Nanozyme-Enhanced Probiotic Spores Regulate the Intestinal Microenvironment for Targeted Acute Gastroenteritis Therapy

Antibiotic therapeutics to combat intestinal pathogen infections often exacerbate microbiota dysbiosis and impair mucosal barrier functions. Probiotics are promising strategies, because they inhibit pathogen colonization and improve intestinal microbiota imbalance. Nevertheless, their limited targeting ability and susceptibility to oxidative stress have hindered their therapeutic potential. To tackle these challenges, Ces₃ is synthesized by in situ growth of CeO₂ nanozymes with positive charges on probiotic spores, facilitating electrostatic interactions with negatively charged pathogens and possessing a high reactive oxygen species (ROS) scavenging activity. Importantly, Ces₃ can resist the harsh environment of the gastrointestinal tract. In mice with S. Typhimurium-infected acute gastroenteritis, Ces₃ shows potent anti-S. Typhimurium activity, thereby alleviating the dissemination of S. Typhimurium into other organs. Additionally, owing to its O₂ deprivation capacity, Ces₃ promotes the proliferation of anaerobic probiotics, reshaping a healthy intestinal microbiota. This work demonstrates the promise of combining antibacterial, anti-inflammatory, and O₂ content regulation properties for acute gastroenteritis therapy.

A potential novel biomarker: comprehensive analysis of prognostic value and immune implication of CES3 in colonic adenocarcinoma.

PURPOSEColon cancer is the most common malignant tumor in the intestine. Abnormal Carboxylesterases 3 (CES3) expression had been reported to be correlated to multiple tumor progression. However, the association among CES3 expression and prognostic value and immune effects in colonic adenocarcinoma (COAD) were unclear.PATIENTS AND METHODSThe transcription and expression data of CES3 and corresponding clinical information was downloaded from The Cancer Genome Atlas (TCGA). The CES3 protein expression and the prognostic value were verified based on tissue microarray data. The Cancer immune group Atlas (TCIA), Tumor Immune Dysfunction and Exclusion (TIDE) algorithm and the GSE78220 immunotherapy cohort were used to forecast immunotherapy efficacy. Finally, a prognostic immune signature was constructed and verified.RESULTSCompared with normal colon tissues, the expression of mRNA and protein levels of CES3 were downregulated in tumor tissues. CES3 expression was associated with TIICs. Hihg-CES3 COAD patients had better efficacy of concurrent immunotherapy. CES3-related immune genes (CRIs) were identified and were then used to construct prognostic immune signature and had been successfully verified in GES39582.CONCLUSIONCES3 might be a potential immune-related gene and promising prognostic biomarker in COAD.

Identification of lncRNA-miRNA-mRNA Regulatory Network and Therapeutic Agents for Skin Aging by Bioinformatics Analysis

Skin aging is the most intuitive manifestation of aging. Skin aging inevitably leads to cosmetic and psychological problems, and even diseases. The present study aims to research the pathological and molecular mechanisms underlying skin aging and identify the therapeutic agents for reversing skin aging. Two available gene expression datasets (GSE55118 and GSE72264) for skin aging were downloaded from Gene Expression Omnibus, followed by bioinformatic analyses performed on the datasets. Firstly, 169 crucial mRNAs, 27 crucial miRNAs and 50 crucial lncRNAs closely related to skin aging were identified by weighted gene co-expression network analysis. Then, function Enrichment Analysis performed by Metascape database showed that skin aging involves a variety of biological functions, such as detection of stimulus, response to steroid hormone and water channel activity, regulation of muscle contraction. Next, ten hub genes including AQP4, TRPM8, TBR1, NTSR2, MPPED1, BARHL2, PAX9, CPN1, CES3, and CHGB were screened out by the protein-protein interaction analysis. Next, the "lncRNA-miRNA-mRNA" network and the "lncRNA-miRNA-hub mRNA" network were constructed to explore the competing endogenous RNAs mechanism of skin aging. Finally, ten significant potential small molecules mitigating skin aging were screened using CMAP platform, including tretinoin, pifithrin, selamectin, entinostat, bretazenil, syringic-acid, BRD-K96475865, emedastine, abacavir, and rotenone, and their reliability was verified by molecular docking experiments. The present study provided basis for revealing the molecular mechanism of skin aging and identified the potential candidate drugs for mitigating skin aging.
