Introduction

Tyrosine carboxypeptidase that removes the C-terminal tyrosine residue of alpha-tubulin, thereby regulating microtubule dynamics and function (PubMed:29146869, PubMed:31171830, PubMed:31235910, PubMed:31235911, PubMed:31270470). Critical for spindle function and accurate chromosome segregation during mitosis since microtubule detyronisation regulates mitotic spindle length and postioning (PubMed:31171830). Acts as an angiogenesis inhibitor: inhibits migration, proliferation and network formation by endothelial cells as well as angiogenesis (PubMed:15467828, PubMed:16488400, PubMed:16707096, PubMed:19204325). This inhibitory effect is selective to endothelial cells as it does not affect the migration of smooth muscle cells or fibroblasts (PubMed:15467828, PubMed:16488400, PubMed:16707096).

MicroRNA mechanisms instructing Purkinje cell specification

MicroRNAs (miRNAs) are critical for brain development; however, if, when, and how miRNAs drive neuronal subtype specification remains poorly understood. To address this, we engineered technologies with vastly improved spatiotemporal resolution that allow the dissection of cell-type-specific miRNA-target networks. Fast and reversible miRNA loss of function showed that miRNAs are necessary for Purkinje cell (PC) differentiation, which previously appeared to be miRNA independent, and identified distinct critical miRNA windows for dendritogenesis and climbing fiber synaptogenesis, structural features defining PC identity. Using new mouse models that enable miRNA-target network mapping in rare cell types, we uncovered PC-specific post-transcriptional programs. Manipulation of these programs revealed that the PC-enriched miR-206 and targets Shank3, Prag1, En2, and Vash1, which are uniquely repressed in PCs, are critical regulators of PC-specific dendritogenesis and synaptogenesis, with miR-206 knockdown and target overexpression partially phenocopying miRNA loss of function. Our results suggest that gene expression regulation by miRNAs, beyond transcription, is critical for neuronal subtype specification.

circSIRT2/miR‐542‐3p/VASH1 axis regulates endothelial‐to‐mesenchymal transition (EndMT) in subretinal fibrosis in age‐related macular degeneration models

AbstractNeovascular age‐related macular degeneration (nAMD), characterized by choroidal neovascularization (CNV), is one of the leading causes of severe visual impairment and irreversible vision loss around the world. Subretinal fibrosis (SRF) contributes to the incomplete response to anti‐vascular endothelial growth factor (VEGF) treatment and is one of the main reasons for long‐term poor visual outcomes in nAMD. Reducing SRF is urgently needed in the anti‐VEGF era. The role of non‐coding RNAs has been implicated in CNV; however, their roles in SRF have not been elucidated yet. Herein, we comprehensively investigated circular RNA (circRNA) profiles in the laser‐induced mouse SRF model and the transforming growth factor‐β (TGF‐β) induced human umbilical vein endothelial cell (HUVEC) fibrosis model. A novel circRNA, circSIRT2, was identified, and its function in SRF and endothelial‐to‐mesenchymal transition (EndMT) regulation was investigated. circSIRT2 was consistently upregulated in fibrotic models in vivo and in vitro. circSIRT2 overexpression downregulated the fibrotic markers and inhibited the proliferation and migration of endothelial cells in vitro. circSIRT2 overexpression in vivo also reduced SRF area in mice. Mechanistically, circSIRT2 functioned by sponging miR‐542‐3p, which further upregulated the expression of vasohibin‐1 (VASH1) and reduced SRF lesion development. Vitreous delivery of miR‐542‐3p and VASH1 in the mouse SRF model also confirmed the pro‐fibrotic function of miR‐542‐3p and anti‐fibrotic function of VASH1, respectively. In conclusion, circSIRT2 inhibited SRF by binding miR‐542‐3p, which stimulated the VASH1 expression and subsequently suppressed EndMT. The circSIRT2/miR‐542‐3p/VASH1 axis may serve as a promising therapeutic target for SRF in nAMD.

Characterization of tubulin post-translational modifications and their enzymes during human oocyte meiosis

RESEARCH QUESTIONWhat are the profiles of tubulin post-translational modifications (PTM) in the meiotic spindles of oocytes with different developmental competence: in-vivo matured MII oocytes (IVO) versus cumulus-free in-vitro matured germinal vesicles (IVM)?DESIGNCumulus-free germinal vesicles were matured in vitro and compared with IVO in terms of their tubulin PTM, characterizing tubulin PTM patterns (acetylation, tyrosination, detyrosination, D2-tubulin formation and polyglutamylation) of their meiotic spindles by immunofluorescence and by evaluation of the expression levels of the enzymes involved by quantitative reverse transcription polymerase chain reaction.RESULTSLevels of D2-tubulin, tyrosination and polyglutamylation were similar in IVO and IVM oocytes; acetylation displayed different patterns in both groups, whereas detyrosination was detected only in MII oocytes matured in vivo. The PTM enzyme RNA expression levels in germinal vesicles, IVO and IVM were analysed, as well as in oocytes that failed to mature. Of the 24 PTM enzyme transcripts tested, 17 were present in at least one of the groups under study (aΤΑΤ1, NAA50, HDAC6, SIRT2, TTL, TTLL1, TTLL2, TTLL4, TTLL5, TTLL6, TTLL9, TTLL11, TTLL12, CCP1, CCP5, CCP6, VASH1), of which three were differentially expressed (NAA50, TTLL12, CCP1).CONCLUSIONAll modifications studied were present in human oocytes regardless of whether their meiotic maturation occurred in vivo or in vitro, except for detyrosination, which was found at low levels in the IVO group, but not in the IVM group. This suggests that the presence of tubulin PTMs may play a role in human oocyte maturation, completion of meiosis and developmental competence.
