Introduction

Acts as a hormone-regulated adipokine/pro-inflammatory cytokine that is implicated in causing chronic inflammation, affecting cellular expansion and blunting insulin response in adipocytes. May have a role in the onset of insulin resistance. Associates with immunoglobulin M (IgM) heavy and light chains and promotes IgM assembly and secretion. May exert its effect by acting as a molecular chaperone or as an oxidoreductase as it displays a low level of oxidoreductase activity (By similarity). Isoform 2 may be involved in regulation of apoptosis. Helps to diversify peripheral B-cell functions by regulating Ca(2+) stores, antibody secretion and integrin activation.

Integrated metabolomics and transcriptomics to reveal the anti-tumor mechanisms of Sanghuangporus mongolicus ethyl acetate extract in H22 tumor-bearing mice

This study aimed to systematically evaluate the anti-tumor efficacy of petroleum ether, ethyl acetate (EAE), and water extracts from Sanghuangporus mongolicus and decipher the molecular mechanisms of the most efficacious extract - EAE. Using H22 tumor-bearing mice, high-dose EAE (37.72 mg/kg) exhibited the highest tumor inhibition rate (75.14 %) without toxicity. Histopathology examination demonstrated that EAE effectively mitigated tumor progression and multi-organ damage, whereas ELISA and serum biochemical assays indicated modulated levels of immune mediators (IL-6, IL-2, IFN-γ, TNF-α, and VEGF) and restored serum levels of hepatic (ALT, AST) and renal (BUN, Cr, UA) functional markers. Integrated transcriptomics and metabolomics demonstrated that EAE suppressed tumor growth via multi-target regulation involving immune responses, biosynthesis of amino acids, and mitochondrial apoptosis pathways. Western blotting validated EAE upregulated pro-apoptotic cleaved caspase-3, caspase-3, Bax, and TNF-α (p < 0.01 vs. model group (MG)), upregulated immune-related protein JCHAIN, MZB1 (p < 0.01 vs. MG), downregulated anti-apoptotic Bcl-2 (p < 0.01 vs. MG). LC-MS identified 33 EAE compounds, with 32 showing strong binding (ΔG ≤ -5.0 kcal/mol) to core targets, JCHAIN and MZB1, via molecular docking. Phellibaumin C, inoscavin A, and phelligridin D exhibited the highest binding affinities. This study provides a multi-target mechanistic framework for developing S. mongolicus EAE as a natural anti-hepatocellular carcinoma (HCC) agent.

Investigation into the molecular mechanisms of biomarkers in psoriasis vulgaris with reduced high-density lipoprotein levels based on bulk transcriptome sequencing data

It has been found that severe lipid metabolism disorders are often present in patients with Psoriasis, including decreased levels of high-density lipoprotein (HDL). This study initially explored the impact of HDL level variations on psoriasis by collecting. This study collected 12 blood samples and 9 skin samples from psoriasis vulgaris and psoriasis vulgaris with reduced HDL levels and performed bulk RNA sequencing. The genes expressed explicitly in both tissue and blood samples from psoriasis vulgaris patients with low HDL levels were selected to explore their molecular regulation in psoriasis vulgaris further, to elucidate the pathogenesis of psoriasis. A total of 421 specific DEGs in blood and 143 specific DEGs in skin from PN groups were obtained, and these genes were enriched in the terms and pathways related to inflammation and immune system. Also, biomarkers were screened out with same expression pattern in both blood and skin samples. Five intersecting differential genes (METRNL, NDEL1, HLA-DRA, MZB1, MKRN3) were obtained. Their function was further predicted. In conclusion, our research identified five biomarkers in psoriasis that are associated with low HDL levels. Furthermore, our findings revealed that alterations in HDL levels in psoriasis may exacerbate the clinical manifestations of psoriasis through regulation of immune response and lipid metabolism.

Ribosome profiling and single-cell RNA sequencing identify the unfolded protein response as a key regulator of pigeon lactation

Pigeons and certain other avian species produce a milk-like secretion in their crop sacs to nourish offspring, yet the detailed processes involved are not fully elucidated. This study investigated the crop sacs of 225-day-old unpaired non-lactating male pigeons (MN) and males initiating lactation on the first day after incubation (ML). Using RNA sequencing, ribosome profiling, and single-cell transcriptome sequencing (scRNA-seq), we identified a significant up-regulation of genes associated with ribosome assembly and protein synthesis in ML compared to MN. Results from scRNA-seq analysis identified 12 distinct cell types and 22 clusters, with secretory epithelial cells (SECs) exhibiting marked expression of plasma cell markers, including IGLL1 and MZB1. RNA fluorescence in situ hybridization (RNA FISH) and IgY quantification confirmed the critical role of SECs in producing endogenous IgY during lactation. We propose that fibroblast-derived BAFF signals activate SECs, mimicking B cell transformation and enhancing protein production through the unfolded protein response (UPR). These findings shed light on the cellular dynamics of pigeon milk production and contribute to a broader understanding of avian biology.
