notoginseng, so it is crucial to develop environmentally friendly methods to increase their production. The symbiotic relationship between endophytic bacteria and host plants offers a sustainable approach to enhance secondary metabolite biosynthesis. In this study, it was reported that the co-cultivation of an endophytic bacterium Enterobacter cloacae PN7, isolated from P. notoginseng and its host plant could greatly promote saponin accumulation in the root of seedlings. After six days of PN7 treatment, the total saponin concentration reached 21.64 mg/g, representing a 2.01-fold increase over the control. Transcriptome sequencing revealed that PN7 induction upregulated key genes in the saponin biosynthetic pathway (including DXS, HMGR, PMK, DS, CYP450, and GTs), modulated 253 plant hormone signaling genes (such as those related to JA, ETH, and ABA), and affected 284 transcription factor genes and 47 ABC transporter genes. Co-expression network analysis identified DEGs related to plant hormone signaling, transcription factors, and ABC transporters in saponin biosynthesis and distribution. The results suggested that JA signaling, mediated by transcription factors, such as bHLH and MYBs, and its interaction with ETH, played crucial roles in saponin biosynthesis. Additionally, potential ABC transporter candidates involved in saponin transport were identified. This study highlights the role of endophytic bacteria in enhancing saponin production in P. notoginseng and opens avenues for further research on microbial-plant interactions in secondary metabolite production.

Novel Drug Targets for Atrial Fibrillation Identified Through Mendelian Randomization Analysis of the Blood Proteome

PURPOSENovel, effective, and safe preventive therapy targets for AF are still needed. Circulating proteins with causal genetic evidence are promising candidates. We aimed to systematically screen circulating proteins for AF drug targets and determine their safety and efficacy using genetic methods.METHODSThe protein quantitative trait loci (pQTL) of up to 1949 circulating proteins were retrieved from nine large genome-proteome-wide association studies. Two-sample Mendelian Randomization (MR) and colocalization analyses were used to estimate the causal effects of proteins on the risk of AF. Furthermore, phenome-wide MR was conducted to depict side effects and the drug-target databases were searched for drug validation and repurposing.RESULTSSystematic MR screen identified 30 proteins as promising AF drug targets. Genetically predicted 12 proteins increased AF risk (TES, CFL2, MTHFD1, RAB1A, DUSP13, SRL, ANXA4, NEO1, FKBP7, SPON1, LPA, MANBA); 18 proteins decreased AF risk (PMVK, UBE2F, SYT11, CHMP3, PFKM, FBP1, TNFSF12, CTSZ, QSOX2, ALAD, EFEMP1, FLRT2, LRIG1, OLA1, SH3BGRL3, IL6R, B3GNT8, FCGR2A). DUSP13 and TNFSF12 possess strong colocalization evidence. For the proteins that were identified, extended phe-MR analysis was conducted to assess their side-effect profiles, while drug-target databases provided information on their approved or investigated indications.CONCLUSIONWe identified 30 circulating proteins as potential preventive targets for AF.
