Differential Effects of IL4I1 Protein on Lymphocytes From Healthy and Multiple Sclerosis Patients

ABSTRACTMultiple sclerosis (MS) is a chronic inflammatory disease characterized by immune‐mediated demyelination of the central nervous system, resulting in extensive neurological deficit and remyelination impairment. We have previously found that interleukin‐four induced one (IL4I1) protein modulates CNS inflammation and enhances remyelination in mouse models of experimental demyelination. However, it remained unclear if IL4I1 regulates lymphocyte activity in MS. To assess the therapeutic potential of IL4I1 in MS, we investigated the impact of IL4I1 treatment on human lymphocytes from peripheral blood mononuclear cells (PBMCs) obtained from healthy individuals and MS patients. We found that IL4I1 increased the relative densities of Th2 and regulatory T‐cells, while reducing Th17 cell density in healthy control (HC) samples. Furthermore, IL4I1‐treated lymphocytes promoted CNS remyelination when grafted into demyelinated spinal cord lesions in mice. We found that baseline endogenous IL4I1 expression was reduced in people with MS. However, unlike HCs, IL4I1 treatment had no significant effect on IL17 or TOB1 expression in lymphocytes derived from MS patients. These results suggest that IL4I1 skews CD4+ T‐cells to a regulatory state in healthy human lymphocytes, which may be essential for promoting remyelination. However, IL4I1 appears unable to exert its influence on lymphocytes in MS, indicating that impaired IL4I1‐mediated activity may underlie MS pathology.

Colorimetric aptasensor for tobramycin based on a highly stabilized aptamer-enhanced enzyme-mimicking activity of iron oxide/graphene oxide nanospheres

Antibiotic contaminants in food are associated with detrimental health issues, and thus, the design of rapid and reliable detection methods to monitor them is in high demand. Although colorimetric aptamer sensing methods can achieve accurate and quick visual read-out analysis, they still have morphological defects that affect the detection efficiency and stability of the aptasensor. Hence, a simple and highly selective iron oxide/graphene oxide (Fe₃O₄/GO) nanospheres was developed and modified with an aptamer as nanozymes for colorimetric detection of tobramycin (Tob). Due to the electrostatic and π-π stacking interaction between the aptamer and the spherical Fe₃O₄/GOs nanozyme, the catalytic activity of the nanozyme was enhanced by the robust affinity between the aptamer and the 3,3',5,5'-tetramethylbenzidine (TMB) substrate. However, the presence of Tob could obstruct the catalytic activity through the specific binding of aptamer to Tob and the detachment of aptamer from the surface of the Fe₃O₄/GO, causing a low catalytic activity. Based on this strategy, a visual and quantitative detection for Tob was achieved with a detection limit of 70.7 nM. The proposed aptasensor demonstrated high selectivity, ease of operation, and potential application in food samples with satisfactory recoveries of 98.4-108.5 %. Therefore, this proposed method has great prospects for monitoring food safety and could be applied for sensing varied antibiotics by just switching the target-specific aptamer.

Conventional and microfluidic methods: Design and optimization of lipid-polymeric hybrid nanoparticles for gene therapy

AbstractGene therapy holds significant promise as a therapeutic approach for addressing a diverse range of diseases through the suppression of overexpressed proteins and the restoration of impaired cell functions. Developing a nanocarrier that can efficiently load and release genetic material into cells remains a challenge. The primary goal of this study is to develop formulations aimed to enhance the therapeutic potential of GapmeRs through technological approaches. To this end, lipid-polymeric hybrid nanoparticles (LPHNPs) with PLGA, DC-cholesterol, and DOPE-mPEG2000 were produced by conventional single-step nanoprecipitation (SSN) and microfluidic (MF) methods. The optimized nanoparticles by SSN have a size of 149.9 ± 18.07 nm, a polydispersity index (PdI) of 0.23 ± 0.02, and a zeta potential of (ZP) of 29.34 ± 2.44 mV, while by MF the size was 179.8 ± 6.3, a PdI of 0.24 ± 0.01, and a ZP of 32.25 ± 1.36 mV. Furthermore, LPHNPs prepared with GapmeR-protamine by both methods exhibit a high encapsulation efficiency of approximately 90%. The encapsulated GapmeR is completely released in 24 h. The LPHNP suspensions are stable for up to 6 h in 10% FBS at pH 5.4 and 7.4. By contrast, LPHNPs remain stable in suspension in 4.5% albumin at pH 7.4 for 24 h. Additionally, LPHNPs were successfully freeze-dried using trehalose in the range of 2.5-5% as cryoprotectant The LPHNPs produced by MF and SSN increase, 6 and 12 fold respectively, GapmeR cell uptake, and both of them reduce by 60–70% expression of Tob1 in 48 h.Our study demonstrates the efficacy of the developed LPHNPs as carriers for oligonucleotide delivery, offering valuable insights for their scale up production from a conventional bulk methodology to a high-throughput microfluidic technology.Graphical Abstract
