Insilico, a clinical-stage generative artificial intelligence (AI)-drive drug discovery company, says recent research demonstrates that protein phase separation (PPS) is widely present in cells and drives a range of important biological functions. PPS at the wrong place or time, it says, could create clogs or aggregates of molecules linked to neurodegenerative diseases, and poorly formed cellular condensates could contribute to cancers and may explain the ageing process. An emerging link between human disease and the PPS process has led scientists to look for ways to identify potential targets for therapeutic interventions based on PPS regulation. The university and Insilico have jointly published a paper in Proceedings of the National Academy of Sciences (PNAS) ​, presenting an approach to identify therapeutic targets for human diseases associated with PPS. It recognizes a significant research milestone in their collaboration launched in September 2021, they say. Protein separation prediction In this study, researchers combined Insilico's proprietary artificial intelligence (AI)-driven target identification engine PandaOmics with the FuzDrop method for predicting protein separation to identify PPS-prone disease-associated proteins. PandaOmics integrates multiple omics and text-based AI bioinformatics models to assess the potential of proteins as therapeutic targets. Large scale multi-omic study ​ Using this approach, the researchers conducted a large-scale multi-omic study of human sample data, quantified the relative impact of PPS in regulating various pathological processes associated with human disease, prioritized candidates with high PandaOmics and FuzDrop scores, and generated a list of possible therapeutic targets for human diseases linked with PPS. Researchers validated the differential phase separation behaviors of three predicted Alzheimer’s disease targets (MARCKS, CAMKK2 and p62) in two cell models of Alzheimer’s disease, which provides experimental validation for the involvement of these predicted targets in Alzheimer's disease and support their potential as therapeutic targets. In RSV, we were pleased to report positive Phase 1 results for EDP-323 in healthy volunteers, demonstrating favorable safety, tolerability, and pharmacokinetics. Research and development expenses totaled $43.0 million for the three months ended June 30, 2023, compared to $39.1 million for the three months ended June 30, 2022. The increase was primarily due to the timing of clinical trial expenses in the company’s virology programs. General and administrative expenses totaled $12.6 million for the three months ended June 30, 2023, compared to $12.9 million for the three months ended June 30, 2022. Enanta recorded income tax expense of $4.2 million for the three months ended June 30, 2023 driven by the receipt of the $200.0 million from the royalty sale agreement which is taxable for federal and state purposes. COVID-19 (SARS-CoV-2) Enanta announced additional analyses of SPRINT (SARS-Cov-2 PRotease INhibitor Treatment), its Phase 2 clinical trial of EDP-235, an oral, 3CL protease inhibitor in non-hospitalized, symptomatic adults with mild or moderate COVID-19. These analyses demonstrated a virologic effect of EDP-235 in the subset of patients who had not been recently infected, known as nucleocapsid-negative patients, as measured by lack of antibodies to the SARS-CoV-2 nucleocapsid. Specifically, in this nucleocapsid-negative patient subset, an 0.8 log viral load decline was observed at Day 5 with 400 mg of EDP-235 compared to placebo, and a 1.0 log viral load decline in nucleocapsid-negative patients who were treated within three days of symptom onset. These data build upon the positive topline SPRINT data announced in May in which a dose-dependent improvement in symptoms was observed with EDP-235 treatment compared to placebo, which achieved statistical significance (p<0.05) in the 400 mg treatment group at multiple time points, starting as early as one day after the first dose. In a prespecified population consisting of patients enrolled within three days of symptom onset, a statistically significant improvement was observed with EDP-235 at 400 mg at all time points. Further, an analysis of a subset of symptoms showed a two day shorter time to symptom improvement in patients receiving EDP-235 400 mg who were enrolled within three days of symptom onset (p<0.01). The study also demonstrated that EDP-235 was generally safe and well-tolerated. Enanta plans to conduct all future COVID work in the context of a collaboration. In a prespecified population consisting of patients enrolled within 3 days of symptom onset, a statistically significant improvement was observed with EDP-235 at 400mg at all time points. While no difference was observed in time to improvement of 14 targeted COVID-19 symptoms, an analysis of a subset of these symptoms showed a 2-day shorter time to improvement in patients receiving EDP-235 400mg who were enrolled within 3 days of symptom onset (p<0.01). No effect on virologic endpoints as measured in the nose was detected due to the rapid viral decline in the placebo arm of this highly immunologically-experienced, standard risk population. Enanta will continue to evaluate data from SPRINT and is focusing on partnership opportunities for Phase 3 and on the potential for a different Phase 2 study in acute or long COVID that could further demonstrate the efficacy of EDP-235. Corporate In April, Enanta strengthened its balance sheet with the sale to OMERS of 54.5% of Enanta’s future royalty payments from AbbVie Inc. on worldwide sales of MAVYRET/MAVIRET. The upfront purchase price paid to Enanta was $200 million. OMERS right to receive royalty payments is based on net sales of the product beginning in July 2023 through June 2032, with total payments capped at 1.42 times the purchase price. Enanta’s retains 45.5% of all royalties until the cap is hit, at which point 100% of all further royalties revert to Enanta.
