Cationic AIEgens with large rigid π-planes: Specific bacterial imaging and treatment of drug-resistant bacterial infections

In this study, four D-π-A type cationic photosensitisers with aggregation-induced emission (AIE) properties were developed based on the electron-donating group triphenylamine and pyrene molecules acting as auxiliary electron donors and main π-bridges, as well as pyridinium salts of different charge numbers acting as electron acceptors: TPP1, MeOTPP1, TPP2 and MeOTPP2. The introduction of pyrene endowed the AIE photosensitizers with a high solid fluorescence quantum yield and long fluorescence lifetime. All four photosensitizer molecules were able to efficiently generate type I (·OH) and type II (¹O₂) under white light irradiation, achieving efficient inactivation of methicillin-resistant Staphylococcus aureus (MRSA) at low concentrations, and TPP1 and TPP2 successfully promoted wound healing in MRSA-infected mice. The introduction of a methoxy group effectively enhanced the intramolecular charge transfer effect, achieved longer wavelength absorption and fluorescence emission redshift, and effectively reduced ΔE_st thereby promoting ROS (Reactive Oxygen Species) generation. However, after the introduction of the methoxy group, the CAC (Critical Aggregate Concentration) of MeOTPP1 and MeOTPP2 became smaller and the hydrophobicity was enhanced, which affected the interaction with bacteria. In fact, the photodynamic antimicrobial activity and imaging ability against bacteria were reduced. TPP2 achieves efficient killing of MRSA and MDR E.coli (Multidrug-resistant Escherichia coli) by disrupting the bacterial cell membrane due to its high photosensitization efficiency, two positive charges and very high CAC value. Under light (40 mW·cm^-2), only 1 μM of TPP2 inactivated 87 % of MRSA, followed by TPP1, which inactivated 59 %, while MeOTPP1 and MeOTPP2 showed no significant antibacterial activity at this concentration. At a concentration of 10 μM, TPP2 deactivated more than 95 % of MDR E.coli, TPP1 deactivated about 41 %, and MeOTPP1 and MeOTPP2 had no antimicrobial activity against MDR E.coli at this concentration. In addition, TPP1, MeOTPP1 and TPP2 were able to rapidly identify MRSA and MDR E.coli under the irradiation of 365 nm UV light, which provides a visual method for the rapid identification of MRSA and MDR E.coli.

Proximate composition, peptide characterization and bioactive properties of faba bean blanching water

Faba bean (Vicia faba L.) offers a rich nutritional profile with high protein content and abundant vitamins and minerals. Processing of faba beans for freezing requires blanching, yielding liluva (legume processing water), possibly containing leached macronutrients, with potential for upcycling. Past evidence has shown that legume processing water may be high in protein. Peptides generated from faba bean proteins during processing have been shown to have bioactivity and can, for example, inhibit HMG-CoA reductase. HMG-CoA reductase is rate-limiting in the biosynthesis of cholesterols and high cholesterol increases the risk of cardiovascular diseases. Thus, this study examined the composition of legume blanching water, analysed the peptides resulting from in vitro digestion and assayed HMG-CoA reductase inhibitory activity of liluva from faba beans sourced from two farms in Canterbury, New Zealand. Results showed that the blanching water contained around 1.7 g/100 mL solid content. These solids were approximately 30 % protein, 12 % water-soluble carbohydrates, 4 % dietary fibre, and 0.17 % ash. Mineral analysis showed high levels of potassium in macro minerals and zinc in trace minerals. Free amino acid analysis revealed high levels of arginine, alanine, asparagine, and glutamic acid, and low levels of methionine and tryptophan. Mass spectrometry analysis identified 111 and 72 endogenous peptides in farm 1 and 2 raw samples, respectively. Most of these peptides were derived from tripeptidyl-peptidase II and subtilisin-like protease. To understand potential bioactivity of these peptides, peptides were also analyzed after in vitro digestion. The resulting identified peptides indicated in silico predicted bioactivities such as anti-thrombotic, antioxidative, ACE dipeptidyl peptidase -III and -IV inhibition and HMG-CoA reductase inhibition activities. Further validation of the faba bean blanching water after in vitro digestion demonstrated approximately 67 % inhibition of HMG-CoA reductase activity, suggesting potential hypocholesterolemic properties. These findings suggest that faba bean blanching water may serve as a sustainable and functional ingredient with potential cholesterol-lowering effects in food production.

AbstractHuman Inborn Errors of Immunity (IEIs) encompass a clinically and genetically heterogeneous group of disorders, ranging from mild cases to severe, life-threatening types. Among these, Primary Immune Regulatory Disorders (PIRDs) constitute a subset of IEIs characterized by diverse clinical phenotypes, prominently featuring severe atopy, autoimmunity, lymphoproliferation, hyperinflammation, autoinflammation, and susceptibility to malignancies. According to the latest report from the International Union of Immunological Societies (IUIS), PIRDs arise from mutations in various genes including LYST, RAB27A, AP3B1, AP3D1, PRF1, UNC13D, STX11, STXBP2, FAAP24, SLC7A7, RASGRP1, CD70, CTPS1, RLTPR, ITK, MAGT1, PRKCD, TNFRSF9, SH2DIA, XIAP, CD27 (TNFRSF7), FAS (TNFRSF6), FASLG (TNFSF6), CASP10, CASP8, FADD, LRBA, STAT3, AIRE, ITCH, ZAP70, TPP2, JAK1, PEPD, FOXP3, IL2RA, CTLA4, BACH2, IL2RB, DEF6, FERMT1, IL10, IL10RA, IL10RB, NFAT5, TGFB1, and RIPK1 genes. We designed a targeted next-generation sequencing (TNGS) workflow using the Ion AmpliSeq™ Primary Immune Deficiency Research Panel to sequence 264 genes associated with IEIs on the Ion S5™ Sequencer. In this study, we report the identification of 38 disease-causing variants, including 16 novel ones, detected in 40 patients across 15 distinct PIRD genes. The application of next-generation sequencing enabled rapid and precise diagnosis of patients with PIRDs.
