Evaluation of hypoxia pathway genes and serum parameters in new coronavirus pneumonia (COVID-19)

BACKGROUNDCoronavirus disease-2019 (COVID-19) causes severe hypoxemia. Unlike normal pneumonia, pneumonia due to COVID-19 causes oxygen deprivation without breathing difficulties (i.e., silent hypoxia). We evaluated the relationship between COVID-19 and hypoxemia and examined possible mechanisms of pneumonia from the perspective of gene expression (HIF1A, vascular endothelial growth factor [VEGF], NF-kB, MEKK1, and EGFR) using real-time PCR and ELISA for serum parameters.METHODSWe evaluated 100 individuals (50 patients and 50 controls). The patients were individuals with respiratory symptoms and pneumonia who were COVİD-19 positive. The relative quantification of standardized samples wa s calculated according to the formula 2 ^-ΔΔCT. Receiver operating curve (ROC) analysis was made to define the diagnostic power of the genes. The expression changes of four genes in the hypoxia pathway were significant (excluding VEGF) and upregulated in the patients' serums.RESULTSThe fold change values of the HIF1A, VEGF, NF-kB, MEKK1, and EGFR genes were 0.048, 0.688, 0.168, 0.207, and 0.171, respectively, in the cases checked against to the controls. The areas under the ROC values indicating the diagnostic power of the genes were 0.727, 0.538, 0.815, 0.734, and 0.936, respectively. Some serum parameters were significant (age, PCR, urea, LDH, WBC, ferritin, and pO₂).CONCLUSIONSThe upregulation of some genes in the hypoxia pathway in COVID-19 pneumonia shows that these genes and protein products are candidates for treatment targets. At the same time, the high discriminative power of two genes (NF-κB and EGFR) in patients compared to controls indicates their diagnostic potential in serum samples.

Integrative and comparative analysis of whole-transcriptome sequencing in circCOL1A1-knockdown and circCOL1A1-overexpressing goat hair follicle stem cells

Objective: Hair morphogenesis is tightly related to hair follicle stem cells (HFSCs) proliferation and hair follicle (HF) development. Yangtze River Delta white goats (YRDWG) HFSCs are important for producing superior-quality brush hair (SQBH). Nonetheless, the known regulatory mechanisms are not sufficient to explain YRDWG gHFSCs growth, HF development, and SQBH formation.Methods: To deeply investigate the interaction networks and mechanisms of circCOL1A1 in the HF development and SQBH formation of YRDWG in detail, we applied whole-transcriptome sequencing and bioinformatics analysis of circCOL1A1-knockdown and circCOL1A1-overexpressing HFSCs from YRDWG. STRING and other databases were used to construct multiple interaction networks. Differentially expressed (DE) genes, DE-miRNAs, and DE-circRNAs were further confirmed via real-time quantitative polymerase chain reaction and Sanger sequencing.Results: A total of 87 genes, 96 miRNAs, and 135 circRNAs were DE between circCOL1A1-knockdown and circCOL1A1-overexpressing gHFSCs. Functional enrichment, gene ontology annotation and Kyoto encyclopedia of genes and genomes analyses identified marked enrichment of these DE- genes, DE-miRNAs, and DE-circRNAs in the MAPK, PI3K/Akt, and focal adhesion signaling pathways, which are closely associated with gHFSCs growth and HF development. In addition, through interaction network construction, four important regulatory axes were obtained, namely, the chi-circCOL1A1-miR-149-5p-CMTM3-AR, chi-circACTN1- miR-671-5p-MAPK3/COL13A1, chi-circITGA6-miR-18a-5p-FGF1/MAP3K1 and chi-circ COBLL1-miR-30a-5p/miR-128-3p-ITGA6/MAPK14/FGF14 axes.Conclusion: These novel findings provide a valuable and comprehensive basis for investigating the complex mechanism by which circRNAs participate in and regulate HF development and SQBH formation in YRDWG.

KMT2C Polymorphism in Familial Hypospadias

Hypospadias, a common congenital anomaly of male genitalia, shows significant heritability and familial recurrence, particularly in consanguineous families. This study explored the role of KMT2C polymorphisms in a Yemeni family with two affected siblings. Comprehensive analysis identified 475 unique SNPs in KMT2C, with 59 shared between parents, suggesting common ancestry. Key interactions with genes such as MAP3K1, ATRX, and CHD7 implicated in sex differentiation were noted. Novel findings included a deleterious BAHD1 mutation and a de novo rs201834857 variant linked to Kleefstra syndrome 2. The study emphasizes the multifactorial etiology of hypospadias, integrating genetic, epigenetic, and environmental factors, and highlights the need for further research.
