Introduction

Validation of reference genes for cardiac RT-qPCR studies spanning the fetal to adult period

Many genes used as internal controls for mRNA expression studies are unstable (change) over development. This study determined an approach to validate reference genes for mRNA studies spanning the fetal period to adulthood in sheep hearts.•We determined the mRNA expression of 12 candidate reference genes (ACTB, GAPDH, H3-3A, HYAL2, PPIA, RNA18S1, RPL32, RPL37A, RPL41, RPLP0, RPS15, and YWHAZ) via RT-qPCR. Per RefFinder, which incorporates computational algorithms by BestKeeper, comparative delta Ct, GeNorm, and NormFinder, RPL32, RPL37A, HYAL2, ACTB and GAPDH were the most stable reference genes, although none were unchanged across all ages.•Systematical calculation of the geometric means of 3 reference genes revealed the combination of HYAL2, RPL32, and RPL37A was unchanged across the 5 fetal, neonatal, and adult ages.•We determined the most stable combination of reference genes for cardiac gene expression studies in sheep from fetus to newborn to adult; these steps are applicable to determine internal controls for mRNA studies in other organs, other species, and periods in which reference gene instability is high.

Injury From Nematode Lung Migration Induces an IL‐13‐Dependent Hyaluronan Matrix

ABSTRACTA consistent feature of lung injury is a rapid and sustained accumulation of hyaluronan (HA). The rodent gut‐dwelling nematode Nippostrongylus brasiliensis (Nb) induces tissue damage as it migrates through the lungs. Type 2 immune responses are essential for the repair of the lungs, hence Nb infection is a well‐established model to study immune‐mediated lung repair. We found that Nb infection was associated with increased HA in the lung, which peaked at d7 post‐infection (p.i.). Deposition of HA in the alveolar epithelium correlated with regions of damaged tissue and the type 2 immune response, which is characterized by eosinophilia and increased type 2 cytokines such as IL‐13. Consistent with the accumulation of HA, we observed increased expression of the major synthase Has2, alongside decreased expression of Hyal1, Hyal2, and Tmem2, which can degrade existing HA. Expression of Tsg6 was also increased and correlated with the presence of inter‐α‐inhibitor heavy chain–HA complexes (HC·HA) at d7 p.i. Using IL‐13‐deficient mice, we found that the accumulation of HA during Nb infection was IL‐13 dependent. Our data thus provide further evidence that IL‐13 is a modulator of the HA matrix during lung challenge and links IL‐13‐mediated HA regulation to tissue repair pathways.

Blood-based HYAL2 methylation as a potential marker for the preclinical detection of coronary heart disease and stroke.

BACKGROUNDCoronary heart disease (CHD) and stroke have become the leading cause of premature mortality and morbidity worldwide. Therefore, sensitive and accurate biomarkers for early detection of CHD and stroke are urgently needed for effective prevention and treatment. We aim to investigate the association between blood-based HYAL2 methylation and the risk of CHD and stroke in Chinese population.METHODSIn a prospective nested case-control study comprising 171 CHD cases, 139 stroke cases, who developed the diseases after recruitment and 356 controls who remained healthy during the 2.5 years of follow-up time, the methylation level of HYAL2 in the peripheral blood was quantified using mass spectrometry, and the association was calculated by logistic regression adjusted for covariant.RESULTSSignificant association between HYAL2 methylation in the peripheral blood and increased risk of preclinical CHD and stroke were identified [odds ratios (ORs) per - 10% methylation: 1.35-1.64, p ≤ 0.045 for HYAL2_CpG_1, HYAL2_CpG_2 and HYAL2_CpG_3 in CHD; ORs per - 10% methylation: 0.76-1.64, p ≤ 0.033 for HYAL2_CpG_2 and HYAL2_CpG_4 in stroke]. The association in CHD was further enhanced by female gender, younger age (< 70 years old), without the history of hypertension and cancer. The combination of four HYAL2 methylation sites showed an effective discrimination of CHD and stroke cases without hypertension from controls [area under curve (AUC) = 0.78 and 0.75, respectively].CONCLUSIONSThis study presents a strong association of altered HYAL2 methylation in peripheral blood with preclinical CHD and stroke, providing a novel biomarker for risk assessment and early detection of cardiovascular diseases.
